Hypoxia-preconditioned bone marrow-derived mesenchymal stem cells protect neurons from cardiac arrest-induced pyroptosis

Cardiac arrest can lead to severe neurological impairment as a result of inflammation,mitochondrial dysfunction,and post-cardiopulmonary resuscitation neurological *** preconditioning has been shown to improve migration and survival of bone marrow–derived mesenchymal stem cells and reduce pyroptosis after cardiac arrest,but the specific mechanisms by which hypoxia-preconditioned bone marrow–derived mesenchymal stem cells protect against brain injury after cardiac arrest are *** this end,we established an in vitro co-culture model of bone marrow–derived mesenchymal stem cells and oxygen–glucose deprived primary neurons and found that hypoxic preconditioning enhanced the protective effect of bone marrow stromal stem cells against neuronal pyroptosis,possibly through inhibition of the MAPK and nuclear factor κB ***,we transplanted hypoxia-preconditioned bone marrow–derived mesenchymal stem cells into the lateral ventricle after the return of spontaneous circulation in an 8-minute cardiac arrest rat model induced by *** results showed that hypoxia-preconditioned bone marrow–derived mesenchymal stem cells significantly reduced cardiac arrest–induced neuronal pyroptosis,oxidative stress,and mitochondrial damage,whereas knockdown of the liver isoform of phosphofructokinase in bone marrow–derived mesenchymal stem cells inhibited these *** conclude,hypoxia-preconditioned bone marrow–derived mesenchymal stem cells offer a promising therapeutic approach for neuronal injury following cardiac arrest,and their beneficial effects are potentially associated with increased expression of the liver isoform of phosphofructokinase following hypoxic preconditioning.

Dynamic Deformation Behaviors and Constitutive Relations of High-Strength Weldox700E Steel

High-strength Weldox700E steel has been increasingly applied to dynamic environment because of its high strength and good toughness. The plastic deformation behaviors of Weldox700E steel at strain rates ranging from 10^-4 to 6200 s^-1 are investigated by the quasistatic and dynamic uniaxial compression tests. The Weldox700E steel exhibits rate-related plastic behavior, work hardening and thermal softening behaviors. Due to the nonlinear strain rate effect of the material and the adiabatic temperature rise caused by high-speed impact compression, the properties of material are greatly affected. By improving the strain rate enhancement term and temperature term in the Johnson-Cook constitutive model, a new constitutive model was proposed to describe the dynamic mechanical behavior of Weldox700E steel. This constitutive equation was implemented into the finite element software, ABAQUS, via an explicit user material subroutine utilizing the stress update algorithm. The simulation results at different strain rates were in good agreement with the experimental data, verifying the validity of the constitutive model.

Influence of colloidal nanosilica on hydration kinetics and properties of CaO/CaSO_(4)-activated slag binder

To solve the energy consumption and CO_(2) emission during cement production,the new binders must be developed as an alternative to ***/CaSO_(4)-activated slag binder is an eco-friendly and safe cementitious material;however,its low strength during initial stages limits its *** this study,colloidal nanosilica(CNS)was employed as an additive to improve the strength of CaO/CaSO_(4)-activated slag binder,and the effects of CNS on the workability,hydration kinetics,hydration products(type,quantity,and polymerization degree),and binder microstructure were thoroughly investigated.A moderate CNS content,through its nucleation effect,significantly increased the hydration rate of the nucleation and crystal growth(NG),phase boundary interaction(I)and diffusion(D)processes,which generated large quantities of calcium aluminosilicate hydrate(C-A-S-H)gel in the initial hydration ***,the addition of CNS improved the polymerization degree of C-A-S-H *** amorphous reactant well-filled the pore space between slag particles and yielded a compact microstructure,consequently enhancing the binder *** the reduction in fluidity and the increase in production cost,the CNS mass fraction was controlled as3%,and the binder reached the satisfactory strengths of 3.87,24.47,31.43,and 41.78 MPa at 1,3,7,and 28 d,respectively.

Tissue distribution of cadmium and its effect on reproduction in Spodoptera exigua

Vegetable fields are often contaminated by heavy metals,and Spodoptera exigua is a major vegetable pest which is stressed by heavy metals mainly by *** this study,cadmium accumulation in the tissues of *** exposed to cadmium and its effects on the growth and development of the parents and the offspring were *** the stress of different concentrations of cadmium(0.2,3.2,and 51.2 mg kg^(-1)),the cadmium content in each tissue of *** increased in a dose-dependent *** the larval stage,the highest cadmium accumulation was found in midgut in all three cadmium treatments,but at the adult stage,the highest cadmium content was found in fat *** addition,the cadmium content in ovaries was much higher than in *** *** was stressed by cadmium and the F_(2)generation was not fed a cadmium-containing diet,the larval survival,pupation rate,emergence rate and fecundity of the F_(2)generation were significantly reduced in the 51.2 mg kg^(-1)treatment compared to the corresponding *** in the F_(2)generation of the 3.2 mg kg^(-1)treatment,the fecundity was significantly lower than in the parental *** fecundity of the only-female stressed treatment was significantly lower than that of the only-male stressed treatment at the 3.2 and 51.2 mg kg^(-1)cadmium exposure *** only mothers were stressed at the larval stage,the fecundity of the F_(2)generation was significantly lower than that of the F1generation in the 51.2 mg kg^(-1)treatment,and it was also significantly lower than in the 3.2 and 0.2 mg kg^(-1)*** results of our study can provide useful information for forecasting the population increase trends under different heavy metal stress conditions and for the reliable environmental risk assessment of heavy metal pollution.

The G-protein α subunit GhGPA positively regulates Gossypium hirsutum resistance to Verticillium dahliae via induction of SA and JA signaling pathways and ROS accumulation

Verticillium wilt, a devastating disease in cotton caused by Verticillium dahliae, reduces cotton quality and yield. Heterotrimeric GTP-binding proteins, consisting of Ga, Gb, and Gc subunits, transducers of receptor signaling, function in a wide range of biological events. However, the function of Ga proteins in the regulation of defense responses in plants is largely unexplored, except for a few reports on model species. In the present study, a cotton G-protein a-subunit-encoding gene(GhGPA) was isolated from Verticillium wilt-resistant Gossypium hirsutum(upland cotton) cv. ND601. GhGPA transcription was up-regulated under V. dahliae stress, with higher expression in tolerant than in susceptible cotton *** localization revealed GhGPA to be located in the plasma membrane. GhGPA shows high(85.0%) identity with Arabidopsis AT2 G26300(AtGPA1), and AtGPA1 gpa1-4 mutants displayed susceptibility to V. dahliae. Ectopic expression of GhGPA successfully restored the resistance of Arabidopsis gpa1-4 mutants to Verticillium wilt and made them more resistant than the wild type. Overexpression of GhGPA in Arabidopsis markedly increased the resistance and resulted in dramatic up-regulation of pathogenesis-related(PR) genes and increased in H2 O2 accumulation and salicylic acid(SA) and jasmonic acid(JA) contents. However, suppressing GhGPA expression via virus-induced gene silencing(VIGS)increased susceptibility to Verticillium wilt, down-regulated the expression of PR and marker genes in SA and JA signaling pathways, and reduced H2 O2 content. The contents of SA and JA in Arabidopsis gpa1-4 and VIGS cotton were lower than those in the wild type and empty-vector control. However,GhGPA-overexpressing Arabidopsis contained more SA and JA than the wild type when inoculated with V. dahliae. Thus, GhGPA plays a vital role in Verticillium wilt resistance by inducing SA and JA signaling pathways and regulating the production of reactive oxygen species. These findings

In this work,we discovered an unexpected mechanoluminescence (ML) phenomena occurring when transforming amorphous into crystalline,due to the stress-induced precipitation of CsPbBr3 perovskite nanocrystals on glass *** is revealed that,unlike the conventional thermal-induced phase transformation mechanism,the breakage of bonding of glass network provides the energy for nucleation and growth,and the shear stress avoids the long-range migration of structural units for *** unique ML phenomenon enables the visualization of dynamical force that is inaccessible by common strategy,and so,opens up some novel applications,such as the pressure-sensitive "glassy pencil" to learn people's writing habits,and the pb^2+-detection with good sensitivity and *** findings not only demonstrate an effective route for the preparation of perovskite materials in a green,time-saving,low cost,and scalable way,enrich the knowledge of glass crystallization mechanism,but also exploit a useful avenue to quantitatively visualize the dynamical force.

As one of the most promising materials for two-dimensional transition metal chalcogenides(2D TMDs),molybdenum diselenide(MoSe_(2))has great potential in photodetectors due to its excellent properties like tunable bandgap,high carrier mobility,and excellent air *** 2D MoSe_(2)-based photodetectors have been reported to exhibit admired performance,the large-area 2D MoSe_(2)layers are difficult to be achieved via conventional synthesis methods,which severely impedes its future ***,we present the controllable growth of large-area 2D MoSe_(2)layers over 3.5-inch with excellent homogeneity by a simple post-selenization ***,a high-quality n-MoSe_(2)/p-Si van der Waals(vdW)heterojunction device is in-situ fabricated by directly growing 2D n-MoSe_(2)layers on the patterned p-Si substrate,which shows a self-driven broadband photoresponse ranging from ultraviolet to mid-wave infrared with an impressive responsivity of 720.5 mA·W^(−1),a high specific detectivity of 10^(13) Jones,and a fast response time to follow nanosecond pulsed optical *** addition,thanks to the inch-level 2D MoSe_(2)layers,a 4×4 integrated heterojunction device array is achieved,which has demonstrated good uniformity and satisfying imaging *** large-area 2D MoSe_(2)layer and its heterojunction device array have great promise for high-performance photodetection and imaging applications in integrated optoelectronic systems.

Changes of the Thermal Environment Caused by a City-Wide Lockdown:The Case of Wuhan City

The impact of the COVID-19 epidemic in Wuhan caused the city to go into a lockdown in January,2020,and most outdoor activities stopped,which provided a chance to investigate the impact of human activities on the urban thermal *** the land surface temperature(LST)and population distribution data,this paper analyzes the changes between normal workdays and the lockdown *** results show that maximum population and LST changes can be seen in industrial and commercial areas,and the LST hot spots shrink and cold spots expand in many areas during the lockdown,which identifes that the impact of the lockdown on the urban thermal environment varies according to different types of ***,it should be noted that the LST has a similar spatial distribution during the two periods,in spite of the signifcant changes in population *** with population size,the urban morphology indicators have a higher correlation with the *** indicates that the urban thermal environment is dominated by urban morphology,and the infuence of human activities on it is still *** fndings can help urban planners to improve the existing methods to address both climate change and public health issues.
