Accurate measurement of film thickness and its uniformity are very important to the performance of many coated surfaces and for many applications are critical. Effective inspection of the film thickness and uniformity is the key to high performance. Conventionally, film thickness is meas- ured using a spectrophotometer/reflectometer, ellipsometer or a physical step measurement;however, these techniques all have limitations. Coherence Scanning Interferometry (CSI) is an established method to measure surface topography as this technique offers many advantages such as speed, ease of use and accuracy. The measurement of “thick” films (exceeding ~1.5 μm) which gives rise to clearly separate fringe bunches is a well-established CSI capability. However, a methodology known as the ‘helical complex field’ (HCF) [1] [2] function allows film thickness to be measured down to ~25 nm. This new method, combined with Coherence Correlation Interferometry (CCI) [3] offers film thickness measurements with sub-nanometre vertical resolution and ~1 μm lateral resolution. It is ideally suited for detailed analysis of coated optical surfaces. In this paper, the fundamentals of the techniques are described and some case studies are presented.

Background: This study investigated the validity of the DNA-marker based test to determine susceptibility to ETECF4 diarrhoea by comparing the results of two DNA sequencing techniques in weaner pigs following experimental infection with F4 enterotoxigenic Escherichia coli(ETEC-F4). The effects of diet and genetic susceptibility were assessed by measuring the incidence of piglet post-weaning diarrhoea(PWD), faecal E. coli shedding and the diarrhoea ***: A DNA marker-based test targeting the mucin 4 gene(MUC4) that encodes F4 fimbria receptor identified pigs as either fully susceptible(SS), partially or mildly susceptible(Sr), and resistant(rr) to developing ETEC-F4 diarrhoea. To further analyse this, DNA sequencing was undertaken, and a significantly higher proportion of C nucleotides was observed for rr and Sr at the Xba I cleavage site genotypes when compared to SS. However, no significant difference was found between Sr and rr genotypes. Therefore, results obtained from Sanger sequencing retrospectively allocated pigs into a resistant genotype(MUC4–), in the case of a C nucleotide, and a susceptible genotype(MUC4+), in the case of a g nucleotide, at the single nucleotide polymorphism site. A total of 72 weaner pigs(age ~ 21 days), weighing 6.1 ± 1.2 kg(mean ± SEM), were fed 3 different diets:(i) positive control(PC) group supplemented with 3 g/kg zinc oxide(Zn O),(ii) negative control(NC) group(no Zn O or HAMSA),and(iii) a diet containing a 50 g/kg high-amylose maize starch product(HAMSA) esterified with acetate. At days five and six after weaning, all pigs were orally infected with ETEC(serotype O149:F4;toxins LT1, ST1, ST2 and EAST). The percentage of pigs that developed diarrhoea following infection was higher(P = 0.05) in MUC4+ pigs compared to MUC4– pigs(50% vs. 26.8%, respectively). Initial Validation of Cytokine Measurement by ELISA in Canine Feces

Measurement of fecal cytokines has been used as a marker of intestinal inflammation in people and correlates with endoscopic findings. The aim of this study was to evaluate the use of canine-specific enzyme-linked immunosorbant assays (ELISAs) for quantification of cytokines in canine fecal samples as a non-invasive biomarker. Interleukin (IL)-6, -8, -10, -23/12p40 and TNF-a were assessed by using spiked fecal samples from 3 healthy dogs. Standard curve validation was performed, and the impact of time to freeze, duration of storage and number of freeze-thaw cycles on cytokine concentration were also examined. All the cytokines assayed could be detected, with varying accuracy. The mean coefficient of variation (CV) for all standard curves ranged from 2.95%-9.8%. The mean intra-assay CV ranged from 3.1%-11.14%, and inter-assay CV from 4.36%-18.83%. recovery of IL-23 was poor (7.23%-17.12%), precluding further interpretation of stability studies. Mean recovery did not appear to be affected by time to freeze and repeat freeze-thaw cycles in all cytokines investigated. recovery for all cytokines after short-term storage of 30 days at -80℃ showed a recovery of 70% or >130%. In conclusion, although fecal IL-6, -8, -10, and TNF-a could be used as biomarkers of intestinal inflammation in the dog, the quality of laboratory performance and poor recovery at lower concentrations limit their application. Bench-top and freeze-thaw stability was acceptable, and samples should ideally be analyzed within a week. Investigation involving dogs with acute and chronic inflammatory intestinal disease is required to determine the role of this methodology in a clinical setting.

genetic manipulation of cellulose biosynthesis in trees may provide novel insights into the growth and development of trees. To explore this possibility, the overexpression of an aspen secondary wall-associated cellulose synthase （PtdCesAS） gene was attempted in transgenic aspen （Populus tremuloides L.） and unexpectedly resulted in silencing of the transgene as well as its endogenous counterparts. The main axis of the transgenic aspen plants quickly stopped growing, and weak branches adopted a weeping growth habit. Furthermore, transgenic plants initially developed smaller leaves and a less extensive root system. Secondary xylem （wood） of transgenic aspen plants contained as little as 10% cellulose normalized to dry weight compared to 41% cellulose typically found in normal aspen wood. This massive reduction in cellulose was accompanied by proportional increases in lignin （35%） and non-cellulosic polysaccharides （55%） compared to the 22% lignin and 36% non-cellulosic polysaccharides in control plants. The transgenic stems pro- duced typical collapsed or ＇irregular＇ xylem vessels that had altered secondary wall morphology and contained greatly reduced amounts of crystalline cellulose. These results demonstrate the fundamental role of secondary wall cellulose within the secondary xylem in maintaining the strength and structural integrity required to establish the vertical growth habit in trees.
