Alzheimer’s disease(AD)is the most common type of dementia,but it is very difficult to diagnose with certainty,so many AD studies have attempted to find early and relevant diagnostic *** upon activation,normal T cell expressed and secreted(rANTES,also known as C-C chemokine ligand)is a chemokine involved in the migration of T cells and other lymphoid *** in rANTES levels and its expression in blood or in cerebrospinal fluid have been reported in some neurodegenerative diseases,such as Parkinson’s disease and multiple sclerosis,but also in metabolic diseases in which inflammation plays a *** aim of this observational study was to assess rANTES levels in peripheral blood as clinical indicators of *** levels of rANTES were investigated in 85 AD patients in a relatively early phase of AD(median 8.5 months after diagnosis;39 men and 46 women;average age 75.7 years),and in 78 control subjects(24 men and 54 women;average age 66 years).We found much higher plasma levels of rANTES in AD patients compared to controls.A negative correlation of rANTES levels with age,disease duration,Fazekas scale score,and the medial temporal lobe atrophy(Mta)score(Scheltens’s scale)was found in AD patients,i.e.,the higher levels corresponded to earlier stages of the *** rANTES levels were not correlated with cognitive *** AD patients,rANTES levels were positively correlated with the levels of pro-inflammatory cytokines interleukin-6 and tumor necrosis factor-α,which is consistent with the wellknown fact that AD is associated with inflammatory *** levels were also positively correlated with insulin levels in AD patients,with insulin resistance(HoMA-r)and pancreatic beta cell function(HoMA-F).This study evaluated several clinical and metabolic factors that may affect plasma levels of rANTES,but these factors could not explain the increases in rANTES levels observed in AD *** levels of rANTES appear to be an interesting

The identification of jets originating from quarks and gluons,often referred to as quark/gluon tagging,plays an important role in various analyses performed at the Large Hadron Collider,as Standard Model measurements and searches for new particles decaying to quarks often rely on suppressing a large gluon-induced *** paper describes the measurement of the efficiencies of quark/gluon taggers developed within the ATLAS Collaboration,using √s=13 TeV proton–proton collision data with an integrated luminosity of 140 fb^(-1) collected by the ATLAS *** taggers with high performances in rejecting jets from gluon over jets from quarks are studied:one tagger is based on requirements on the number of inner-detector tracks associated with the jet,and the other combines several jet substructure observables using a boosted decision tree.A method is established to determine the quark/gluon fraction in data,by using quark/gluon-enriched subsamples defined by the jet *** in tagging efficiency between data and simulation are provided for jets with transverse momentum between 500 GeV and 2 TeV and for multiple tagger working points.

Single-cell mass cytometry(SCMC)combines features of traditional flow cytometry(i.e.,fluorescence-activated cell sorting)with mass spectrometry,making it possible to measure several parameters at the single-cell level for a complex analysis of biological regulatory *** this study,we optimized SCMC to analyze hemocytes of the Drosophila innate immune *** used metal-conjugated antibodies(against cell surface antigens H2,H3,H18,L1,L4,and P1,and intracellular antigens 3A5 and L2)and anti-igM(against cell surface antigen L6)to detect the levels of antigens,while anti-GFP was used to detect crystal cells in the immune-induced *** investigated the antigen expression profile of single cells and hemocyte populations in naive states,in immune-induced states,in tumorous mutants bearing a driver mutation in the Drosophila homologue of Janus kinase(hopTum)and carrying a deficiency of the tumor suppressor gene lethal(3)malignant blood neoplasm-1[l(3)mbn1],as well as in stem cell maintenance-defective hdcD84 mutant *** analysis enabled the discrimination of the functionally different major hemocyte subsets for lamellocytes,plasmatocytes,and crystal cells,and delineated the unique immunophenotype of Drosophila *** have identified subpopulations of L2^(+)/P1^(+)and L2^(+)/L4^(+)/P1^(+)transitional phenotype cells in the tumorous strains l(3)mbn1 and hopTum,respectively,and a subpopulation of L4^(+)/P1^(+)cells upon immune *** results demonstrated for the first time that SCMC,combined with multidimensional bioinformatic analysis,represents a versatile and powerful tool to deeply analyze the regulation of cell-mediated immunity of Drosophila.

目的基于非酒精性脂肪肝(NAFLD)与糖调节受损(iGr)研究代谢病肝病传脾的基因本质。方法108只血糖正常SD大鼠,按体重分层、血糖随机分为对照组(饲以常规饲料)、模型组、预防组(饲以高糖饲料),建立NAFLD及iGr模型。预防组同步用痰瘀同治的丹瓜方干预。共12周。观察NAFLD和iGr的发病率、相关指标,做肝组织HE、MASSoN及油红o染色。以illumina Hiseq2500仪做转录组全基因表达及isoform对照分析。结果与对照组比较,模型组超声检测脂肪肝发病率显著增高及肝重增加(PoGTT 0 h及2 h血糖、2 h血浆胰岛素水平(P2iNS)、葡萄糖曲线下面积(G-AUC)、胰岛素抵抗指数(HoMA-ir)、iGr发病率较对照组显著升高(PoMA-β)、胰岛素敏感指数(HoMA-iSi)显著下降(PoGTT 2 h血糖、G-AUC、iGr发病率,P2iNS、糖负荷后HoMA-ir显著降低(PoMA-iSi显著升高(P<0.01),并恢复了四类基因中大部分异常表达的基因。结论代谢病肝病传脾存在成脂基因、能量代谢基因、胰岛功能基因、胰岛素抵抗基因的异常表达,干预这些基因的表达阻滞了肝病传脾的发展,这些异常表达的基因可能在代谢病肝病传脾的演变机制中起到了一定的作用。
