Effects of different cooking methods on health-promoting compounds of broccoli

The effects of five domestic cooking methods,including steaming,microwaving,boiling,stir-frying,and stir-frying followed by boiling(stir-frying/boiling),on the nutrients and health-promoting compounds of broccoli were investigated.The results show that all cooking treatments,except steaming,caused significant losses of chlorophyll and vitamin C and significant decreases of total soluble proteins and soluble sugars.Total aliphatic and indole glucosinolates were significantly modified by all cooking treatments but not by steaming.In general,the steaming led to the lowest loss of total glucosinolates,while stir-frying and stir-frying/boiling presented the highest loss.Stir-frying and stir-frying/boiling,the two most popular methods for most homemade dishes in China,cause great losses of chlorophyll,soluble protein,soluble sugar,vitamin C,and glucosinolates,but the steaming method appears the best in retention of the nutrients in cooking broccoli.

Background：Quantifying syphilis prevalence is important for planning interventions and advocating for resources on syphilis.However,data on large sample studies regarding the prevalence of syphilis among reproductive-age women in rural China were not available for analysis.The aim of the study was to determine the prevalence,epidemiological characteristics,and related factors of syphilis infection among reproductive-age women in rural China.Methods：Data were obtained from a nationwide,population-based,cross-sectional study under the National Free Preconception Health Examination Project which covered all the 31 provinces in China's Mainland.Women intending to get pregnant within the next 6 months were enrolled between January 1,2010,and December 31,2012.Sociodemographic,gynecological and obstetric characteristics,and other relevant information were obtained through face-to-face interviews.Treponema pallidum particle agglutination assay test was used to detect positive samples of syphilis.Univariate and multivariate logistic regressions were performed to assess the associations between syphilis seropositivity and related factors.Results：The overall seroprevalence of syphilis （SPS） among the 2,044,126 women who received syphilis screening test during 2010-2012 was 0.37% （95% confidence interval [CI]：0.36-0.37%）.The SPS appeared 0.24% （95% CI：0.23-0.26%） and 0.66% （95% CI：0.59-0.72%） in women at 21-24 and 40-44 years of age,respectively,showing an increase of SPS,parallel with age,and the difference was significant.SPS was significantly higher in ethnic minorities than that in Han nationality （0.58% vs.0.35%,respectively,odds ratio [OR] =1.41,95% CI：1.30-1.53） and higher in workers than that in farmers （0.45% vs.0.36%,respectively,OR =1.27,95% CI：1.14-1.41）.Women with primary school or below level had a higher SPS as compared to those with college or above educational level （0.61% vs.0.32%,respectively,OR =2.49,95% CI：2.14-2.89）,and the increase reversely correlated with the levels of education.Women whose spouses w

The carotenoid isomerase gene(BoaCRTISO)of Chinese kale was targeted and edited using the CRISPR/Cas9 system in the present study.The results showed a high mutation rate(81.25%),and 13 crtiso mutants were obtained.Only two types of mutations,insertions and replacements,were found.Both the total and individual carotenoid and chlorophyll concentrations of the biallelic and homozygous mutants were reduced,and the total levels declined by 11.89–36.33%.The color of the biallelic and homozygous mutants changed from green to yellow,likely reflecting a reduction in the color-masking effect of chlorophyll on carotenoids.The expression levels of most carotenoid and chlorophyll biosynthesis-related genes,including CRTISO,were notably lower in the mutants than in the WT plants.In addition,the functional differences between members of this gene family were discussed.In summary,these findings indicate that CRISPR/Cas9 is a promising technique for the quality improvement of Chinese kale and other Brassica vegetables.

Genetic variation in alkaloid accumulation in leaves of Nicotiana

Alkaloids are plant secondary metabolites that are widely distributed in Nicotiana species and contribute greatly to the quality of tobacco leaves. Some alkaloids, such as nornicotine and myosmine, have adverse effects on human health. To reduce the content of harmful alkaloids in tobacco leaves through conventional breeding, a genetic study of the alkaloid variation among different genotypes is required. In this study, alkaloid profiles in leaves of five Nicotiana tabacum cultivars and Nicotiana tomentosiformis were investigated. Six alkaloids were identified from all six genotypes via gas chromatograph-mass spectrometry(GC-MS). Significant differences in alkaloid content were observed both among different leaf positions and among cultivars. The contents of nornicotine and myosmine were positively and significantly correlated(R2=0.881), and were also separated from those of other alkaloids by clustering. Thus, the genotype plays a major role in alkaloid accumulation, indicating a high potential for manipulation of alkaloid content through traditional breeding.

目的:探究外源施加葡萄糖和赤霉酸对芥蓝芽菜中芥子油苷的积累,花青素、多酚和维生素C等抗氧化物的含量,以及总抗氧化活性的影响。创新点:首次发现葡萄糖和赤霉酸可以协同促进芥蓝芽菜中几乎所有种类芥子油苷以及总芥子油苷的积累,并且可以大幅度提升总多酚的含量以及抗氧化能力。方法:以芥蓝芽菜为材料,使用3%(0.03 g/ml)葡萄糖和5μmo/L赤霉酸进行外源处理,以水处理作为对照组。用高效液相色谱法分析芥子油苷和维生素C的含量;采用分光光度法检测花青素的含量;用Folin-Ciocalteu试剂法测定总多酚的含量;利用亚铁还原能力实验(FRAP)法进行总抗氧化能力的评估。结论:葡萄糖和赤霉酸共同处理可以有效提升芥蓝芽菜中有益健康的植物化学物质含量以及抗氧化能力。

Drying soil has been conducive to a high frequency of extreme high-temperature events over many regions worldwide in recent decades.However,changes in the intraseasonal variability of soil moisture can also influence the likelihood of extremely high temperatures.Although previous investigators have examined the association between extremely high temperatures and large-scale atmospheric circulation variability,the role of land-atmosphere coupling dominated by soil moisture variability in extremely high temperatures,particularly over the Eurasian continent,is not well understood.In this study,on the basis of the Land Surface,Snow,and Soil Moisture Model Intercomparison Project,we found that land-atmosphere feedback amplified the variability of soil moisture in most regions of Eurasia during summer from 1980 to 2014.This amplification of soil moisture variability is closely correlated with more intensive intraseasonal variability of surface air temperature and more frequent occurrences of extreme high-temperature events,particularly in Europe,Siberia,Northeast Asia,and the Indochina Peninsula.This correlation implies that increasing the intraseasonal variability of soil moisture results in a high likelihood of heat extremes during summer in most parts of Eurasia except Asian desert areas.On the intraseasonal timescale,the land-atmosphere coupling increases the variability of surface sensible heat flux and net long-wave radiation heating the atmosphere by intensifying the soil moisture variability,thus amplifying the variability of surface air temperature and enhancing the extreme high-temperature days.This finding demonstrates the importance of changes in intraseasonal soil moisture variability for the increasing likelihood of heat extremes in summer.

In this paper, we propose an efficient and accurate method for pricing Guaranteed Minimum Death Benefit (GMDB) under time-changed Le′vy processes. Suppose that the GMDB payoff depends on a dollar cost averaging (DCA) style periodic investment, and the activity rate process in stochastic time change is modeled by a square-root process. We develop a recursive method to derive the closed form valuation formula by using the frame duality projection method. Numerical examples are reported for demonstrating the effectiveness of our approach and illustrating the interplay between contract parameters and the valuation.

BACKGROUND Gastric cancer(GC)is a highly aggressive malignancy with a heterogeneous nature,which makes prognosis prediction and treatment determination difficult.Inflammation is now recognized as one of the hallmarks of cancer and plays an important role in the aetiology and continued growth of tumours.Inflammation also affects the prognosis of GC patients.Recent reports suggest that a number of inflammatory-related biomarkers are useful for predicting tumour prognosis.However,the importance of inflammatory-related biomarkers in predicting the prognosis of GC patients is still unclear.AIM To investigate inflammatory-related biomarkers in predicting the prognosis of GC patients.was constructed using the least absolute shrinkage and selection operator Cox regression model based on the GEO database.GC patients from the GSE26253 cohort were used for validation.Univariate and multivariate Cox analyses were used to determine the independent prognostic factors,and a prognostic nomogram was established.The calibration curve and the area under the curve based on receiver operating characteristic analysis were utilized to evaluate the predictive value of the nomogram.The decision curve analysis results were plotted to quantify and assess the clinical value of the nomogram.Gene set enrichment analysis was performed to explore the potential regulatory pathways involved.The relationship between tumour immune infiltration status and risk score was analysed via Tumour Immune Estimation Resource and CIBERSORT.Finally,we analysed the association between risk score and patient sensitivity to commonly used chemotherapy and targeted therapy agents.RESULTS A prognostic model consisting of three inflammatory-related genes(MRPS17,GUF1,and PDK4)was constructed.Independent prognostic analysis revealed that the risk score was a separate prognostic factor in GC patients.According to the risk score,GC patients were stratified into high-and low-risk groups,and patients in the high-risk group had significantly worse prognoses according to age
