Bradykinin postconditioning protects rat hippocampal neurons after restoration of spontaneous circulation following cardiac arrest via activation of the AMPK/mTOR signaling pathway

Bradykinin(BK)is an active component of the kallikrein-kinin system that has been shown to have cardioprotective and neuroprotective effects.We previously showed that BK postconditioning strongly protects rat hippocampal neurons upon restoration of spontaneous circulation(ROSC)after cardiac arrest.However,the precise mechanism underlying this process remains poorly understood.In this study,we treated a rat model of ROSC after cardiac arrest(induced by asphyxiation)with 150μg/kg BK via intraperitoneal injection 48 hours after ROSC following cardiac arrest.We found that BK postconditioning effectively promoted the recovery of rat neurological function after ROSC following cardiac arrest,increased the amount of autophagosomes in the hippocampal tissue,inhibited neuronal cell apoptosis,up-regulated the expression of autophagy-related proteins LC3 and NBR1 and down-regulated p62,inhibited the expression of the brain injury marker S100βand apoptosis-related protein caspase-3,and affected the expression of adenosine monophosphate-activated protein kinase/mechanistic target of rapamycin pathway-related proteins.Adenosine monophosphate-activated protein kinase inhibitor compound C clearly inhibited BK-mediated activation of autophagy in rats after ROSC following cardiac arrest,which aggravated the injury caused by ROSC.The mechanistic target of rapamycin inhibitor rapamycin enhanced the protective effects of BK by stimulating autophagy.Our findings suggest that BK postconditioning protects against injury caused by ROSC through activating the adenosine monophosphate-activated protein kinase/mechanistic target of the rapamycin pathway.

An optimization method for passive muzzle arc control devices in augmented railguns

In this paper,a passive muzzle arc control device(PMACD)of the augmented railguns is studied.By discussing its performance at different numbers of extra rails,a parameter optimization model is proposed.Through the calculation model,it is found that the PMACD works well in the simple railgun,which refers to the gun that there is only one pair of rails in the inner bore.The PMACD may decrease the simple railgun’s armature peak current and muzzle arc,but affect its muzzle velocity not much.However,in the augmented railguns it has different characteristics.If the parameters of the PMACD are not selected suitable.It may increase the armature peak current and muzzle arc,but greatly decrease the velocity.The reason for this problem is that the extra rails generate a strong magnetic field in front of the armature,which induces a large current to change the armature current.It is also found that when the resistance and inductance parameters of the PMACD satisfy with the optimization formula,the PMACD can also play a good role in arc suppression in the augmented railguns.Experiments of an augmented railgun with a stainless steel PMACD are carried out to verify this optimization method.Results show that the muzzle arc is obviously controlled.This work may provide a reference for the design of the muzzle arc control device.

Bone marrow-derived mesenchymal stem cell transplantation attenuates overexpression of inflammatory mediators in rat brain after cardiopulmonary resuscitation

Emerging evidence suggests that bone marrow-derived mesenchymal stem cell transplantation improves neurological function after cardiac arrest and cardiopulmonary resuscitation;however, the precise mechanisms remain unclear. This study aimed to investigate the effect of bone marrow-derived mesenchymal stem cell treatment on expression profiles of multiple cytokines in the brain after cardiac arrest and cardiopulmonary resuscitation. Cardiac arrest was induced in rats by asphyxia and cardiopulmonary resuscitation was initiated 6 minutes after cardiac arrest. One hour after successful cardiopulmonary resuscitation, rats were injected with either phosphate-buffered saline(control) or 1 × 10~6 bone marrow-derived mesenchymal stem cells via the tail vein. Serum S100 B levels were measured by enzyme-linked immunosorbent assay and neurological deficit scores were evaluated to assess brain damage at 3 days after cardiopulmonary resuscitation. Serum S100 B levels were remarkably decreased and neurological deficit scores were obviously improved in the mesenchymal stem cell group compared with the phosphate-buffered saline group. Brains were isolated from the rats and expression levels of 90 proteins were determined using a RayBio Rat Antibody Array, to investigate the cytokine profiles. Brain levels of the inflammatory mediators tumor necrosis factor-α, interferon-γ, macrophage inflammatory protein-1α, macrophage inflammatory protein-2, macrophage inflammatory protein-3α, macrophage-derived chemokine, and matrix metalloproteinase-2 were decreased ≥ 1.5-fold, while levels of the anti-inflammatory factor interleukin-10 were increased ≥ 1.5-fold in the mesenchymal stem cell group compared with the control group. Donor mesenchymal stem cells were detected by immunofluorescence to determine their distribution in the damaged brain, and were primarily observed in the cerebral cortex. Numerical simulation of dynamic large deformation and fracture damage for solid armature in electromagnetic railgun

The design of solid armature of railgun should take full account of its operating conditions and material properties because the armature is subjected to dynamic loading conditions and experiences a complicated electrical,thermal and mechanical process in the interior ballistic cycle.In this paper present,we first introduced a multi-physical field model of railgun,followed by several examples to investigate the launching process.Especially,we used the explicit finite element method,in which material nonlinearity and geometric nonlinearity were accounted,to investigate the deform behaviors of solid armature.The results show that the dynamic mechanical process of armature is dependent on the armature geometry,material and exciting electric current.By the numerical simulation,the understanding of the fracture mechanism of solid armature was deepened.

Fatty acid-binding protein gene is indispensable for molting process in Heortia vitessoides(Lepidoptera:Crambidae)

As intracellular fatty acid(FA) carriers,FA-binding proteins(FABPs) widely participate in the absorption,transport,and metabolism of FAs.It is a key protein in insect lipid metabolism and plays an important role in various physiological activities of insects.An FABP gene(HvFABP) was cloned from the transcriptional library of Heortia vitessoides Moore(Lepidoptera:Crambidae),and its expression patterns were determined using reverse transcription quantitative PCR(RTqPCR).Stage-and tissue-specific expression profiles indicated that HvFABP highly expressed from prepupal to adult stages and in larval midgut and adult wings.HvFABP expression may be induced through starvation,mRNA expression was downregulated at 24 and 48 h and upregulated at 72 h after starvation.Furthermore,20-hydroxyecdysone can induce the upregulation of its expression.RNA interference-mediated silencing of Hv FABP significantly inhibited HvFABP expression,resulting in delayed development,abnormal molting or lethal phenotypes,and a significantly reduced survival rate.These results indicate that HvFABP plays a key role in the molting of H.vitessoides.

Dense Face Network:A Dense Face Detector Based on Global Context and Visual Attention Mechanism

Face detection has achieved tremendous strides thanks to convolutional neural networks. However, dense face detection remains an open challenge due to large face scale variation, tiny faces, and serious occlusion. This paper presents a robust, dense face detector using global context and visual attention mechanisms which can significantly improve detection accuracy. Specifically, a global context fusion module with top-down feedback is proposed to improve the ability to identify tiny faces. Moreover, a visual attention mechanism is employed to solve the problem of occlusion. Experimental results on the public face datasets WIDER FACE and FDDB demonstrate the effectiveness of the proposed method.

Analysis and discussion on launching mechanism and tactical electromagnetic railgun technology

This paper begins with a discussion on the significance of multi-physical fields in the research of railgun launching mechanism, followed by an introduction of research work about dynamic measurements and numerical simulations. The application of some measurement methods including the atomic emission spectrum, fiber-optic strain, optical level, pulsed X-ray, and high-speed video in observing the launching process was introduced. The models about the electromechanical dynamic processes and multi-physical fields in a railgun were developed. The mechanisms of the grooving, arc transition, and gouging were analyzed. These work provided a deeper understanding of launching mechanism of electromagnetic railgun. Some issues about the tactical railgun, such as the fiber composite overwrapped barrel, recoil,loading modes and eddy current loss were also discussed. Taking a medium-caliber launcher as an example, the basic process of design and analysis was introduced.

A midgut-specific lytic polysaccharide monooxygenase of Locusta migratoria is indispensable for the deconstruction of the peritrophic matrix

Lytic polysaccharide monooxygenases(LPMOs)are important enzymes that boost the hydrolysis of recalcitrant polysaccharides,such as chitin.They are found extensively in different insect species and are classified as auxiliary activities family 15(AA15)LPMOs(LPMO15).Some of them were identified from the insect midgut and proven to act on chitin.However,knowledge about their physiological roles during insect growth and development remains limited.Here,we found that midgut-specific LPMO15s are widely distributed in different insect orders,such as the orthopteran Locusta migratoria and the lepidopteran Bombyx mori.Using L.migratoria as a model insect,the function of midgut-specific LmLPMO15-3 during development was investigated.Double-stranded RNA-mediated downregulation of LmLPMO15-3 expression at the 4th or 5th instar nymph stage severely decreased the survival rate and resulted in lethal phenotypes.Hematoxylin and eosin staining results indicated that the deficient individuals exhibited incompletely digested peritrophic matrix(PM),which suggested that LmLPMO15-3 is essential for the deconstruction of the PM during molting.This study provides direct evidence of the physiological importance of a midgut-specific LPMO15 during insect development.As L.migratoria is one of the most destructive agricultural pests,LmLPMO15-3 is a potential target for pest management.

Irritable bowel syndrome(IBS)is a common disorder,reported to be found in 5%-20%of the general population.Its management accounts for up to 25%of a gastroenterologist’s workload in the outpatient department,and the main symptoms are abdominal pain,bloating,and altered bowel habits.Despite a great amount of available pharmacological treatments aimed at a wide variety of gastrointestinal and brain targets,many patients have not shown adequate symptom relief.In recent years,there has been increasing evidence to suggest that psychological treatments,in particular cognitive-behavioral therapy(CBT),are effective for the management of IBS.This review discusses CBT for the management of IBS.CBT has proved to be effective in alleviating the physical and psychological symptoms of IBS and has thus been recommended as a treatment option for the syndrome.

Intravenous injection of Xuebijing attenuates acute kidney injury in rats with paraquat intoxication

BACKGROUND: The study aimed to investigate the therapeutic benefits of intravenous Xuebijing on acute kidney injury(AKI) in rats with paraquat intoxication.METHODS: Male Sprague-Dawley rats were randomly divided equally into three groups: sham group(n=8), paraquat group(n=8) and Xuebijing-treated group(n=8) using a random number table. The rats were intraperitoneally injected with 50 mg/kg of paraquat. One hour after paraquat administration, the rats were treated intravenously with Xuebijing(8 mL /kg). At 12 hours after paraquat administration, serum was collected to evaluate kidney function, then the rats were sacrificed and kidney samples were immediately harvested. AKI scores were evaluated by renal histopathology and pro-in? ammatory cytokines mR NA levels in kidney were assayed using real-time RT-PCR.RESULTS: Serum urea nitrogen, creatinine and AKI scores were significantly higher in the paraquat group, compared with the sham group(P<0.05, respectively). Moreover, interleukin(IL)-1β, IL-6 and TNF-α m RNA levels were signi? cantly higher in the paraquat group(P<0.01, respectively). However, intravenous Xuebijing signi? cantly decreased serum urea nitrogen, creatinine, AKI scores and IL-1β, IL-6 and TNF-α m RNA levels, compared with the paraquat group(P<0.05, respectively).CONCLUSION: Intravenous Xuebijing attenuates AKI following paraquat poisoning by suppressing in? ammatory response.
