Functional Interaction of the SNArE Protein NtSyp121 in Ca^2＋ Channel Gating, Ca^2＋ Transientsand ABA Signalling of Stomatal Guard Cells

There is now growing evidence that membrane vesicle trafficking proteins, especially of the superfamily of SNArEs, are critical for cellular signalling in plants. Work from this laboratory first demonstrated that a soluble, inhibitory （dominant-negative） fragment of the SNArE NtSyp121 blocked K^＋ and CI^- channel responses to the stress-related hormone abscisic acid （ABA）, but left open a question about functional impacts on signal intermediates, especially on Ca^2＋-mediated signalling events. Here, we report one mode of action for the SNArE mediated directly through alterations in Ca^2＋ channel gating and its consequent effects on cytosolic-free [Ca^2＋] （[Ca^2＋]i） elevation. We find that expressing the same inhibitory fragment of NtSyp121 blocks ABA-evoked stomatal closure, but only partially suppresses stomatal closure in the presence of the NO donor, SNAP, which promotes [Ca^2＋]i elevation independently of the plasma membrane Ca^2＋ channels. Consistent with these observations, Ca^2＋ channel gating at the plasma membrane is altered by the SNArE fragment in a manner effective in reducing the potential for triggering a rise in [Ca^2＋]i, and we show directly that its expression in vivo leads to a pronounced suppression of evoked [Ca^2＋]i transients. These observations offer primary evidence for the functional coupling of the SNArE with Ca^2＋ channels at the plant cell plasma membrane and, because [Ca^2＋]i plays a key role in the control of K^＋ and CI^- channel currents in guard cells, they underscore an important mechanism for SNArE integration with ion channel regulation during stomatal closure.

Assemblages of <i>Giardia duodenalis</i>Isolates from Dogs by Amplification and restriction of tpi and <i>β</i>-Giardin Genes and Sequencing of SSU rDNA Gene

Giardia duodenalis exhibits seven assemblages (A-G) that are distributed in different hosts. The A and B assemblages are commonly found in humans and several mammals, while C and D assemblages are typically found in dogs. The purpose of this study was to determine the assemblage of Giardia duodenalis present in the stool samples of ten canines using an assay based on PCr amplification, restriction analysis, and sequencing of the small subunit ribosomal DNA (SSU-rDNA), β-giardin, and triosephosphate isomerase (tpi) genes in order to establish the similarities or differences between the assemblage obtained with each gene. The results indicated that all positive isolates belonged to assemblage A, and specifically to the sub-assemblage A-I. A comparison of the SSU-rDNA gene sequence revealed the presence of three subgroups of assemblage A. These findings highlight the importance of canine transmission of Giardia in Mexico and its genetic plasticity. They also establish a method for additional and more molecularly extensive epidemiological studies to improve sanitation and hygiene in the most affected areas.

Numerical Simulation and Experiments of Hydroforming of rectangular-section Tubular Component

In order to reduce high calibration pressure in hydroforming of components with too small radii, a method wasproposed to manufacture automotive hollow components with rectangular shape by relatively lower pressure. Theprocess is simulated and analyzed. It is thought that the friction force between the die surface and tube is a mainreason that high pressure is needed to form small radii. Using the method proposed in this paper, a petal-like sectionshape is first preformed so that the central zones of the four sides of the preform section do not contact with the diesides, thus the tube metal is easy to flow into the transition radii area in calibration stage. Moreover, a positive forcealong the sides is produced by the internal pressure, which is beneficial to overcome the friction force and push thematerial into the radii. Therefore, the pressure for forming the transition radii is greatly reduced and the componentswith small radii can be formed with relatively lower pressure. For the experimental case conducted in this paper, theforming pressure is reduced by about 28.6% than the estimated forming pressure.

Management of Complex Wounds with Dermal Substitute Assisted by a Negative Pressure System

Introduction: The standard treatment for complex wound care is autografting. The advent of dermal substitutes has provided a novel tool for the preparation of the bed to be grafted. However, most types of dermal matrices require the application of a skin graft a second time. Currently, other strategies have been developed to improve the vascularization process, such as negative pressure wound therapy (NPWT), which has been reported to reduce the time required for vascular growth and dermal matrix integration and thus achieve a shorter waiting period for autologous graft application. The present study aims to evaluate the effectiveness and safety of dermal matrix management associated with NPWT in the treatment of complex wounds. Methods: Seven patients with a diagnosis of complex wounds were enrolled in this study between July 1, 2015, and June 31, 2016. After debridement and having an adequate wound bed, patients who met the criteria for the application of combined therapy were treated with dermal substitutes and a negative pressure system. The percentage of graft integration into the wound bed, complications, length of hospital stay, and duration of therapy were analyzed. results: The mean age was 42.5 ± 16 (39 - 54) years old;three women and four men were included in the study. The approximate size of skin loss was 120.7 ± 75 cm2 (25 - 250 cm2). The combined therapy of dermal matrix plus NPWT was instituted in all cases for a period of 14 days. There were no complications, with 100% graft integration in 6 of 7 cases. Patients were discharged after a mean hospital stay of 5.4 days. Conclusions: This study demonstrates that the utilization of combined dermal matrix plus NPWT therapy can be performed safely and effectively in patients with complex wounds with low complication rates and a short hospital stay.

Effect of Thin Metallic Layers on the refractive Index of a Multilayer System

We present a simplified structure made of periodic metallic layers on a glass substrate,which offers a potentially simpler approach to building photonic *** microwave experiments and Fresnel formulas,we investigate the refractive index properties of these *** make both transmission and reflection measurements and find that the refractive index of a periodic layered media decreases depending on the periodicity of the geometric arrangement,dielectric contrast and filling fraction of metallic *** is shown that metallic layers lead to a substantial change of interference pattern which can be interpreted as a result of interference in a uniform medium with a refractive index smaller than 1.

Evaluation of integration methods for hybrid simulation of complex structural systems through collapse

This study examines the performance of integration methods for hybrid simulation of large and complex structural systems in the context of structural collapse due to seismic excitations. The target application is not necessarily for real-time testing, but rather for models that involve large-scale physical sub-structures and highly nonlinear numerical models. Four case studies are presented and discussed. In the first case study, the accuracy of integration schemes including two widely used methods, namely, modified version of the implicit Newmark with fixed-number of iteration （iterative） and the operator-splitting （non-iterative） is examined through pure numerical simulations. The second case study presents the results of 10 hybrid simulations repeated with the two aforementioned integration methods considering various time steps and fixed-number of iterations for the iterative integration method. The physical sub-structure in these tests consists of a single-degree-of-freedom （SDOF） cantilever column with replaceable steel coupons that provides repeatable highly- nonlinear behavior including fracture-type strength and stiffness degradations. In case study three, the implicit Newmark with fixed-number of iterations is applied for hybrid simulations of a 1：2 scale steel moment frame that includes a relatively complex nonlinear numerical substructure. Lastly, a more complex numerical substructure is considered by constructing a nonlinear computational model of a moment frame coupled to a hybrid model ofa 1：2 scale steel gravity frame. The last two case studies are conducted on the same porotype structure and the selection of time steps and fixed number of iterations are closely examined in pre-test simulations. The generated unbalance forces is used as an index to track the equilibrium error and predict the accuracy and stability of the simulations.

Lie symmetry group transformation for MHD natural convection flow of nanofluid over linearly porous stretching sheet in presence of thermal stratification

The magnetohydrodynamics （MHD） convection flow and heat transfer of an incompressible viscous nanofluid past a semi-infinite vertical stretching sheet in the pres- ence of thermal stratification are examined. The partial differential equations governing the problem under consideration are transformed by a special form of the Lie symmetry group transformations, i.e., a one-parameter group of transformations into a system of ordinary differential equations which are numerically solved using the runge-Kutta-Gill- based shooting method. It is concluded that the flow field, temperature, and nanoparticle volume fraction profiles are significantly influenced by the thermal stratification and the magnetic field.
