Protective effects of Wuwei Xiaodu Drink against chronic osteomyelitis through Foxp3^(+)CD25^(+)CD4^(+) Treg cells via the IL-2/STAT5 signaling pathway

To explore the effectiveness and safety of a Chinese medicinal decoction Wuwei Xiaodu Drink(WWXDD)in inhibiting chronic osteomyelitis via regulatory T cells signaling.The effective constitutes of WWXDD and osteomyelitis related genes were screened.Target proteins were cross-validated using the Venny database.GO function and KEGG pathway analysis were performed for target proteins,while pharmacological network was constructed.The bone properties were analyzed by HE staining and the concentrations of immune factors were measured by ELISA.The expression of CTLA-4 and Foxp3 mRNA and STAT5,p-STAT5,CTLA-4 and Foxp3 protein were detected using Real-time PCR and Western blot,respectively.FACS was used to analyze the percentages of cells.A total of 117 genes overlapped between 785 target genes of the active compounds of WWXDD and 912 osteomyelitis related genes.Inflammation-related genes,including IL-6,TNFα,IL-1βand IL-2 showed high connection degree in the drug-compound-disease-target network.GO function and KEGG pathway analysis revealed that 117 intersection genes mainly enriched in virus infection related pathways,immune related pathways and chemokine signaling pathway.Furthermore,the development of chronic osteomyelitis was suppressed in model rats after treatment with WWXDD.Meanwhile,the concentrations of IL-2 and CD4^(+)CD25^(+)Foxp3 Treg perr centages together with the levels of p-STAT5,CTLA-4 and Foxp3 were also down-regulated.Furthermore,IL-2 and WWXDD drug-containing serum exhibited opposite effects on regulating IL-2,IL-10,TGF-β1,Foxp3,CTLA4 and STAT5.In addition,a STAT5 phosphorylation inhibitor suppressed the expression of Foxp3 and CTLA-4.WWXDD can treat chronic osteomyelitis through suppressing the main regulating factors of Tregs and interfere its immunodepression.Our results bring a new solution for chronic osteomyelitis.

Passenger emergency evacuation from bus carriage:Results from realistic data and modeling simulations

One of critical challenges in the development of pedestrian models is the lack of relevant data under emergency conditions.In this paper,we reveal several behavior characteristics of passenger emergency evacuation from a burning bus carriage,by observing and analyzing a video recording,attempting to close the gap between practical observation and theoretical modeling.The analysis results show that there are considerable differences between real emergency evacuation and experimental normal evacuation with respect to the cumulative flows,flows,and time gap distributions.Additionally,the behavior of falling-down,which contributes to the formation of movable obstacles,interrupts the continuity of the evacuation process.Then,we incorporate these behavioral characteristics into a microscopic pedestrian model with a fine lattice space representation.The simulation results indicate that the escape is slowed down by the falling behavior,as well as by the luggage-carrying behavior.

Small extracellular vesicles secreted by induced pluripotent stem cell-derived mesenchymal stem cells improve postoperative cognitive dysfunction in mice with diabetes

Postoperative cognitive dysfunction(POCD)is a common surgical complication.Diabetes mellitus(DM)increases risk of developing POCD after surgery.DM patients with POCD seriously threaten the quality of patients’life,however,the intrinsic mechanism is unclear,and the effective treatment is deficiency.Previous studies have demonstrated neuronal loss and reduced neurogenesis in the hippocampus in mouse models of POCD.In this study,we constructed a mouse model of DM by intraperitoneal injection of streptozotocin,and then induced postoperative cognitive dysfunction by transient bilateral common carotid artery occlusion.We found that mouse models of DM-POCD exhibited the most serious cognitive impairment,as well as the most hippocampal neural stem cells(H-NSCs)loss and neurogenesis decline.Subsequently,we hypothesized that small extracellular vesicles secreted by induced pluripotent stem cell-derived mesenchymal stem cells(iMSC-sEVs)might promote neurogenesis and restore cognitive function in patients with DM-POCD.iMSC-sEVs were administered via the tail vein beginning on day 2 after surgery,and then once every 3 days for 1 month thereafter.Our results showed that iMSC-sEVs treatment significantly recovered compromised proliferation and neuronal-differentiation capacity in H-NSCs,and reversed cognitive impairment in mouse models of DM-POCD.Furthermore,miRNA sequencing and qPCR showed miR-21-5p and miR-486-5p were the highest expression in iMSC-sEVs.We found iMSC-sEVs mainly transferred miR-21-5p and miR-486-5p to promote H-NSCs proliferation and neurogenesis.As miR-21-5p was demonstrated to directly targete Epha4 and CDKN2C,while miR-486-5p can inhibit FoxO1 in NSCs.We then demonstrated iMSC-sEVs can transfer miR-21-5p and miR-486-5p to inhibit EphA4,CDKN2C,and FoxO1 expression in H-NSCs.Collectively,these results indicate significant H-NSC loss and neurogenesis reduction lead to DM-POCD,the application of iMSC-sEVs may represent a novel cell-free therapeutic tool for diabetic patients with postoperative cognitive dysfun

Un displaced subtle ligamentous Lisfranc injuries, con servative or surgical treatment with percutaneous position screws?

Purpose: Un displaced subtle ligamentous Lisfranc injuries are easy to miss or underestimate, and many cases are treated without surgical fixation. It has not yet widely known whether conservative treatment for undisplaced subtle ligamentous Lisfranc injuries may lead to a poor outcome. The purpose of this study is to compare the outcomes of conservative versus surgical management (percutaneous position screw) of undisplaced subtle ligamentous Lisfranc injury. Methods: We analysed 61 cases in this retrospective study, including 38 males and 23 females. Forty-one patients were managed conservatively, while 20 patients received surgical treatment involving minimal invasive percutaneous pos让ion screw. American orthopaedic foot &ankle society (AOFAS), foot function index (FFI, including FFI disability, FFl pain score and activity limitation scale) scores, Maryland foot score and short form-36 (SF-36) were recorded and compared after a follow-up of 10-16 months (average 12.3). Results: Patients in the surgical management group had higher scores in all evaluation methods (p < 0.05). The complications in the conservative management group had higher incidenee, mainly including secondary diastasis (34.1% vs. 5.0%), joint stiffness after 3 months (82.9% vs. 0%), and secondary arthrodesis (12.2% vs. 0%). The highest rate of complication in surgical management group was temporary forefoot pain (55.0%). Conclusion: The results of this study suggest that the outcomes of the surgical management with percutaneous position screw fixation are better than the conservative management to treat undisplaced subtle ligamentous Lisfranc injuries. This study can serve as a resource for orthopaedic surgeons in recog nizing and man aging such injuries.

Exploration of potential biomarkers and therapeutic targets for trauma-related acute kidney injury

Purpose:Acute kidney injury(AKI)is one of the most common functional injuries observed in trauma patients.However,certain trauma medications may exacerbate renal injury.Therefore,the early detection of trauma-related AKI holds paramount importance in improving trauma prognosis.Methods:Qualified datasets were selected from public databases,and common differentially expressed genes related to trauma-induced AKI and hub genes were identified through enrichment analysis and the establishment of protein-protein interaction(PPI)networks.Additionally,the specificity of these hub genes was investigated using the sepsis dataset and conducted a comprehensive literature review to assess their plausibility.The raw data from both datasets were downloaded using R software(version 4.2.1)and processed with the"affy"package19 for correction and normalization.Results:Our analysis revealed 585 upregulated and 629 downregulated differentially expressed genes in the AKI dataset,along with 586 upregulated and 948 downregulated differentially expressed genes in the trauma dataset.Concurrently,the establishment of the PPI network and subsequent topological analysis highlighted key hub genes,including CD44,CD163,TIMP metallopeptidase inhibitor 1,cytochrome b-245 beta chain,versican,membrane spanning 4-domains A4A,mitogen-activated protein kinase 14,and early growth response 1.Notably,their receiver operating characteristic curves displayed areas exceeding 75%,indicating good diagnostic performance.Moreover,our findings postulated a unique molecular mechanism underlying trauma-related AKI.Conclusion:This study presents an alternative strategy for the early diagnosis and treatment of trauma-related AKI,based on the identification of potential biomarkers and therapeutic targets.Additionally,this study provides theoretical references for elucidating the mechanisms of trauma-related AKI.

First synthesis and characterization of key stereoisomers related to Ezetimibe

uring the laboratory optimization and the late phase manufacturing studies of the cholesterol absorption inhibitor Ezetimibe 1, the formation of several stereoisomers was observed. To study the complete stereoisomer profile of Ezetimibe 1, we have synthesized and completely characterized several key stereoisomers of Ezetimibe 1 for the first time. This study will provide an access to the reference standard of these stereoisomers and may have some implications in the development of new medicines.

Fe-catalyzed regioselective Friedel–Crafts hydroxyalkylation of N-substituted glyoxylamide with indoles

An efficient regioselective Friedel-Crafts hydroxyalkylation of N-substituted glyoxylamide with various indoles catalyzed by Lewis acids was developed. The reactions proceeded smoothly at room temperature and the 2-hydroxy-2-（1H-indol-3-yl）-N-substituted acetamide resulted from the reactions catalyzed by FeSO4 were synthesized in excellent yields （up to 93%）. While the bisindole compounds were obtained when FeCl3 was used as a catalyst in excellent yields （up to 92%）. A possible mechanism was proposed.

Identification and functional prediction of long intergenic noncoding RNAs in fetal porcine longissimus dorsi muscle

Pigs are globally farmed animals which provide protein for human consumption in the form of skeletal muscle. To better understand the function of long intergenic noncoding RNAs(linc RNAs) in porcine skeletal muscle growth and development, we collected RNA-seq data from porcine longissimus dorsi muscle(LDM) during embryonic development. We identified a total of 739 linc RNA transcripts, which were distributed on all chromosomes except the chromosome Y, and analyzed their molecular characteristics. Compared to protein-coding genes, linc RNAs showed shorter transcripts, longer exons, fewer exons and higher tissue specificity. In addition, the abundance of linc RNAs in five embryonic development stages were analyzed and 45 differentially expressed linc RNAs were screened, three of which were highly expressed in LDM during porcine embryonic development. Finally, we predicted the potential target genes and functions of the linc RNAs, and identified 1 537 cis-target genes and 8 571 trans-target genes. Furthermore, we identified two key candidate linc RNAs involved in muscle development, XLOC_024652 and XLOC_001832, for post-trial validation. Our results provide a genome-wide resource of linc RNAs which are potentially involved in porcine embryonic skeletal muscle development and lay a foundation for the further study of their functions.

Strategies and challenges in treatment of varicose veins and venous insufficiency

Patients with varicose veins can be treated with conservative or surgical approaches based on the clinical conditions and patient preferences.In the recent decade,the recommendations for managing symptomatic varicose veins have changed dramatically due to the rise of minimally invasive endovascular techniques.The literature was systematically searched on Medline without language restrictions.All papers on the treatment of varicose veins and venous insufficiency with different procedures were included and reviewed.Endovenous laser ablation(EVLA)and radiofrequency ablation(RFA)both are same safe and effective in terms of occlusion rate,and time to return to normal activity.In comparison with RFA or EVLT,Cure conservatrice et Hemodynamique de l’Insufficience Veineuse en Ambulatoire(CHIVA)may cause more bruising and make little or no difference to rates of limb infection,superficial vein thrombosis,nerve injury,or hematoma.In terms of recurrence of varicose veins,there is little or no difference between CHIVA and stripping,RFA,or EVLT.Great saphenous vein recanalization is highest in the ultrasound-guided foam sclerotherapy(FS)group(51%)during 1 year of follow-up.The 2013 National Institute for Health and Care Excellence clinical guidelines recommend surgery as a third-line therapeutic option after EVLA or RFA and sclerotherapy.Although the mechanochemical endovenous ablation(MOCA)is a non-thermal,non-tumescent option and appears to be of similar efficacy to stab avulsion with no potential risk of nerve damage,the overall success rate of MOCA is lower than those of other procedures such as EVLA,RFA,or high ligation and stripping.EVLA is the most cost-effective therapeutic option,with RFA being a close second for the treatment of patients with varicose veins.Endovenous thermal ablation(EVLA or RFA)is recommended as a first-line treatment for varicose veins and has substituted the high ligation of saphenofemoral junctional reflux and stripping of varicose veins.Ultrasound-guided FS is associated with a high recurrence

Research and development of 780 MPa cold rolling TRIP-aided steel

As an industry-university cooperative project, an experimental research was conducted to develop a new cold-rolled transformation-induced plasticity （TRIP） steel with a tensile strength of 780 MPa and an elongation of 25% under the conditions that some processing parameters were preestablished according to the actual conditions of factory production lines. The optimal heat treatment conditions for obtaining excellent strength-ductility balance, specifically in intercritical annealing temperature and isothermal treatment temperature, were investigated by means of thermal simulation tests. Ultimately the desirable mechanical properties were attained successfully, and the microstructure and retained austenite stability were studied by optical microscopy （OM） and electron back-scattered diffraction （EBSD）. It has been discovered that the sizes of retained austenite grains are generally proportional to the sizes of carbon-clear space （ferrite and bainite） around them, and there is marked selectivity in their transition sequences under stress-strain.
