Activation of skeletal carbohydrate-response element binding protein(ChREBP)-mediated de novo lipogenesis increases intramuscular fat content in chickens

The intracellular lipids in muscle cells of farm animals play a crucial role in determining the overall intramuscular fat(IMF)content,which has a positive impact on meat ***,the mechanisms underlying the deposition of lipids in muscle cells of farm animals are not yet fully *** purpose of this study was to determine the roles of carbohydrate-response element binding protein(ChREBP)and fructose in IMF deposition of *** virus-mediated ChREBP overexpression in tibialis anterior(TA)muscle of chickens,seven 5-d-old male yellow-feather chickens were *** 1. d after virus injection,the chickens were slaughtered to obtain TA muscles for *** fructose administration trial,sixty 9-wk-old male yellow-feather chickens were randomly divided into 2 groups,with 6 replicates per group and 5 chickens per *** chickens were fed either a basal diet or a basal diet supplemented with 1.%fructose(purity≥99%).At 4 wk later,the chickens were slaughtered,and breast and thigh muscles were collected for *** results showed that the skeletal ChREBP mRNA levels were positively associated with IMF content in multiple species,including the chickens,pigs,and mice(P<0.05).ChREBP overexpression increased lipid accumulation in both muscle cells in vitro and the TA muscles of mice and chickens in vivo(P<0.05),by activation of the de novo lipogenesis(DNL)***,activation of ChREBP by dietary fructose administration also resulted in increased IMF content in mice and notably chickens(P<0.05).Furthermore,the lipidomics analysis revealed that ChREBP activation altered the lipid composition of chicken IMF and tented to improve the flavor profile of the *** conclusion,this study found that ChREBP plays a pivotal role in mediating the deposition of fat in chicken muscles in response to fructose-rich diets,which provides a novel strategy for improving meat quality in the livestock industry.

Efficient Polytelluride Anchoring for Ultralong-Life Potassium Storage: Combined Physical Barrier and Chemisorption in Nanogrid-in-Nanofiber

Metal tellurides(MTes) are highly attractive as promising anodes for high-performance potassium-ion batteries. The capacity attenuation of most reported MTe anodes is attributed to their poor electrical conductivity and large volume variation. The evolution mechanisms, dissolution properties, and corresponding manipulation strategies of intermediates(K-polytellurides, K-pTe_(x)) are rarely mentioned. Herein,we propose a novel structural engineering strategy to confine ultrafine CoTe_(2) nanodots in hierarchical nanogrid-in-nanofiber carbon substrates(CoTe_(2)@NC@NSPCNFs) for smooth immobilization of K-pTe_(x) and highly reversible conversion of CoTe_(2) by manipulating the intense electrochemical reaction process. Various in situ/ex situ techniques and density functional theory calculations have been performed to clarify the formation, transformation, and dissolution of K-pTe_(x)(K_(5)Te_(3) and K_(2)Te), as well as verifying the robust physical barrier and the strong chemisorption of K_(5)Te_(3) and K_(2)Te on S, N co-doped dual-type carbon substrates. Additionally, the hierarchical nanogrid-in-nanofiber nanostructure increases the chemical anchoring sites for K-pTe_(x), provides sufficient volume buffer space, and constructs highly interconnected conductive microcircuits, further propelling the battery reaction to new heights(3500 cycles at 2.0 A g^(-1.). Furthermore, the full cells further demonstrate the potential for practical applications. This work provides new insights into manipulating K-pTe_(x) in the design of ultralong-cycling MTe anodes for advanced PIBs.

Heavy-atom engineered hypoxia-responsive probes for precisive photoacoustic imaging and cancer therapy

Photoacoustic agents combining photodynamic therapy(PDT) and photothermal therapy(PTT) functions have emerged as potent theranostic agents for combating cancer. The molecular approaches for enhancing the near-infrared(NIR)-absorption and maximizing non-radiative energy transfer are essential for effective photoacoustic imaging(PAI) and therapy applications. In addition, such molecules with high specificity and affinity to cancer cells are urgently needed, which would further decrease the side effect during treatments. In this study, we applied a heavy-atom engineering strategy and introduced p-aminophenol,-thio, and-seleno moieties into NIR heptamethine cyanine(Cy7) skeleton(Cy7-X-NH_(2), X = O, S, Se) to significantly increase photothermal conversion efficiency for PTT and promote intersystem crossing for ***, we designed a series of nitroreductase(NTR)-activated photoacoustic probes(Cy7-X-NO_(2),X = O, S, Se), and target hypoxic tumors with NTR overexpression. Our prostate cancer targeting probe,Cy7-Se-NO_(2)-KUE, exhibited specific tumor photoacoustic signals and effective tumor killing through outstanding synergistic PTT/PDT in vivo. These findings highlighted a versatile strategy for cancer photoacoustic diagnosis and enhanced phototherapy.

目的比较刮吸解剖法和传统电刀法在乳腺癌、肺癌、食管癌手术中的应用情况。方法收集201.年1.月至201.年6月南通市肿瘤医院收治的98例乳腺癌、肺癌、食管癌手术患者的临床资料。48例行刮吸解剖法手术,为刮吸解剖组,其中乳腺癌1.例、肺癌1.例、食管癌1.例;50例行传统电刀法手术,为传统电刀组,其中乳腺癌1.例、肺癌1.例、食管癌1.例。对两组的术中出血量、手术时间及淋巴结清扫数目进行比较。结果刮吸解剖组:乳腺癌术中出血量为(33.21.9.32)ml,手术时间为(65.71.6.75)min,淋巴结清扫数目为(1..1.±2.32)个;肺癌术中出血量为(296.25±67.71.ml,手术时间为(1.4.38±1..02)min,淋巴结清扫数目为(1..63±3.30)个;食管癌术中出血量为(271.67±48.40)ml,手术时间为(1.6.39±1..40)min,淋巴结清扫数目为(1..78±2.02)个。传统电刀组:乳腺癌术中出血量为(51.25±1..72)ml,手术时间为(73.44±9.61.min,淋巴结清扫数目为(9.25±2.24)个;肺癌术中出血量为(397.65±72.40)ml,手术时间为(1.5.59±1..1.)min,淋巴结清扫数目为(1..06±2.86)个;食管癌术中出血量为(336.47±71.40)ml,手术时间为(1.1.76±1..1.)min,淋巴结清扫数目为(1..06±2.01.个。两组均无围手术期死亡病例,刮吸解剖组优于传统电刀组,在术中出血量、手术时间、淋巴结清扫数目方面两组比较,差异有统计学意义(均P<0.05)。结论刮吸解剖法用于乳腺癌、肺癌、食管癌手术安全,可减少术中出血,加快手术速度,提高手术质量。
