In vivo imaging reveals a synchronized correlation among neurotransmitter dynamics during propofol and sevoflurane anesthesia

General anesthesia is widely applied in clinical ***,the precise mechanism of loss of consciousness induced by general anesthetics remains ***,we measured the dynamics of five neurotransmitters,includingγ-aminobutyric acid,glutamate,norepinephrine,acetylcholine,and dopamine,in the medial prefrontal cortex and primary visual cortex of C57Bl/6 mice through in vivo fiber photometry and genetically encoded neurotransmitter sensors under anesthesia to reveal the mechanism of general anesthesia from a neurotransmitter *** revealed that the concentrations of γ-aminobutyric acid,glutamate,norepinephrine,and acetylcholine increased in the cortex during propofol-induced loss of *** levels did not change fol.owing the hypnotic dose of propofol but increased significantly fol.owing surgical doses of propofol ***,the concentrations of the five neurotransmitters generally decreased during sevoflurane-induced loss of ***,the neurotransmitter dynamic networks were not synchronized in the non-anesthesia groups but were highly synchronized in the anesthetic *** findings suggest that neurotransmitter dynamic network synchronization may cause anesthetic-induced loss of consciousness.

Seed coat color affects the appearance and commodity quality of mung beans(Vigna radiata l.).The substances that affect mung bean seed coat color are mainly flavonoids,which have important medicinal *** the seed coat color gene in mung beans would facilitate the development of new varieties and improve their *** this study,an F2 mapping population consisting of 546 plants was constructed using Jilv9(black seed coat)and BIS9805(green seed coat).Using bulk segregated analysis(BSA)sequencing and kompetitive allele-specific PCR(KASP)markers,the candidate region related to seed coat color was finally narrowed to 0.66 Mb on chromosome(Chr.)4 and included eight candidate *** transcriptome and metabolome analyses showed that three of the eight candidate genes(loC106758748,loC106758747,and loC106759075)were differentially expressed,which may have caused the differences in flavonoid metabolite content between Jilv9 and *** findings can provide a research basis for cloning the genes related to seed coat color and accelerate molecular markerassisted selection breeding in mung beans.

BACKGRoUND Rectus abdominis separation(DRA)affects pelvic stability and body *** studies have explored the effects of manual massage on early postpartum DRA and postpartum *** To analyze the curative effect of massage on early postpartum DRA and its impact on postpartum depression and thus its ability promote the overall psychosomatic rehabilitation of postpartum *** Data were retrospectively collected on 70 primiparous women with postpartum DRA who underwent rehabilitation at the Postpartum Rehabilitation Center of Huzhou Maternal and Child Health Hospital from october 2022 to September *** patients were divided into the Group S(35 cases,biomimetic electrical stimulation treatment)and Group l(35 cases,biomimetic electrical stimulation combined with manual massage treatment).Baseline data,the edinburgh postpartum depression scale(EPDS)score,and the visual analog scale(VAS)scores for rectus abdominis distance,waist circumference,and lower back pain before and after treatment were *** No significant differences were found in the baseline data,rectus abdominis distance,waist circumference,and VAS and EPDS scores between the two groups before treatment(P>0.05).After treatment,the distance between rectus abdominis and waist circumference in Group l were significantly smaller than those in Group S(Pore,lower back pain(VAS score)and the EPDS score in Group l were significantly lower than those in Group S(PoNClUSIoN Manual massage can significantly reduce early postpartum DRA,waist circumference,and back pain and improve the patient’s mental state and postpartum depression.

Enhanced activation of peroxymonosulfate on layered molybdenum disulfide loaded with Co–la bimetallic oxide for efficient degradation of chloramphenicol

The use of transition metal-activated peroxymonosulfate（PMS） as an advanced oxidation technology has gained recognition. This study developed a catalyst using cobaltDlanthanum bimetallic oxide supported on layered molybdenum disulfide （Mo S2） as a carrier. The CoDla/Mo S2catalyst was synthesised through coprecipitation, fol.owed by calcination with an optimised metal ratio of Co:la=2:1 to activate PMS and degrade trace chloramphenicol（CAP） in water. The chemical composition of the catalyst was confirmed using X-ray diffraction （XRD） and X-ray photoelectron spectroscopy （XPS）. At catalyst and PMS dosages of 0.1 and 0.5 g·l-1, respectively, the degradation rate of CAP was 95% with in 30 min. The catalyst exhibitedstrong resistance to most interfering anions and maintained a high degradation rate at p H 3D11. liquid chromatographyDmass spectrometry analysis revealed the potential degradation pathways of CAP in the CoDla/Mo S2（2:1）/PMS system. For other pollutants, such as oxytetracycline,complete degradation was achieved within 20 min,demonstrating the broad applicability of the CoDla/Mo S2（2:1）/PMS system for the degradation and removal.of antibiotic organic contaminants. This study offers a feasible approach for the degradation of antibiotic organic pollutants, including CAP, in natural water bodies.

Molecular mechanism of exopolysaccharide biosynthesis regulated by wze and wzd genes on the immunomodulatory effects of probiotic lacticaseibacillus paracasei S-NB

The surface macromolecules of probiotic bacteria play crucial roles in modulating immune responses in the host. Exopolysaccharide (EPS) from lactic acid bacteria (lAB) have been widely reported to exhibit immunomodulatory activity. In this study， the EPS biosynthesis gene cluster of lacticaseibacillus paracasei S-NB was analyzed and a deletion mutant S-NB△7576 (two genes S-NB＿2175/wze and S-NB＿2176/wzd were responsible for the chain length determination and export of EPS) was successfully constructed， resulting a 40.02% decrease in the production of EPS. The deletion of wze and wzd had little effect on the monosaccharide composition and major groups of the two EPS fractions (BEPS1 and BEPS2). Both BEPS1 and BEPS2 could inhibit the transcriptional level.of tumor necrosis factor-α (TNF-α)， interleukin-1β (Il-1β)， inducible nitric oxide synthase (iNoS) and cycl.oxygenase-2 (CoX-2) mRNA in RAW 264.7 cells induced by lipopolysaccharide (lPS)， and enhance host immune tolerance via suppressing NF-κB and MAPK signaling. Notably， the S-NB△7576 mutant supplied with the BEPS1/BEPS2 exhibited more significant inhibition of cytokines production and the phosphorylation of p65 and c-Jun N-terminal kinase (JNK) in lPS-stimulated cells compared with the S-NB△7576 mutant alone. our study provided the immunomodulatory effect of BEPS1 and BEPS2 from l. paracasei S-NB， in which the wze and wzd genes associated with EPS biosynthesis may play an important role.

Electrocatalytic synthesis of urea through C-N bond formation,converting carbon dioxide(Co_(2))and ni-trate(No_(3)^(-)),presents a promising,less energy-intensive alternative to industrial urea production *** this communication,we report the application of Mo_(2)C nanosheets-decorated carbon sheets(Mo_(2)C/C)as a highly efficient electrocatalyst for facilitating C-N coupling in ambient urea *** Co_(2)-saturated 0.2 mol/l Na_(2)So_(4)solution containing 0.05 mol/l No_(3)^(-),the Mo_(2)C/C catalyst achieves an impres-sive urea yield of 579.13μg h^(-1)mg^(-1)with high Faradaic efficiency of 44.80%at-0.5 V versus the reversible hydrogen *** theoretical calculations reveal that the multiple Mo active sites enhance the formation of^(∗)Co and^(∗)NH_(2)intermediates and facilitate their C-N *** research propels the use of Mo_(2)C-based electrodes in electrocatalysis and accentuates the capabilities of binary metal-based catalysts in C-N coupling reactions.

Although green light(Gl)is located in the middle of the visible light spectrum and regulates a series of plant developmental processes,the mechanism by which it regulates seedling development is largely *** this study,we demonstrated that Gl promotes atypical photomorphogenesis in Arabidopsis thaliana via the dual regulations of phytochrome B(phyB)and *** the Pr-to-Pfr conversion rates of phyB and phyA under Gl were lower than those under red light(Rl)in a fluence rate-dependent and time-dependent manner,long-term treatment with Gl induced high Pfr/Pr ratios of phyB and ***,Gl induced the formation of numerous small phyB photobodies in the nucleus,resulting in atypical photomorphogenesis,with smaller cotyledon opening angles and longer hypocotyls in seedlings compared to *** abundance of phyA significantly decreased after short-and long-term Gl *** determined that four major PHYToCHRoME-INTERACTING FACToRs(PIFs:PIF1,PIF3,PIF4,and PIF5)act downstream of phyB in Gl-mediated cotyledon *** addition,Gl plays opposite roles in regulating different *** example,under continuous Gl,the protein levels of all PIFs decreased,whereas the transcript levels of PIF4 and PIF5 strongly increased compared with dark *** together,our work provides a detailed molecular framework for understanding the role of the antagonistic regulations of phyB and phyA in Gl-mediated atypical photomorphogenesis.

miR-125b reverses cisplatin resistance by regulating autophagy via targeting RoRA/BNIP3l axis in lung adenocarcinoma

The platinum-based chemotherapy is one of the most frequently used treatment protocols for lung adenocarcinoma(lUAD),and chemoresistance,however,usually results in treatment failure and limits its application in the *** has been shown that microRNAs(miRNAs)play a significant role in tumor *** this study,miR-125b was identified as a specific cisplatin(DDP)-resistant gene in lUAD,as indicated by the bioinformatics analysis and the real-time quantitative PCR *** decreased serum level.of miR-125b in lUAD patients was correlated with the poor treatment response rate and short survival ***-125b decreased the A549/DDP proliferation,and the multiple drug resistance-and autophagy-related protein expression levels,which were all reversed by the inhibition of *** addition,xenografts of human tumors in nude mice were suppressed by miR-125b,demonstrating that through autophagy regulation,miR-125b could reverse the DDP resistance in lUAD cells,both in vitro and in *** mechanistic studies indicated that miR-125b directly repressed the expression levels of RoRA and its downstream BNIP3l,which in turn inhibited autophagy and reversed *** on these findings,miR-125b in combination with DDP might be an effective treatment option to overcome DDP resistance in lUAD.
