Emerging Concepts of Pathogenesis and Comprehensive Therapeutic Strategies for Spinocerebellar Ataxia Type 3

Spinocerebellar ataxia type 3 (SCA3), also known as Machado-Joseph Disease (MJD), is an autosomal dominant neurodegenerative disorder that predominantly involves the cerebellar, pyramidal, extrapyramidal, motor neuron and oculomotor systems. SCA3 presents strong phenotypic heterogeneity and its causative mutation of SCA3 consists of an expansion of a CAG tract in exon 10 of the ATXN3 gene, situated at 14q32.1. The ATXN3 gene is ubiquitously expressed in neuronal and non-neuronal tissues, and also participates in cellular protein quality control pathways. Mutated ATXN3 alleles present about 45 to 87CAG repeats, which result in an expanded polyglutamine tract in ataxin-3. After mutation, the polyQ tract reaches the pathological threshold (about 50 glutamine residues);the protein is considered that it might gain a neurotoxic function through some unclear mechanisms. We reviewed the literature on the pathogenesis and therapeutic strategies of spinocerebellar ataxia type 3 patients. Conversion of the expanded protein is possible by enhancing protein refolding and degradation or preventing proteolytic cleavage and prevents the protein to reach the site of toxicity by altering its ability to translocate between the nucleus and cytoplasm. Proteasomal degradation and enhancing autophagic aggregate clearance are currently proposed remarkable therapy. In spite of extensive research, the molecular mechanisms of cellular toxicity resulting from mutant ataxin-3 remain no preventive treatment is currently available. These therapeutic strategies might be able to improve sign symptoms of SCA3 as well as slow the disease progression.

Antioxidative effect of folate-modified chitosan nanoparticles

Objective:To evaluate the potency of carboxymethyl chitosan-2,2' ethylenedioxy bisethylamine-folate(CMC-EDBE-FA) on tissue injury,antioxidant status and glutathione system in tissue mitochondria and serum against nicotine-induced oxidative stress in ***: CMC-EDBE-FA was prepared on basis of carboxymethyl chitosan tagged with folic acid by covalently linkage through 2,2' ethylenedioxy *** were divided into four groups,i.e.,control,nicotine(1 mg/kg bw/day),CMC-EDBE-FA(1 mg/kg bw/day) and nicotine(1 mg/kg bw/day) and CMC-EDBE-FA(1 mg/kg bw/day) for 7 *** of lipid peroxidation, oxidized glutathione level,antioxidant enzyme status and DNA damage were observed and ***:The significantly increase of lipid peroxidation,oxidized glutathione levels and DNA damage was observed in nicotine treated group as compared with control group;those were significantly reduced in CMC-EDBE-FA supplemented ***,significantly reduced antioxidant status in nicotine treated group was effectively ameliorated by the supplementation of *** CMC-EDBE-FA treated groups showed no significant change as compared with control group;rather than it repairs the tissue damage of nicotine treated ***:These findings suggest that CMC-EDBE-FA is non-toxic and ameliorates nicotine-induced toxicity.

Objective:To evaluate the wound healing activity of the methanolic root extract of Buchanania lanzan Spreng.(***),with a focus on antimicrobial and anti-biofilm ***:The extract was evaluated for its wound healing properties(excision and incision models)as evident from the analysis of tensile strength and wound *** extract was also screened for antibacterial properties against different Cram positive and Gram negative bacterial *** was also studied for its effect on biofilm formation and disruption of preformed *** synergistic effect of *** was determined in combination with ***:Topical application of ***(10%w/w ointment)significantly increased(40.84%)the tensile strength in the incision wound *** also showed significant wound healing activity in excision model and such significant activity was observed from the 9th *** Soframycin displayed significant wound healing activity from the 6th *** was found that root extracts of *** revealed significant inhibition against all tested *** displayed antimicrobial activity against Cram positive(MIC 0.625 mg/mL)and Cram negative(MIC0.625-1.25 mg/mL).*** was able to reduce biofilm formation and also caused disruption of preformed biofilms in a manner similar to ***,gentamicin was found to be ineffective against biofilms formed by Gram negative *** to the fractional inhibitory concentration index,*** displayed synergistic activity when it was combined with ***:From this study it may be concluded that the root extract of *** revealed significant wound healing potential,which was supported and well correlated with pronounced antibacterial activity of the tested plant parts.

Assessment of Antifungal Activity of Some Himalayan Foliose Lichens Against Plant Pathogenic Fungi

In vitro antifungal activity of the acetone, methanol and chloroform extracts of four lichen species viz, Bulbothirx setschwanensis, Everniastrum nepalense, Heterodermia diademata, Parmelaria thomsonii were investigated against seven plant pathogenic fungi (Aspergillus flavus, A. fumigatus, Alternaria alternata, Fusarium oxysporum, F. solani, F. roseum and Penicillium citrinum) with reference to commercially available synthetic antifungal drug Ketoconazole (positive control). Lichen secondary metabolites were extracted using Soxhlet extractor and were further recovered through gentle evaporation of solvents in rotatory evaporator. Antifungal activity was analysed employing BauerKirby disc diffusion assay. Acetone and methanol extracts of lichenized fungi were found more effective against tested plant pathogenic fungi. Principal component analysis concluded that though, Ketoconazole was effective against four of the tested plant pathogenic fungi, acetone and methanol extracts of lichens were comparatively more effective against some broad spectrum plant pathogenic fungi (Fusarium oxysporum, F. solani, F. roseum).

Integration of Remote Sensing Data and GIS Tools for Accurate Mapping of Flooded Area of Kurigram, Bangladesh

Flood is the most devastating disaster in the present world which causes damage to environmental, social, economical and human lives at about 43% of all natural disasters. There are many flood hazard occurs in Bangladesh during the 19th century and 20th century in the different regions. These flood hazards have more catastrophic damages of huge area within human lives and other necessary properties of Bangladesh. The first step of flood management is to evaluate the area which is under threat of flood disaster. In this study here showed the importance of Remote Sensing (RS) data and Geographic Information System (GIS) tools to manage the flood related problems. Remote Sensing (RS) data and Geographic Information System (GIS) provide a lot of information to flood disaster management. ArcView GIS software tools are used for digitizing the base map and to create a flood risk zone of Kurigram, Bangladesh where images of remote sensing can be helped to determine the flood inundation areas. The integrated application of RS and GIS techniques for monitoring and flood mapping provides information for the decision makers. The study also grows attentions the need of cost-efficient methodology by creating a flood vulnerable map of Bangladesh.

Pediatric thermal burn injury:A retrospective analysis from pediatric care institutes,Eastern India

Objective:Pediatric burns pose difficult healthcare issues in underdeveloped *** to the scarcity of pediatric burn care facilities and qualified burn‑care staff,patients are frequently referred from a rural *** study explores the burn‑treatment approach that could be used in rural hospitals in the absence of a dedicated pediatric burn care *** and Methods:A retrospective analysis of clinical data of 199 children who received treatment for burn‑injury in an isolated general surgical *** debridement under anesthesia at the earliest was the cornerstone of the treatment plan for this *** the wounds healed,the treatment was repeated every alternate *** patient’s demographic data,number of surgical dressings,length of hospital stay,and outcome were ***:The mean age of the patients was 51.68±23.79 *** male‑female ratio was 1.45:*** survival rate was 99.00%.The disability rate was 4.52%.Univariate logistic regression identified the age group,total body surface area affected,depth of the wound,and time of intervention as statistically significant variables(P<0.05)predicting the *** logistic regression identified the time intervals between surgical intervention and injury as an important predictor of *** addition,delayed presentation and delayed intervention led to a longer hospital stay,and more interventions than the early intervention ***:Early debridement accelerates recovery and lessens the need for future skin grafts or contracture *** approach can be used in rural hospitals as well because pediatric burns can be manageable in an isolated general ward.

Role of subjective visual vertical in patients with posterior canal benign paroxysmal positional vertigo as a prognostic marker after canalith repositioning maneuver

Objective:To study the potential role of subjective visual vertical(SVV)as a prognostic marker for canalith repositioning maneuver(CRM)in patients with posterior canal benign paroxysmal positional vertigo(PC-BPPV)for the Indian ***:SVV was examined in 30 patients with PC-BPPV before and after canalith repositioning maneuver and after complete resolution of *** parameters included the mean of 10 angular tilt readings and direction of deviation,which were compared before and after CRM and following complete resolution of ***:The angle of SVV tilt was greater and deviated towards the affected ear before CRM in all patients,which decreased significantly shortly after CRM and continued to decrease after complete resolution of PC-BPPV(p<0.0001).Conclusions:SVV can be used to test utricular dysfunction in *** angle of tilt improves in response to CRM,which may be used as a prognostic marker in patients with PC-BPPV receiving CRM.

Nitric oxide mediated Staphylococcus aureus pathogenesis and protective role of nanoconjugated vancomycin

Objective:To investigate the relation between fruit seeds,plants residuals and appendicitis. Methods:Among cases that underwent appendectomy,the appendicitis cases having fruit seeds and undigested plant residuals in their etiology were examined ***, histopathological features,age,sex,and parameters of morbidity and mortality were used. Results:Fruit seed was found in one case(0.05%) with presence of pus in appendix lumen, undigested plant residuals in 7 cases(0.35%).It was determined that there were appendix inflammation in 2 of the plant residuals cases,while there were obstruction and lymphoid hyperplasia in the appendix lumen of 5 *** mortality was ***:The ratio of acute appendicitis caused by plants is minimal among all appendectomised patients, but avoidence of eating undigested fruit seeds and chewing plants well may help to prevent appendicitis.
