How to make insulin-producing pancreatic β cells for diabetes treatment

Around 400 million people worldwide suffer from diabetes *** major pathological event for Type 1 diabetes and advanced Type 2 diabetes is loss or impairment of insulin-secreting β cells of the *** the past 100 years,daily insulin injection has served as a life-saving treatment for these ***,insulin injection often cannot achieve full glucose control,and over time poor glucose control leads to severe complications and *** an alternative treatment,islet transplantation has been demonstrated to effectively maintain glucose homeostasis in diabetic patients,but its wide application is limited by the scarcity of donated ***,it is important to define new strategies to obtain functional human β cells for transplantation ***,we summarize recent progress towards the production of β cells in vitro from pluripotent stem cells or somatic cell types including a cells,pancreatic exocrine cells,gastrointestinal stem cells,fibroblasts and *** also discuss novel methods for optimizing β cell transplantation and maintenance in *** our perspective,the future of βcell replacement therapy is very promising although it is still challenging to control differentiation of β cells in vitro and to protect these cells from autoimmune attack in Type 1 diabetic ***,tremendous progress has been made in understanding βcell differentiation and producing functional β cells with different *** the coming years,we believe more clinical trials will be launched to move these technologies towards treatments to benefit diabetic patients.

Solvent Effect on the Photoinduced Electron Transfer Reaction Between Thioxanthen-9-one and Diphenylamine

Photoinduced chemical reaction between thioxanthen-9-one (TX) and diphenylamine (DPA) were investigated by the nanosecond laser flash photolysis. With photolysis at 355 nm, the triplet TX (3TX*) is produced via a Franck-Condon excitation and intersystem crossing. In the transient absorption spectra of the reduction of 3TX* by DPA in pure and aqueous CH3CN, four bands are clearly observed and assigned to absorption of 3TX*, TXH·, TX·? and DPA·+, respectively. With the increase of solvent polarity, the blue-shift was observed for all absorption bands of the intermediates. With the aid of dynamic decay curves, an electron transfer followed by a protonation process is confirmed for the reduction of 3TX* by DPA. The quenching rate constants of 3TX* by DPA very slightly decrease from 9.7×109 L/(mol·s) in pure CH3CN, to 8.7×109 L/(mol·s) in CH3CN:H2O (9:1), 8.0×109 L/(mol·s) in CH3CN:H2O (4:1) and 7.5×109 L/(mol·s) in CH3CN:H2O (1:1). Therefore water plays a minor role in the title reaction, and moreover no obvious medium effect of solvent polarity is observed for the electron transfer between 3TX* and DPA, indicating that the 3nπ* and 3ππ* states of TX have the approximate ability to attract an electron from DPA.

Laser Flash Photolysis on Electron Transfer Reactions between 1,8-Dihydroxyanthraquinone with Adenine and Cytosine

Electron transfer （ET） reactions between 1,8-dihydroxyanthraquinone （DHAQ） and two DNA bases, adenine （A） and cytosine （C）, have been investigated in CH3CN/H20 solution with nanosecond time-resolved laser flash photolysis. After irradiation at 355 nm, the triplet DHAQ is produced via intersystem crossing and reacts with two nucleobases. ET processes for both reactions have been definitely identified, in which two bases play a significant role of electron donor. Based on the measured decay dynamics of various intermediates and the corresponding quenching rates, an initial ET process followed by a secondary proton-transfer reaction is suggested for both the overall reactions. By plotting the observed quenching rate against the concentration of two DNA bases, the bimolecular quenching rate constants are determined as 9.0-10s L/（mol.s） for the 3DHAQ＊＋C reaction and 3.3x10^8 L/（mol.s） for the 3DHAQ＊＋A reaction, respectively.

Roxadustat for treatment of anemia in a cancer patient with endstage renal disease:A case report

BACKGROUNDMost cancer patients are accompanied by anemia, which will be more seriouswhen combined with end-stage renal disease (ESRD). At present, cancer-relatedanemia and renal anemia treatments mainly include erythropoiesis-stimulatingagents (ESAs), iron supplementation, and blood transfusion, but their effects areoften poor with several safety concerns. We have used roxadustat to treat anemiain a cancer patient with ESRD and achieved a successful outcome for the *** SUMMARYA 64-year-old man was diagnosed with right renal cancer (clear cell renal cellcarcinoma). He did not receive surgery or radiotherapy before admission. He wastreated with oral soltan (sunitinib malate) on April 18, 2017. During oral chemotherapy,he had numerous complications, including anemia, hypertension,thyroid hypofunction, skin pigment loss, and renal function deterioration. At last,he progressed to ESRD and began hemodialysis treatment. We initially treated thepatient with high-dose ESAs, iron supplementation, adequate dialysis, and evenblood transfusion, but his anemia did not improve. Roxadustat is a newlydeveloped drug for renal anemia treatment, but not for cancer-related anemia, letalone to treat anemia in cancer patients with ESRD. We prescribed oral roxadustatto the patient. After a period, his hemoglobin gradually increased. He did nothave obvious discomfort symptoms, and his tumor did not progress *** roxadustat could achieve good results in treating anemia in cancer patients with ESRD.

Research on the comprehensive performance of PrFeB magnets for synchrotron radiation and free electron laser

PrFeB magnets, which possess excellent magnetic properties at low temperatures, have important application value as cryogenic permanent magnet undulators for synchrotron radiation sources and free electron lasers. In this research study, several high-performance PrFeB magnets(P42H, P48SH, and P48UH) were prepared, and their performance was comprehensively examined, including evaluations of their magnetic properties,microstructures, uniformity, and stability. Next,their application prospects were analyzed and discussed. In China, the first cryogenic permanent magnet undulators(CPMUs) with P48SH magnets with 18 mm cycle lengths have been developed. When the temperature is 80 K and the gap is 6.0 mm,the magnetic field measurement results of the CPMU showed that the effective magnetic field peak was approximately 0.92 T, yielding an increase of 11.76% relative to operation at 300 K, with an RMS phase error of about 4.99°.

The proof of our main results relies on a well-known fixed point theorem in cones. The paper extends some previous results and reports some new results about impulsive differential equations.

A cross-layer design approach on spectrum allocation and resource scheduling in cognitive PMP networks

We propose the spectrum allocation and resource scheduling algorithms in cognitive point to multipoint (PMP) networks with rapid changes of spectrum opportunities and present a media access control (MAC) protocol based on these algorithms. The objective of spectrum allocation is to make efficient use of the spectrum while maintaining the transceiver synchronization on frequency and time in the network. The objective of resource scheduling is to guarantee the quality of service (QoS) requirements of different kinds of connections and to minimize the total energy consumption in the network as well. By sensing only a small set of possible channels in each slot based on the state transition probability of each channel, our spectrum allocation algorithm achieves high spectrum efficiency in the network. The resource scheduling problem is divided into three sub problems and we derive optimal solutions to these problems by greedy algorithm and convex optimization. The simulation results show that our algorithm can make efficient use of the spectrum and the network resources at a cost of low computational complexity.

Effects of chemotherapeutic agent bendamustine for nonhodgkin lymphoma on spermatogenesis in mice

Non-Hodgkin lymphoma(NHL) is one of the most common cancers affecting men of reproductive age. The high response rate of bendamustine as first-line treatment for NHL, coupled with young age of patients, makes elucidation of the impact of treatment on male reproduction important. Our aim was to determine the effects of bendamustine on male reproduction by animal model. Male mice were treated with bendamustine(40 mg/kg) through tail vein injection while cisplatin was given as a standard(3 mg/kg) through intraperitoneal injection. After 3 weeks, bendamustine induced weight loss and sperm morphology abnormalities were compared to the control. Additionally, sperm with folded tails were the most frequent abnormality in bendamustine-treated mice. But the mechanism of sperm abnormality induced by bendamustine remains to be elucidated. These results indicate bendamustine may affect spermatozoa of patients who have been treated for NHL.
