Chr.nic pancr.atitis (CP) is a pr.gr.ssive infl.mmator. disor.er.of the pancr.as. It is pr.dominantl. idiopathic (with an unknown cause) in India and mostl. due to al.ohol.in the West. Diabetes that occur.secondar. to chr.nic pancr.atitis (T3c Diabetes) is often br.ttl., and is difficul. to attain nor.ogl.cemia with conventional.tr.atment r.quir.ng mul.ipl. doses of insul.n. Mil. and sever. model.of CP was induced in mice by r.peated intr.per.toneal.injections of cer.l.in and l.ar.inine r.spectivel. with an intent to study isl.t dysfunction and devel.p ther.peutic str.tegy in animal.model. of CP. Dietar. inter.ention of epigal.ocatechin-3-gal.ate (EGCG) was tested in both the model. of CP for.its beneficial.effects on insul.n secr.tor. functions. Pancr.ata col.ected upon euthanasia wer. used to study al.er.tions in the mor.hol.gy of pancr.atic par.nchyma and infl.mmation by staining with H&E and fibr.tic changes by Masson’s tr.chr.me and picr.sir.us staining. Insul.n secr.tor. functions of isl.ts wer. eval.ated to test the efficacy of the dietar. inter.ention on β-cel. functions. Intr.per.toneal.gl.cose tol.r.nce test was per.or.ed to monitor.the gl.cose homeostasis befor. and after.the dietar. inter.ention. Both the model. r.sul.ed in CP with disper.ed acini, infl.mmation and fibr.sis. The l.ss of acini and extent of fibr.sis was mor. in l.ar.inine model. 2-fol. impr.vement in gl.cose-stimul.ted insul.n secr.tor. functions of isl.ts was obser.ed with 0.5% EGCG dietar. inter.ention in cer.l.in model.of CP and 1.6-fol. in l.ar.inine model.of CP. A fur.her.impr.vement in insul.n secr.tion by 3.2-fol. was obser.ed with additional.dietar. suppl.ments l.ke N-acetyl.cysteine, cur.umin in combination with EGCG. Our.r.sul.s thus demonstr.te and highl.ght the ther.peutic potential.of dietar. gr.en tea (EGCG) suppl.mentation in r.ver.ing isl.t dysfunction and impr.ving gl.cose homeostasis in exper.mental.chr.nic pancr.atitis in mice.
