Mitochondrial calcium uniporter(MCU)is a conserved calcium ion(Ca^(2+))transporter in the mitochondrial inner membrane of eukaryotic *** MCU proteins regulate Ca^(2+)flow and modulate plant cell development remain largely ***,we identified the gene GhMCU4 encoding a MCU protein that negatively regulates plant development and fiber elongation in cotton(Gossypium hirsutum).GhMCU4expressed constitutively in various tissues with the higher transcripts in elongating fiber *** of GhMCU4 in cotton significantly elevated the plant height and root *** calcium signaling pathway was significantly activated and calcium sensor genes,including Ca^(2+)dependent modulator of interactor of constitutively active ROP(GhCMI1),calmodulin like protein(GhCML46),calciumdependent protein kinases(GhCPKs),calcineurin B-like protein(GhCBLs),and CBL-interacting protein kinases(GhCIPKs),were dramatically upregulated in GhMCU4-silenced *** processes were preferentially enriched,and genes related to regulation of transcription were upregulated in GhMCU4-silenced *** contents of Ca^(2+)and H_(2)O_(2)were significantly increased in roots and leaves of GhMCU4-silenced *** length and Ca^(2+)and H_(2)O_(2)contents in fibers were significantly increased in GhMCU4-silenced *** study indicated that GhMCU4 plays a negative role in regulating cell elongation in cotton,thus expanding understanding in the role of MCU proteins in plant growth and development.

Monotropein Induced Apoptosis and Suppressed Cell Cycle Progression in Colorectal Cancer Cells

Objective: To determine whether monotropein has an anticancer effect and explore its potential mechanisms against colorectal cancer(CRC) through network pharmacology and molecular docking combined with experimental verification. Methods: Network pharmacology and molecular docking were used to predict potential targets of monotropein against CRC. Cell counting kit assay, plate monoclonal assay and microscopic observation were used to investigate the antiproliferative effects of monotropein on CRC cells HCT116, HT29 and LoVo. Flow cytometry and scratch assay were used to analyze apoptosis and cell cycle, as well as cell migration, respectively in HCT116, HT29, and LoVo cells. Western blotting was used to detect the expression of proteins related to apoptosis, cell cycle, and cell migration, and the expression of proteins key to the Akt pathway. Results: The Gene Ontology and Reactome enrichment analyses indicated that the anticancer potential of monotropein against CRC might be involved in multiple cancer-related signaling pathways. Among these pathways, RAC-beta serine/threonine-protein kinase(Akt1, Akt2), cyclin-dependent kinase 6(CDK6), matrix metalloproteinase-9(MMP9), epidermal growth factor receptor(EGFR), cell division control protein 42 homolog(CDC42) were shown as the potential anticancer targets of monotropein against CRC. Molecular docking suggested that monotropein may interact with the 6 targets(Akt1, Akt2, CDK6, MMP9, EGFR, CDC42). Subsequently, cell activity of HCT116, HT29 and LoVo cell lines were significantly suppressed by monotropein(PlinD1, CDK4 and CDK6, inhibited cell migration by suppressing the expressions of CDC42 and MMP9(Pling pathway. Atomistic study on the microscopic mechanism of grain boundary embrittlement induced by small dense helium bubbles in iron

The helium bubbles induced by 14 MeV neutron irradiation can cause intergranular fractures in reduced activation ferritic martensitic steel,which is a candidate structural material for fusion *** order to elucidate the susceptibility of different grain boundaries(GBs)to helium-induced embrittlement,the tensile fracture processes of 10 types of GBs with and without helium bubbles in body-centered cubic(bcc)iron at the relevant service temperature of 600 K were investigated via molecular dynamics *** results indicate that in the absence of helium bubbles,the GBs studied here can be classified into two distinct categories:brittle GBs and ductile *** atomic scale analysis shows that the plastic deformation of ductile GB at high temperatures originates from complex plastic deformation mechanisms,including the Bain/Burgers path phase transition and deformation twinning,in which the Bain path phase transition is the most dominant plastic deformation ***,the presence of helium bubbles severely inhibits the plastic deformation channels of the GBs,resulting in a significant decrease in elongation at *** bubble-decorated GBs,the ultimate tensile strength increases with the increase in the misorientation ***,the coherent twin boundary∑3{112}was found to maintain relatively high fracture strength and maximum failure strain under the influence of helium bubbles.

Application of a concrete thermal pile in cooling the warming permafrost under climate change

Permafrost degradation caused by climate warming is posing a serious threat to the stability of cast-in-place pile foundations in warm permafrost *** cold energy can be effectively utilized by two-phase closed thermosyphons(TPCTs)to cool the ***,we installed TPCTs in a cast-in-place pile foundation to create a unique structure called a thermal pile,which effectively utilizes the TPCTs to regulate ground *** we conducted a case study and numerical simulation to exhibit the cooling performance,and optimize the structure of the thermal *** purpose of this study is to promote the application of thermal piles in unstable permafrost *** on the findings,the thermal pile operated for approximately 53%of the entire year and effectively reduced the deep ground temperature at a rate of at least-0.1℃per ***,it successfully raised the permafrost table that is 0.35 m shallower than the natural ground *** characteristics prove highly beneficial in mitigating the adverse effects of permafrost degradation and enhancing infrastructure *** the length of the condenser section and the diameter of the TPCT in a suitable manner can effectively enhance the cooling capability of the thermal pile and ensure the long-term mechanical stability of the pile foundation even under climate warming.

A pair of coumarin-based polycyclic meroterpenoid enantiomers(+)/(-)-gerbeloid A[(+)-1a and(-)-1b]were isolated from the medicinal plant Gerbera piloselloides,which have a unique caged oxatricyclo[4.***.2.0^(3,8)]decene *** planar and three-dimensional structures were exhaustively characterized by comprehensive spectroscopic data and X-ray diffraction *** by the hypothetical biosynthetic pathway,the biomimetic synthesis of racemic 1 was achieved using 4-hydroxy-5-methylcoumarin and citral as the starting material via oxa-6πelectrocyclization and intramolecular[2+2]***,the results of the biological activity assay demonstrated that both(+)-1a and(-)-1b exhibited potent lipid-lowering effects in 3T3-L1 adipocytes and the high-fat diet zebrafish ***,the lipid-lowering activity of(+)-1a is better than that of(-)-1b at the same concentration,and molecular mechanism study has shown that(+)-1a and(-)-1b impairs adipocyte differentiation and stimulate lipolysis by regulating C/EBPα/PPARγsignaling and Perilipin signaling in vitro and in *** findings provide a promising drug model molecule for the treatment of obesity.

Spectral observation of symmetry-protected selection rules for dynamical high-dimensional parity in alignment magnetic resonance

Multidimensional Floquet-driven alignment systems with dynamical symmetry present various exotic phenomena and applications. However, there are challenges in directly characterizing large-spin dynamical symmetry from spectra. Here, we first observe the symmetry-protected selection rules of dynamical high-dimensional parity in a large-spin(F = 4) system. We theoretically construct a Floquet-driven alignment system that can be used to reveal high-dimensional spatiotemporal symmetry. In the experiment, the system is implemented in Cs atomic gas subjected to two-dimensional Floquet-modulated magnetic resonance driving. By developing Floquet detection protocols of alignment double-sided spectra, we directly verify symmetry-protected selection rules of dynamical high-dimensional parity for large-spin systems. This work advances the exploration of dynamical symmetry to large spins, and unravels a universal Floquet scheme for the investigation of symmetry-protected selection rules.

Impact of the Political Risk on Food Reserve Ratio:Evidence Across Countries

Using an unbalanced panel data covering 75 countries from 1991 to 2019, the authors explore how the political risk impacts on food reserve ratio. The empirical findings show that an increasing political risk negatively affects food reserve ratio, and the same effects hold for both internal risk and external risk. Moreover, the authors find that the increasing external or internal risks both negatively affect food production and food exports, but external risk does not significantly impact food imports and it positively impacts food consumption, while internal risk negatively impacts food imports and food consumption. The results suggest that most governments have difficulty raising subsequent food reserve ratios in face of an increasing political risk, no matter if it is an internal risk or an external risk although the mechanisms behind the impacts are different.

Relationship between the expression of Toll-like receptor 2 and 4 in mononuclear cells and postoperative acute lung injury in orthotopic liver transplantation

Background The aim of this study was to investigate the potential relationship between the dynamic expression of Toll-like receptor 2 and 4 （TLR2/4） in peripheral blood mononuclear cells as well as changes in serum concentration of inflammatory factors and acute lung injury （ALl） in patients after orthotopic liver transplantation （OLT）. Methods The peripheral blood samples of 27 patients （23 men and 4 women with ASA Ⅲ to Ⅳ） who received OLT were collected for measurement of TLR2/4 at T1 （after induction of anesthesia）, T2 （25 minutes after anhepatic phase）, T3 （3 hours after graft reperfusion） and T4 （24 hours after graft reperfusion）. The expression of TLR2/4 in mononuclear cells was measured by flow cytometry. The serum concentrations of tumor necrosis factor （TNF）-α, interleukin （IL）-1β and IL-8 were measured by enzyme-linked immunosorbent assay （EliSA）. Twenty-seven patients were assigned to ALl group （n=-9） and non-ALl group （n=-18） according to the diagnostic criteria of ALl. The expression of TLR2/4 in the ALl group or non-ALl group was analyzed. Results Compared to the non-ALl group, the volumes of blood loss, ascites, total output and transfused red blood cells were higher in the ALl group, and the anhepatic phase lasted longer （P 〈0.05, P 〈0.01）. The expression of TLR2/4 in mononuclear cells increased significantly at T3 and T4, and serum concentrations of TNF-α, IL-1β and IL-8 increased significantly too. There was no significant difference in Child-Turcotte-Pugh （CTP） scores between the ALl group and non-ALl group （P 〉0.05）. The expression of TLR2/4 in mononuclear cells increased significantly at T3 and T4 in the ALl group （P 〈0.05, P〈0.01）. A positive correlation was noted between the expression of TLR4 in mononuclear cells and the serum concentrations of TNF-α, IL-1β （P=0.041, P=0.046） in the ALl group. Effects of cross-Kerr coupling on transmission spectrum of double-cavity optomechanical system

We theoretically study the transmission spectrum of the cavity field in a double-cavity optomechanical system with cross-Kerr(CK) effect. The system consists of two tunneling coupling optomechanical cavities with a mechanical resonator as a coupling interface. By doping CK medium into the mechanical resonator, CK couplings between the cavity fields and the mechanical resonator are introduced. We investigate the effects of CK coupling strength on the transmission spectrum of the cavity field, including the transmission rate, nonreciprocity and four-wave mixing(FWM). We find that the transmission spectrum of the probe field can show two obvious transparent windows, which can be widened by increasing the CK coupling strength. For the transmission between the two cavity fields, the perfect nonreciprocity and reciprocity are present and modulated by CK coupling and phase difference between two effective optomechanical couplings. In addition, the effects of the optomechanical and CK couplings on FWM show that the single peak of FWM is split into three symmetrical peaks due to the introduction of the CK effect.
