Calcium-fortified fresh milk ameliorates postmenopausal osteoporosis via regulation of bone metabolism and gut microbiota in ovariectomized rats

The aging of the global population has made postmenopausal osteoporosis prevention essential;however,pharmacological treatments are ***,we evaluate the effect of calcium-fortified fresh milk(FM)in ameliorating postmenopausal osteoporosis in a rat model established using bilateral *** 3 months of FM(containing vitamin D,and casein phosphopeptides,1000 mg Ca/100 g)or control milk(110 mg Ca/100 g milk)supplementation,bone changes were assessed using dual-energy X-ray absorptiometry,microcomputed tomography,and bone biomechanical *** results revealed that FM can regulate bone metabolism and gut microbiota composition,which act on bone metabolism through pathways associated with steroid hormone biosynthesis,relaxin signaling,serotonergic synapse,and unsaturated fatty acid ***,FM administration significantly increased bone mineral content and density in the lumbar spine and femur,as well as femoral compressive strength,while improving femoral trabecular bone parameters and ***,we found that the effects may be due to increased levels of estrogen,bone formation marker osteocalcin,and procollagen typeⅠN-propeptide,and decreased expression of the bone resorption marker C-telopiptide and tartrate-resistant acid phosphatase ***,the findings suggest that FM is a potential alternative therapeutic option for ameliorating postmenopausal osteoporosis.

Diabetic osteoporosis(DOP) is the leading complication continuously threatening the bone health of patients with diabetes. A key pathogenic factor in DOP is loss of osteocyte viability. However, the mechanism of osteocyte death remains unclear. Here, we identified ferroptosis, which is iron-dependent programmed cell death, as a critical mechanism of osteocyte death in murine models of DOP. The diabetic microenvironment significantly enhanced osteocyte ferroptosis in vitro, as shown by the substantial lipid peroxidation, iron overload, and aberrant activation of the ferroptosis pathway. RNA sequencing showed that heme oxygenase-1(HO-1) expression was notably upregulated in ferroptotic osteocytes. Further findings revealed that HO-1 was essential for osteocyte ferroptosis in DOP and that its promoter activity was controlled by the interaction between the upstream NRF2 and c-JUN transcription factors. Targeting ferroptosis or HO-1 efficiently rescued osteocyte death in DOP by disrupting the vicious cycle between lipid peroxidation and HO-1 activation, eventually ameliorating trabecular deterioration. Our study provides insight into DOP pathogenesis, and our results provide a mechanism-based strategy for clinical DOP treatment.

慢性乙型肝炎疾病进展影响母婴健康。探讨摄入益生菌发酵乳对乙肝孕妇孕期便秘、血生化及血、尿常规的影响。纳入24-38岁之间,孕8-12周的志愿者,慢性乙肝孕妇11例为乙肝实验组,健康孕妇10例为健康实验组,10例健康孕妇为对照组进行队列跟踪研究。乙肝实验组和健康实验组志愿者每日摄入一瓶200 mL益生菌发酵乳,对照组志愿者每日摄入一瓶200 mL普通酸奶(不含有益生菌),均持续饮用16周。干预后乙肝组平均血红蛋白浓度水平上升,组间比较差异有统计学意义(P<0.05)。对照组红细胞计数上升,组间比较差异有统计学意义(P<0.05)。乙肝孕妇孕期补充益生菌发酵乳影响常规生化及健康指标,具有改善肝功能的潜在可能,可在临床中进行进一步探究。

合金钢、不锈钢等金属工程材料在服役过程中受到交变荷载以及腐蚀性介质的共同作用,使工程结构发生腐蚀疲劳,造成金属工程材料的承载能力降低和服役寿命缩短。因此,揭示载荷-环境体系下材料的腐蚀疲劳开裂规律对工程结构的前期设计及使用寿命预测都有深远意义。金属工程材料腐蚀疲劳失效的机制包括裂纹萌生和扩展两部分。目前的研究主要集中于裂纹扩展寿命,对裂纹萌生的研究较少。裂纹萌生机制仍存在争议,主要的观点有点蚀加速裂纹形成理论、滑移带优先溶解理论、保护膜破裂理论和吸附理论等。目前,关于腐蚀疲劳裂纹扩展机制的研究主要有阳极溶解和氢致开裂两种观点。影响腐蚀疲劳寿命的主要因素分为材料因素、力学因素及环境因素三方面。目前力学因素的研究主要集中于应力比、加载频率、加载波形等;材料因素的研究主要为显微组织及合金元素;环境因素的研究包括介质温度、浓度、含氧量、pH值及外加电位等。本文综述了金属工程材料腐蚀疲劳开裂机制及影响因素的研究现状,归纳了金属腐蚀疲劳的研究成果与不足,并在此基础上提出了下一步的研究方向及发展趋势,为相关领域的研究人员提供新思路。

Mammalian Sirtuin 6(SIRT6) plays an important role in the transcriptional silencing of silent mating-type loci,telomeres,and ***,the roles of porcine SIRT6 in cell proliferation are poorly understood,and a better knowledge of these will help improve our understanding of the biological mechanisms of cell growth and *** this study,a novel variant of porcine SIRT6(SIRT6 V2) identified by reverse transcription-polymerase chain reaction and BLAST analysis showed a 124-bp deletion compared to wild-type SIRT6 mRNA(SIRT6 V1).Two recombinant plasmids overexpressing SIRT6 V1 and SIRT6 V2 were produced and their roles in the proliferation of porcine fetal fibroblasts(PFFs) were *** transfected with SIRT6 V1 proliferated significantly faster than those transfected with SIRT6 V2(P<0.01),though both proliferated significantly faster than cells expressing an empty vector(P<0.01).These results might be caused by altered proportions of α helices and β sheets in the SIRT6 V2 structure relative to SIRT6 *** results indicate that overexpression of SIRT6 V1/V2 was involved in promoting PFF *** of a 124-bp sequence attenuated the effects of SIRT6 on cell proliferation,possibly as a result of changes in the proportions of α helices and β sheets in the protein secondary structure.

The cyclic plastic straining electrode technique has been used to investigate the transient electrochemical behaviour of Fe-26Cr1Mo stainless steel in 1M H2SO4 solution at a passive *** influence of plastic strain amplitude and plastic strain rate on the dissolution current response was analysed. The experimental results showed that the transient current was dependent on the competitive process of the surface film rupture and repassivation of the new surface. 采用疲劳实验研究了不同加载波形下X65钢在空气和海水中的疲劳行为。结合SEM结果,对疲劳断口和次生裂纹进行了观察。结果表明,在空气和海水中,正锯齿波加载下X65钢的疲劳寿命最大,三角波次之,正弦波最短。与在空气中相比,X65钢在海水中的疲劳寿命显著降低。正弦波与三角波加载时应力上升时间较短,有利于位错开动,加快裂纹萌生。其中正弦波在σ_(max)的保载时间最长,有利于位错滑移形成,疲劳裂纹扩展速度最快。在海水环境中,Cl^(-)促进了X65钢表面点蚀萌生,成为腐蚀疲劳裂纹源。当裂纹形成后,电解质进入裂纹间隙,在交变应力作用下裂纹反复张开与闭合,导致裂纹快速扩展。在海水中,当加载波形为正锯齿波时,X65钢的腐蚀疲劳裂纹的扩展机制为阳极溶解,而当加载波形为正弦波和三角波时,X65钢的腐蚀疲劳的扩展机制均为氢脆+阳极溶解混合机制,其中加载正弦波时腐蚀疲劳开裂敏感性最大。
