Elucidation and engineering mitochondrial respiratory-related genes for improving bioethanol production at high temperature in Saccharomyces cerevisiae

Industrial manufacturing of bioproducts,especially bioethanol,can benefit from high-temperature fermentation,which requires the use of thermotolerant yeast *** activity in yeast is closely related to its over-all ***,the mitochondrial respiratory changes in response to adaptive thermotolerance are still poorly understood and have been rarely utilized for developing thermotolerant yeast cell ***,adap-tive evolution and transcriptional sequencing,as well as whole-genome-level gene knockout,were used to obtain a thermotolerant strain of Saccharomyces ***,thermotolerance and bioethanol production efficiency of the engineered strain were *** evaluation showed the boosted fermentation ca-pacity and suppressed mitochondrial respiratory activity in the thermotolerant *** improved fermentation produced an increased supply of adenosine triphosphate required for more active energy-consuming *** analysis revealed significant changes in the expression of the genes involved in the mitochondrial respiratory *** of mitochondria-associated gene knockout confirmed that ADK1,DOC1,or MET7 were the key factors for the adaptive evolution of thermotolerance in the engineered yeast ***,overexpression of DOC1 with TEF1 promoter regulation led to a 10.1%increase in ethanol production at 42℃.The relationships between thermotolerance,mitochondrial activity,and respiration were explored,and a ther-motolerant yeast strain was developed by altering the expression of mitochondrial respiration-related *** study provides a better understanding on the physiological mechanism of adaptive evolution of thermotolerance in yeast.

1 Introduction Completely decomposed granite(CDg)is widely distributed in South China(Xu et al.,2022;wa.g et al.,2023).The parent granite rock mass gradually loses features during the weathering process,and thus needs to be reinforced when used(Lan et al.,2003;Dassekpo et al.,2017;Alamanis et al.,2021).On the other hand,considering the huge demand for construction materials and limited natural resources,it is increasingly important to fully utilize solid waste(gruhler et al.,2019;Anagnostopoulos et al.,2020;Jiang et al.,2022,2023a,2023b).With high strength and environment-friendly characteristics,incineration bottom ash(IBA)seems to be a suitable reinforcement material for CDg(Ahmed and Khalid,2011;Alhassan and Tankó,2012;Toraldo et al.,2013;Lynn et al.,2017;Xuan et al.,2018;Tang et al.,2020).

Autophagy is an important factor in reducing the efficacy of tumor phototherapy(including PTT and PDT).Accurate regulation of autophagy in tumor cells is a new strategy to improve the anti-tumor efficiency of PTT/*** project intended to construct a tumor-activated autophagy regulator to efficiently block PTT/PDT-induced autophagy and realize synergistic sensitization to tumor *** achieve this goal,we first synthesized TRANSFERRIN(Tf)biomimetic mineralized nano-tellurium(Tf-Te)as photosensitizer and then used disulfide bond reconstruction technology to induce Tf-Te *** autophagy inhibitor hydroxychloroquine(HCQ)and iron ions carried by Tf were simultaneously loaded to prepare a tumor-responsive drug reservoir Tf-Te/*** entering breast cancer cells through the“self-guidance system”,Tf-Te/HCQ can generate hyperpyrexia and ROS under NIR laser irradiation,to efficiently induce PTT/PDT ***,the disulfide bond broke down in response to gSH,and the nanoparticles disintegrated to release Fe2+and HCQ at fixed *** simultaneously induce lysosomal alkalinization and increased osmotic pressure,effectively inhibit autophagy,and synergistically enhance the therapeutic effect of *** vivo anti-tumor results have proved that the tumor inhibition rate of Tf-Te/HCQ can be as high as 88.6%on 4T1 tumor-bearing *** multifunctional drug delivery system might provide a new alternative for more precise and effective tumor phototherapy.

Optical temperature sensing based on the fluorescence intensity ratio(FIR)of red emission for lanthanide ions holds significant relevance in non-contact temperature measurement for biological *** this study,the perovskite-structured KZnF_(3)is utilized as a host material for Er^(3+)to achieve a high-purity upconversion(UC)red *** observed Stark splitting of the red emission peak provides evidence of the energy level splitting of Er^(3+).group theory is employed to decompose the spectral branching of Er^(3+)under the point group symmetry of KZnF_(3),allowing for the derivation of Stark splitting energy levels induced by the crystal field *** optical temperature-sensing behavior of the red UC luminescence was investigated,specifically examining the FIR of the splitting sub-peaks,which exhibited an exponential relationship with *** KZnF_(3):Yb^(3+),Er^(3+)demonstrated a relative sensitivity(S_(r))of 0.00182%·K^(-1)at 298 K,highlighting its excellent response to *** vivo bio-thermometry experiments conducted on chicken breast validated the material's ability to penetrate biological tissues and showed its significant sensitivity of the FIR to *** results establish KZnF_(3):Yb^(3+),Er^(3+)as a promising material for optical thermometry in various biological applications.

Objective: To evaluate the efficacy of guanxin Danshen Dripping Pill(gXDSDP) in treating anxiety and depression in patients with coronary heart disease(CHD). Methods: A total of 1,428 patients diagnosed with CHD screened for anxiety, depression, and quality of life(QOL) at baseline received 0.4 g of gXDSDP treatment 3 times per day and returned for monthly reassessment. Patients were recruited after stable treatment for CHD and received assessment of general Anxiety Disorder-7(gAD-7), Patient Health Questionnaire-9(PHQ-9), and Seattle Angina Questionnaire(SAQ) for evaluating anxiety, depression, and *** were followed up 3 times, once every 4 weeks, during outpatient visits for 12 weeks. Results: At the third follow-up(F3), the anxiety symptom of 63.79%(673/1,055) of the patients improved to sub-clinical level, and the gAD-7 score improved significantly(8.11 vs. 3.87, Plinical level, with a significant improvement in PHQ-9 score(8.69 vs. 4.41, Pgnificantly improved at the end of treatment compared to those at baseline(all Pginal stability(2.74 vs. 4.14), anginal frequency(8.16vs. 9.10), treatment satisfaction(13.43 vs. 16.29), and disease perception(8.69 vs. 11.02). Conclusions: A fixed dosage of gXDSDP may be a potential treatment option for CHD patients comorbid with anxiety or depression.(Registration No. ChiCTR2100051523)

In recent years,endoscopic resection,particularly endoscopic submucosal dis-section,has become increasingly popular in treating non-metastatic superficial esophageal squamous cell carcinoma(ESCC).In this evolving paradigm,it is crucial to identify factors that predict higher rates of lymphatic invasion and poorer *** tumor size,deeper invasion,poorer differentiation,more infiltrative growth patterns(INF-c),higher-grade tumor budding,positive lymphovascular invasion,and certain biomarkers have been associated with lymph node metastasis and increased morbidity through retrospective reviews,leading to the construction of comprehensive nomograms for outcome *** validated by future prospective studies,these nomograms would prove highly applicable in guiding the selection of treatment for superficial ESCC.

global aging has brought attention to addressing the elderly mobility dilemma,and a commitment to address inclusive transport in *** inclusion for older adults aims at establishing an age-friendly transport *** a direct policy goal and promoting active aging as an indirect social goal,to help older populations gain access to transport and social *** on the analysis of scientific knowledge mapping over the past 30 years,this study visualizes the hotspots and trends in elderly mobility *** a qualitative analysis of 138 papers,this paper reviews the theoretical foundations for transport inclusion and three themes of elderly mobility research,proposes a framework for public transport(PT)inclusion research as well as future research *** study suggests that transport inclusion can help enhance elderly health,social participation,and subjective *** can improve their travel opportunities for active aging in four ways:opportunity accessibility,physical accessibility,age-friendly informationalization,and cost *** research should pay attention to examining the mechanism of perceived transport inclusion on active aging,expanding the multilevel research space of the capability approach(CA),and exploring the subjectiveobjective integrated research model for PT *** research contributes to the overall knowledge of the theoretical and empirical researches on PT inclusion for the elderly and promotes the integrated application of interdisciplinary knowledge in the field.

With continuous advancements in interventional radiology,considerable progress has been made in transarterial therapies for hepatocellular carcinoma(HCC)in recent years,and an increasing number of research papers on transarterial therapies for HCC have been *** this editorial,we comment on the article by Ma et al published in the recent issue of the World Journal of gastrointestinal Oncology:“Efficacy and predictive factors of transarterial chemoembolization combined with lenvatinib plus programmed cell death protein-1 inhibition for unresectable HCC”.We focus specifically on the current research status and future directions of transarterial *** the future,more studies are needed to determine the optimal transarterial local treatment for *** the emergence of checkpoint immunotherapy modalities,it is expected that the results of trials of transarterial local therapy combined with systemic therapy will bring new hope to HCC patients.
