The CD19-targeting bispecific T-cell engager blinatumomab has shown remarkable efficacy in patients with relapsed/refractory B-cell precursor acute lymphoblastic leukemia.However,several studies showed that blinatumomab has a short plasma half-life due to its low molecular weight,and thus its clinical use is limited.Furthermore,multiple trials have shown that approximately 30%of blinatumomab-relapsed cases are characterized by CD19 negative leukemic cells.Here,we design and characterize two novel antibodies,A-319 and A-2019.Blinatumomab and A-319 are CD3/CD19 bispecific antibodies with different molecular sizes and structures,and A-2019 is a novel CD3/CD19/CD20 trispecific antibody with an additional anti-CD20 function.Our in vitro,ex vivo,and in vivo experiments demonstrated that A-319 and A-2019 are potent antitumor agents and capable of recruiting CD3 positive T cells,enhancing T-cell function,mediating B-cell depletion,and eventually inhibiting tumor growth in Raji xenograft models.The two molecules are complementary in terms of efficacy and specificity profile.The activity of A-319 demonstrated superior to that of A-2019,whereas A-2019 has an additional capability to target CD20 in cells missing CD19,suggesting its potential function against CD19 weak or negative CD20 positive leukemic cells.

DEAR EDITOR Centenarians are a typical longevity model characterized by delayed onset of morbidity in age-related diseases such as cancer, cardiovascular disease, dementia, and stroke （Andersen et al, 2012）. Though there may be a number of underlying mechanisms behind this longevity, curiously it seems that the survival advantage persists in their offspring （Terry et al, 2003）, suggesting a potentially important role for genetic factors. Previous studies suggested that the heritability of human longevity may be ~25% （Herskind et al, 1996; Mcgue et al, 1993）, whicih is consistent with other studies on model organisms that identified several longevity-related genes, such as age-l, daf-2, daf-16, and sir-2 （Friedman ＆ Johnson, 1988; Kenyon et al, 1993; Lin et al, 1997; Tissenbaum ＆ Guarente, 2001）. Likewise, several studies have reported the existence of many mutations related to human longevity （Holstege et al, 2014; Sebastiani et al, 2012）.

In view of the geographically closest location to Andaman archipelago, Myanmar was suggested to be the origin place of aboriginal Andamanese. However, for lacking any genetic information from this region, which has prevented to resolve the dispute on whether the aboriginal Andamanese were originated from mainland India or Myanmar. To solve this question and better understand the origin of the aboriginal Andamanese, we screened for haplogroups M31 （from which Andaman-specific lineage M31al branched off） and M32 among 846 mitochondrial DNAs （mtDNAs） sampled across Myanmar. As a result, two Myanmar individuals belonging to haplogroup M31 were identified, and completely sequencing the entire mtDNA genomes of both samples testified that the two M31 individuals observed in Myanmar were probably attributed to the recent gene flow from northeast India populations. Since no root lineages of haplogroup M31 or M32 were observed in Myanmar, it is unlikely that Myanmar may serve as the source place of the aboriginal Andamanese. To get further insight into the origin of this unique population, the detailed phylogenetic and phylogeographic analyses were performed by including additional 7 new entire mtDNA genomes and 113 M31 mtDNAs pinpointed from South Asian populations, and the results suggested that Andaman-specific M31al could in fact trace its origin to northeast India. Time estimation results further indicated that the Andaman archipelago was likely settled by modem humans from northeast India via the land-bridge which connected the Andaman archipelago and Myanmar around the Last Glacial Maximum （LGM）, a scenario in well agreement with the evidence from linguistic and palaeoclimate studies.

Tailoring thermal conductivity of bulk graphene oxide by tuning the oxidation degree

Bulk graphene oxide （GO） shows great potential in a variety of applications, such as sensors,photodetectors, supercapacitors, lithium ion batteries and catalysts. However, its thermal conductivity,one of the most important and fundamental physical properties, is still less known. Herein, we havesystematically investigated the thermal conductivity of bulk GOs and find that it can be tailored by tuningtheir oxidation degree during preparation process. Notably, the cross-plane thermal conductivity of bulkGO, in comparison with its precursor graphite, exhibits more than 100 times decrease at roomtemperature. The dependence of thermal conductivity of GO on oxidation degree is attributed to thechemical and structural changes by introducing oxygen atoms and oxygen-containing functional groups,which can lead to a significant enhancement in atomic- and nano-scale phonon scattering. Furthermore,we reveal that the thermal conductivity of bulk GOs exhibits evident anisotropic behavior. These resultsprovide fundamental understanding and valuable information on thermal transport properties of bulkGOs for various practical applications.

Automatic segmentation of the liver and hepatic lesions from abdominal 3D comput-ed tomography(CT)images is fundamental tasks in computer-assisted liver surgery planning.However,due to complex backgrounds,ambiguous boundaries,heterogeneous appearances and highly varied shapes of the liver,accurate liver segmentation and tumor detection are stil-1 challenging problems.To address these difficulties,we propose an automatic segmentation framework based on 3D U-net with dense connections and globally optimized refinement.First-ly,a deep U-net architecture with dense connections is trained to learn the probability map of the liver.Then the probability map goes into the following refinement step as the initial surface and prior shape.The segmentation of liver tumor is based on the similar network architecture with the help of segmentation results of liver.In order to reduce the infuence of the surrounding tissues with the similar intensity and texture behavior with the tumor region,during the training procedure,I x liverlabel is the input of the network for the segmentation of liver tumor.By do-ing this,the accuracy of segmentation can be improved.The proposed method is fully automatic without any user interaction.Both qualitative and quantitative results reveal that the pro-posed approach is efficient and accurate for liver volume estimation in clinical application.The high correlation between the automatic and manual references shows that the proposed method can be good enough to replace the time-consuming and non-reproducible manual segmentation method.

Difference of Clinical Characteristics in Patients with Omicron and Delta Variants of SARS-CoV-2 in Beijing,China

Background Delta and Omicron are two main variants that have been prevalent since 2021.However,the Omicron variant of severe acute respiratory syndrome coronavirus 2 shows a less severe clinical presentation and high transmissibility.Therefore,we carried out this retrospective study to evaluate Omicron severity compared with the Delta variant and further comprehend the differences in clinical characteristics in patients with the Omicron variant.Methods We extracted clinical data and compared clinical severity,symptoms,vaccination status,laboratory parameters,viral shedding time,and computed tomography(CT)imaging between the two groups of patients,which included 109 COVID-19 cases with the Delta variant and 183 cases with the Omicron variant,from January 19 to April 1,2022,in Beijing Ditan Hospital.In addition,the Beijing Center for Disease Prevention and Control conducted whole-genome sequencing.Results We obtained 94 strains of variants of concern/Delta and 110 strains of variants of concern/Omicron.For the 110 Omicron strains,three were assigned as BA.1.1,53 as BA.2,and 54 as BA.2.2.Among patients with the Delta variant,54%(59/109)were moderate,which was significantly higher than that of patients with the Omicron variant(7%(12/183),P<0.001).The number of patients with mild symptoms in the Omicron group was significantly higher than in the Delta group(80%vs.35%,P<0.001).Compared with the Omicron group,patients with underlying diseases or obesity,60 years or older,or unvaccinated in the Delta group had more severe disease,and there was a significant difference between the two groups.The viral shedding time in the Omicron group was shorter than in the Delta group((11.9±5.9)vs.(14.0±5.8)days,P=0.003).Among the 183 patients in the Omicron group,104(57%)had dry or sore throat symptoms,more than those in the Delta group(34%(37/109);P<0.001).In the Delta group,patients in the moderate group had more fever and cough symptoms than those in the mild group.The remission time of CT imaging in the Omicron group was shorter t

Soybean cyst nematode Heterodera glycines is one of the most serious soil-borne pathogens in soybean production. However, the researches were limited in China due to lack of an effective pathosystem. In this study, we screened 21 legume Medicago plants in both Medicago truncatula and Medicago sativa to obtain candidate model plants for establishing a new pathosystem for legume-H. glycines interactions. The nematode infection of tested plants was assayed with Race 3 and 4 respectively, which were two dominant H. glycines inbred races in China soybean producing areas. The results showed that the model legume plant M. truncatula A17 failed to allow Race 3 of H. glycines to complete its life cycle, in contrast, it provided the Race 4 population to form several cyst nematodes, however, the female index（FI） value was approximately 1.6. Three M. sativa cultivars, including Xunlu, Aergangjin and Junren, provided either Race 3 or 4 of H. glycines to develop into mature cysts with their FI value below 5 as well. Our results demonstrated that legume plants in both M. truncatula and M. sativa were not likely to be a model plant for H. glycines because of an extreme high resistance.
