Objective:In this paper,we will discuss if the CETP polymorphism contributes to the centenarians in Hainan ***:We tested the Taq IB and I405 V polymorphisms of CETP gene among 276 centenarians and 301 matched healthy individuals by AS-PCR and analyzed the data with SPSS software package(Version 19.0).Results:Our data indicated that allele B1 and V have the significant differences between centenarians and healthy control groups with Plied that genotypes B1B1(Psuch as B1B1-II(Psults implied that allele B1B1 and VV,as well as the combination B1B1-II,B1B1-IV,B1B2-VV and B2B2-VV may contribute to the longevity in centenarians of Hainan,south of China.

SIRT6 belongs to the conserved NAD^(+)-dependent deacetylase superfamily and mediates multiple biological and pathological *** SIRT6 by allosteric modulators represents a novel direction for therapeutics,which can overcome the selectivity problem caused by the structural similarity of orthosteric sites among ***,developing a reversed allosteric strategy Allo Reverse,we identified a cryptic allosteric site,Pocket Z,which was only induced by the bi-directional allosteric signal triggered upon orthosteric binding of NAD^(+).Based on Pocket Z,we discovered an SIRT6 allosteric inhibitor named ***-42 selectively targets SIRT6 among other histone deacetylases and effectively inhibits SIRT6 deacetylation,with an IC50 of 2.33μmol/***-42 significantly suppresses SIRT6-mediated cancer cell migration and pro-inflammatory cytokine ***-42,to our knowledge,is the most potent and selective allosteric SIRT6 *** study provides a novel strategy for allosteric drug design and will help in the challenging development of therapeutic agents that can selectively bind SIRT6.

The growth periods（GPs, from planting/emergence to reproductive stage 8（R8） of soybean cultivars vary in different ecological regions, especially in China with a very complex soybean cropping system. In this study, a 3-yr experimental study was undertaken in three geographical locations of China from 2008 to 2010, including the Northeast（40.66-45.85°N）, Huang-Huai（34.75-38.04°N） and southern（22.82-30.60°N） eco-regions with about 250 accessions in each region to clarify the classification of maturity group（MG） and identify the cultivars with stable GP to increase the knowledge about the GP distribution of soybean cultivars in China. GPs of soybean cultivars in different eco-regions were significant different with a gradual decrease from 115-125 d in the Northeast part to the 85-100 d in the southern part of China. The geographical location was the major factor for GP of cultivars from the Northeast, while the year of planting was the major factor affecting the stability of GPs in Huang-Huai summer and southern summer soybean. AMMI2（additive main effects and multiplicativeinteraction）-Biplot analysis showed that the GPs of soybean cultivars from the Northeast eco-region have a comparatively satisfactory environmental stability. Moreover, soybean cultivars with moderate GP/MG and stable environment adaptability in different eco-regions were identified based on the linear regression and AMMI analysis, which was important for the accurate classification of soybean MGs in future. Taken together, our results reflected the genetic diversity, geographical distribution and environmental stability of the Chinese soybean GP trait. Soybean cultivars with stable GP for various Chinese eco-regions would be beneficial for Chinese soybean genetic improvement, varietal introduction, exchange, and soybean breeding program for wide adaptability.

A low-energy ion spectrometer(LEIS) for use aboard three-axis stabilized spacecraft has been developed to measure ion energy per charge distribution in three-dimensional space with good energy-, angular-and temporal-resolutions. For the standard top-hat electrostatic analyzer used widely in space plasma detection, three-axis stabilized spacecraft makes it difficult to obtain complete coverage of all possible ion arrival directions. We have designed angular scanning deflectors supplementing to a cylindrically symmetric top-hat electrostatic analyzer to provide a half-space field of view as 360°×90°(–45°–+45°), and fabricated the LEIS flight model for detecting magnetospheric ions in geosynchronous orbit. The performance of this payload has been evaluated in detail by a series of simulation and environmental tests, and the payload has also been calibrated through laboratory experiments using a low-energy ion source. The results show that capabilities of the LEIS payload are in accordance with the requirements of a magnetospheric mission.

summary:In late December 2019,COVID-19 was firstly recognized in Wuhan,China and spread rapidly to all of the provinces of *** West Campus of Wuhan Union Hospital,the designated hospital to admit and treat the severe and critically ill COVID-19 cases,has treated a large number of such patients with great success and obtained lots of valuable experiences based on the Chinese guideline(V7.0).To standardize and share the treatment procedures of severe and critically ill cases,Wuhan Union Hospital has established a working group and formulated an operational recommendation,including the monitoring,early warning indicators,and several treatment principles for severe and critically ill *** treatment experiences may provide some constructive suggestions for treating the severe and critically ill COVID-19 cases all over the world.

B to O erythrocyte conversion by the recombinant α-galactosidase

Background Human group O red blood cells have great benefit in specialized transfusion areas such as armed conflict and natural calamity. The group B antigen differs structurally from group O antigen only by the addition of one terminal α-linked galactose residue. In this study we aimed to remove the terminal galactose from group B red blood cell to get group O red blood cell. Methods α-galactosidase cDNA was cloned by RT-PCR from Catimor coffee beans grown on Hainan Island of China. The vector for α-galactosidase cDNA expression was constructed and transferred into Pichia pastoris cells by electroporation. The transgenic cells were cloned by fermentation and the recombinant α-galactosidase was purified by ion exchange chromatography. After studying the biochemical characters of α-galactosidase, we have used it in converting human erythrocytes from group B to group O. Results The purity of recombinant α-galactosidase was higher than 96%, which was thought to be suitable for the use of blood conversion. Enzymatically converted human group O red blood cells （ECHORBC） exhibited membrane integrity, metabolic integrity, normal cell deformation and morphology. There were no coagulation between ECHORBC and any group of human blood. The ECHORBC will keep normal structure and function for a period of 21 days at 4℃ in monoammoniumphosphate nutrient solution. Experiments with Rhesus monkeys and gibbons showed that transfusion of enzymatically converted erythrocytes was safe. Conclusion ECHORBC can be easily obtained from group B red blood cell by α-galactosidase digestion. This study suggests that ECHORBC could be transfused to patients safely and efficiently.

Neutron resonance imaging(NRI)has recently emerged as an appealing technique for neutron *** complexity surpasses that of conventional transmission imaging,as it requires a high demand for both a neutron source and ***,the progression of NRI technology has been sluggish since its inception in the 1980s,particularly considering the limited studies analyzing the neutron energy range above *** white neutron source(Back-n)at the China Spallation Neutron Source(CSNS)provides favorable beam conditions for the development of the NRI technique over a wide neutron energy range from eV to ***-sensitive microchannel plates(MCP)have emerged as a cutting-edge tool in the field of neutron detection owing to their high temporal and spatial resolutions,high detection efficiency,and low *** this study,we report the development of a 10B-doped MCP detector,along with its associated electronics,data processing system,and NRI experiments at the *** heavy elements such as gold,silver,tungsten,and indium can be easily identified in the transmission images by their characteristic resonance peaks in the 1–100 eV energy range;the more difficult medium-weight elements such as iron,copper,and aluminum with resonance peaks in the 1–100 keV energy range can also be *** particular,results in the neutron energy range of dozens of keV(Aluminum)are reported here for the first time.
