Based on global initiatives such as the clean energy transition and the development of renewable energy,the pumped storage power station has become a new and significant way of energy storage and regulation,and its construction environment is more complex than that of a traditional *** particular,the stability of the rock strata in the underground reservoirs is affected by the seepage pressure and rock stress,which presents some challenges in achieving engineering safety and *** the advantages of the numerical simulation method in dealing deal with nonlinear problems in engineering stability,in this study,the stability of the underground reservoir of the Shidangshan(SDS)pumped storage power station was numerically calculated and quantitatively analyzed based on fluid-structure coupling theory,providing an important reference for the safe operation and management of the underground ***,using the COMSOL software,a suitablemechanicalmodel was created in accordance with the geological structure and project characteristics of the underground ***,the characteristics of the stress field,displacement field,and seepage field after excavation of the underground reservoir were simulated in light of the seepage effect of groundwater on the nearby rock of the underground ***,based on the construction specifications and Molar-Coulomb criterion,a thorough evaluation of the stability of the underground reservoir was performed through simulation of the filling and discharge conditions and anti-seepage strengthening *** findings demonstrate that the numerical simulation results have a certain level of reliability and are in accordance with the stress measured in the project *** underground reservoir excavation resulted in a maximum displacement value of the rock mass around the caverns of 3.56 mm in a typical section,and the safety coefficient of the parts,as determined using the Molar-Coulomb criterion,was highe

Smoke and fire recognition are of great importance on foreseeing fire disasters and preventing environmental pollution by monitoring the burning process of objects(e.g., straw, fuels). However, since fire images suffer from problems like the variability of the features, complexity of scenarios, interference from background, changeable weather conditions as well as image quality problems, identifying smoke and fire accurately and promptly from a given image still remains a substantial challenge. Automatically learning the features of smoke images by CNNs has improved the target recognition ability compared to traditional approaches,nonetheless, convolutions and pooling operations in CNNs may cause severe information loss which may lead to *** tackle the above problems, this paper proposed a hybrid attention model based on the characteristics of smoke images. This model adopted multiple optimized attention mechanism in several stages to quickly and precisely capture the important features,achieving state-of-the-art performance on smoke and fire recognition in terms of accuracy and speed. Our proposed module mainly consists of two stages: pooling and attention. In the first stage, we conducted several newly proposed first-order pooling methods. Through traversing the data space in a larger scope, features are better reserved, thus constructing a more intact feature space of smoke and fire in an image. In the second stage, feature maps are aggregated together to perform channel and spatial attention. The channel and spatial dependencies allow us to quickly catch the important features presented in an image. By fully exploring the feature space and prominent salient features, characteristics of smoke and fire are better presented so as to obtain better smoke and fire detection results. Molting and metamorphosis are important physiological processes in insects that are tightly controlled by ecdysone receptor(EcR)through the 20-hydroxyecdysone(20E)signaling *** is a steroid nuclear receptor(SR).Several FK506-binding proteins(FKBPs)have been identified from the mammal SR complex,and are thought to be involved in the subcellular trafficking of ***,their roles in insects are poorly *** explore whether FKBPs are involved in insect molting or metamorphosis,we injected an FKBP inhibitor(FK506)into a lepidopteran insect,Spodoptera litura,and found that molting was inhibited in 61.11%of the larvae,and that the time for larvae to pupate was significantly extended.A total of 10 FKBP genes were identified from the genome of *** and were clustered into 2 distinct groups,according to their subcellular localization,with FKBP13 and FKBP14 belonging to the endoplasmic reticulum(ER)group and with the other members belonging to the cytoplasmic(Cy)*** the CyFKBPs were significantly upregulated in the prepupal or pupal stages,with the opposite being observed for the ER group ***506 completely blocked the transfer of EcR to the nucleus under 20E induction,and significantly downregulated the transcriptional expression of many 20E signaling genes.A similar phenomenon was observed after RNA interference of2 CyFKBPs(FKBP45 and FKBP12b),but not for *** together,our data indicate that the cytoplasmic FKBPs,especially FKBP45 and FKBP12b,mediate the nuclear localization of EcR,thereby regulating the 20E signaling and ultimately affecting molting and metamorphosis in insects.

A possible quasi-periodic oscillation(QPO) at frequency 7.045 × 10^(-5) Hz is found in the narrow-line Seyfert 1galaxy Mrk 142 in the data of XMM-Newton collected on 2020 April *** find that the QPO signal is statistically significantly larger than the 5σ level and highly coherent with quality factor Q > 5 at the 0.3–10 keV band by using the method of the Lomb–Scargle Periodogram,the Weighted Wavelet Z-transform and the *** analyze the data in 0.3–0.6 keV,0.6–1 keV,1–3 keV and 3–10 keV energy bands,and find obvious QPO signals at 0.3–0.6 keV and 1–3 keV *** then analyze the time-average spectra and time variability at the QPO frequency of 7.045 × 10^(-5) Hz,and use a model to fit *** find that the QPO signal mainly comes from the X-ray hot corona.

The prevalence of hyperlipidemia has increased significantly due to genetic, dietary, nutritional and pharmacological factors, and has become one of the most common pathological conditions in humans. Hyperlipidemia can lead to a range of diseases such as atherosclerosis, stroke, coronary heart disease, myocardial infarction, diabetes, and kidney failure, etc. High circulating low-density lipoprotein cholesterol (LDL-C) is one of the causes of hyperlipidemia. LDL-C in the blood binds to LDL receptor (LDLR) and regulates cholesterol homeostasis through endocytosis. In contrast, proprotein convertase subtilisin/kexin type 9 (PCSK9) mediates LDLR degradation via the intracellular and extracellular pathways, leading to hyperlipidemia. Targeting PCSK9-synthesizing transcription factors and downstream molecules are important for development of new lipid-lowering drugs. Clinical trials regarding PCSK9 inhibitors have demonstrated a reduction in atherosclerotic cardiovascular disease events. The purpose of this review was to explore the target and mechanism of intracellular and extracellular pathways in degradation of LDLR and related drugs by PCSK9 in order to open up a new pathway for the development of new lipid-lowering drugs.

In this study,we aimed to evaluate the toxic effects,changes in life span,and expression of various metabolismrelated genes in Caenorhabditis elegans,using RNA interference(RNAi)and mutant strains,after 3-bromopyruvate(3-BrPA)*** was treated with various concentrations of 3-BrPA on nematode growth medium(NGM)plates,and their survival was monitored every 24 *** expression of genes related to metabolism was measured by the real-time fluorescent quantitative polymerase chain reaction(qPCR).Nematode survival in the presence of 3-BrPA was also studied after silencing three hexokinase(HK)*** average life span of *** cultured on NGM with 3-BrPA was shortened to 5.7 d compared with 7.7 d in the control ***-1,hxk-2,and hxk-3 were overexpressed after the treatment with *** successfully interfering hxk-1,hxk-2,and hxk-3,the 50%lethal concentration(LC50)of all mutant nematodes decreased with 3-BrPA treatment for 24 h compared with that of the *** the cyp35 genes tested were overexpressed,except *** induction of cyp-35A1 expression was most *** LC50 values of the mutant strains cyp-35A1,cyp-35A2,cyp-35A4,cyp-35B3,and cyp-35C1 were lower than that of the ***,the toxicity of 3-BrPA is closely related to its effect on hexokinase metabolism in nematodes,and the cyp-35 family plays a key role in the metabolism of 3-BrPA.

Non-destructive Testing and Intelligent Evaluation of Road Structural Conditions using GPR and FWD

Aiming at the assessment of road structural conditions， a method of combining ground penetrating radar (GPR) and falling weight deflectometer (FWD) for assessing the pavement structural integrity and strength was proposed in this study. First， 3D GPR was performed to detect the thickness and internal distress of pavement structural layers using the canny edge detection and you only look once version eight (YOLOv8) detection algorithms. Results showed that the error of thickness extraction was approximately 3%， and the distress detection achieved a mean average precision (mAP) of 0.859 and an inference time of 11.43 ms with a GTX 1070 GPU. Then， the extracted thickness was used for the modulus (E) inversion of pavement structure layers based on FWD test data and regression analysis. Finally， the distress ratio inside pavement structures (DRIPS) was proposed as a structural integrity index. The relationship between E and DRIPS was revealed and showed a good correlation. The greater the DRIPS value， the worse the pavement structural integrity and strength. It provides a reference for the evaluation of road structural conditions. This strategy proved to be reliable for nondestructive testing and evaluation of road structures， which could improve the comprehensiveness and effectiveness for evaluating road structural conditions.
