Diagnosis of Helicobacter pylori : What should be the gold standard?

Since the discovery of Helicobacter pylori(H. pylori) in 1983, numerous detection methods for the presence of the bacterium have been developed. Each one of them has been associated with advantages and disadvantages. Noninvasive tests such as serology, 13 C urea breath test(UBT) and stool antigen tests are usually preferred by the clinicians. Serology has its own limitation especially in endemic areas while 13 C UBT is technically very demanding. The stool antigen detection method, although specific, is usually associated with poor sensitivity. The 13 C UBT is believed to be specific, but with present revelation of the fact that stomach is colonized by many other urease producing bacteria makes it questionable. Histology, culture, rapid urease test and polymerase chain reaction(PCR) are the tests which are carried out on antral biopsies collected by invasive means. Histology has been proposed to be very sensitive and specific but the question is how by simply looking the morphology of the bacteria in the microscope, one can claim that the curved bacterium is exclusively H. pylori. Rapid urease test(RUT), the doctor's test, is also challenged because the presence of other urease producing bacteria in the stomach cannot be denied. Moreover, RUT has been reported with poor sensitivity specially, when density of the bacterium is low. Isolation of H. pylori is essential to investigate its growth requirements, antibiotic susceptibility testing, studying virulence factor to develop vaccine and many more explorations. It has also got several disadvantages i.e., special condition for transporting, media, incubation and few days waiting for the colonies to appear, apart from the speed essentially needed to process the specimens. Till date, majority of the microbiological laboratories in the world are not equipped and trained to isolate such fastidious bacterium. The option left is PCR methods to detect H. pylori 's DNA in gastric mucosa, gastric juice, saliva, dental plaques

Identification of EST–SSRs and molecular diversity analysis in Mentha piperita

EST sequences of Mentha piperita available in the public domain(NCBI) were exploited to develop SSR markers. A total of 1316 ESTs were assembled into 155 contigs and 653 singletons and of these, 110 sequences were found to contain 130 SSRs, with a frequency of1 SSR/3.4 kb. Dinucleotide repeat SSRs were most frequent(72.3%) with the AG/CT(43.8%)repeat motif followed by AT/AT(16.2%). Primers were successfully designed for 68SSR-containing sequences(62.0%). The 68 primers amplified 13 accessions of M. piperita and 54 produced clear amplicons of the expected size. Of these 54, 33(61%) were found to be polymorphic among M. piperita accessions, showing from 2 to 4 alleles with an average of2.33 alleles/SSR, and the polymorphic information content(PIC) value varied between 0.13 and 0.51(average 0.25). All the amplified SSRs showed transferability among four different species of Mentha, with a highest in Mentha arvensis(87.0%) and minimum in Mentha citrata(37.0%). The newly developed SSRs markers were found to be useful for diversity analysis, as they successfully differentiated among species and accessions of Mentha.

Recovery of Iron Bearing Minerals from Beneficiation Plant 2 Thickner Underflow of JSW Steel Limited

To identify and establish beneficiation techniques for slime, a comprehensive research work was carried out on beneficiation plant 2 thickener underflow (Tailing). Iron ore slimes (-45 micron ~80.0%) generated at beneficiation plant 2 assaying 46.7% Fe, 10.82% SiO2, and 4.58% Al2O3 have been subjected to various beneficiation studies like cyclone, wet high intensity magnetic separator and flotation to recover the iron bearing minerals. The effects of different operating parameters have been studied to get a product suitable for pellet making. Detailed beneficiation studies indicate that it is possible to obtain a product containing 61.8% to 66.6% Fe, 1.80% to 3.35% SiO2, and 1.65% to 2.40% Al2O3 with 8.0% to 22.3% weight recovery by adopting de-sliming followed by magnetic separation. Within this product grade range the pellet grade fines (JSW norms-minimum 63.0% Fe) consist of 63.0% Fe, 2.97% SiO2 and 2.48% Al2O3 with 16.9% weight recovery and 56.9% Fe recovery at 0.5 Tesla magnetic field intensity, pulsation rate 230 rpm and 2 min residence time.

Pseudomonas fluorescens-like bacteria from the stomach:A microbiological and molecular study

AIM:To characterize oxidase-and urease-producing bacterial isolates,grown aerobically,that originated from antral biopsies of patients suffering from acid peptic ***:A total of 258 antral biopsy specimens were subjected to isolation of bacteria followed by tests for oxidase and urease production,acid tolerance and aerobic *** selected isolates were further characterized by molecular techniques *** for 16S rRNA using universal eubacterial and HSP60 gene specific *** amplicons were subjected to restriction analysis and partial sequencing.A phylogenetic tree was generated using unweighted pair group method with arithmetic mean(UPGMA) from evolutionary distance computed with bootstrap test of *** of acidity tolerance of bacteria isolated from antrum was performed using hydrochloric acid from 10-7 mol/L to 10-1 mol/***:Of the 258 antral biopsy specimens collected from patients,179(69.4%) were positive for urease production by rapid urease test and 31%(80/258) yielded typical Helicobacter pylori(***) after 5-7 d of incubation under a microaerophilic environment.A total of 240(93%) antral biopsies yielded homogeneous semi-translucent and small colonies after overnight *** partial 16S rRNA sequences revealed that the isolates had 99% similarity with Pseudomonas species.A phylogenetic tree on the basis of 16S rRNA sequences denoted that JQ927226 and JQ927227 were likely to be related to Pseudomonas fluorescens(***).On the basis ofHSP60 sequences applied to the UPGMA phylogenetic tree,it was observed that isolated strains in an aerobic environment were likely to be ***,and HSP60 sequences had more discriminatory potential rather than 16S rRNA ***,this bacterium was acid tolerant for hours at low ***,a total of 250(96.9%) genomic DNA samples of 258 biopsy specimens and DNA from 240 bacterial isolates were positive for the 613 bp amplicons by targetin

Controlling the Flow of Microscopic Particles—The Role of Beam Size

Flow of micro particles and fluids is important in many microscopic systems. Here we present details of our finding of a directional flow of micro particles due to a single beam optical trap. It was found that the directional flow depends upon the size of optical trap, the number density of particles in the solution and the time after the trap was created. We suggest controlling the motion of microscopic particles in a fluid by varying a simple parameter like beam size for microfluidics applications.

Synergistic effect of Mucuna pruriens and Withania somnifera in a paraquat induced Parkinsonian mouse model

Parkinson’s disease (PD) is a neurodegenerative disorder characterized by the development of rigidity, resting tremors and postural instability. Recently, the focus of PD’s treatment has shifted towards herbal medicines. Mucuna pruriens (Mp) and Withania somnifera (Ws) are traditional herbal medicines known to have neuro-protective effects due to the L-DOPA present in Mp seed powder and withanoloides present in Ws root extract. Here, the synergistic effect of Mp and Ws in Parkinsonian mice induced by chronic exposure to paraquat was evaluated. Co-treatment with Mp and Ws for 9 weeks, significantly decreased the elevated nitrite levels and lipid peroxidation found in Parkinsonian mice. In behavioural tests, Mp and Ws treated mice showed a significant decrease in the time taken to cross a narrow beam, an increase in the time of stay on drum in rotarod test and an improvement in the hanging time. Furthermore, it was found that the use of Mp and Ws considerably improved the tyrosine hydroxylase expression in the substantianigra region of the brain. The results suggest that Mp and Ws may provide a platform for future drug discoveries and novel treatment strategies for PD.

Higher Body Mass Index is an Independent Predictor of Left Atrial Enlargement

Background: The influence of obesity on the cardiac geometry is less clear. Our objective was to determine if body mass index (BMI) was an independent predictor of left atrial enlargement (LAE) relative to LV thickness, dimension and function. Methods: Retrospective analysis of 90 consecutive patients with available echocardiographic data was performed. Patients were categorized into those with LAE (defined as >4.0 cm in transverse dimension by M-mode, n = 36) and those with normal LA size (n = 54). The clinical and echocardiographic variables were compared between the two groups. Results: Compared to patients with normal LA size, patients with LAE had significantly higher BMI (32 ± 8 vsi ± 8, p = 0.006), greater LV thickness, increased LV cavity dimension, and depressed LV systolic function. BMI was a strong and independent predictor of LAE. Left ventricular thickness and LV end-diastolic (LVED) dimension were other independent predictors of LAE. After adjusting for the significant effects of LV thickness and LV diameter, every unit increase in BMI resulted in an 8% increase in the odds of having LAE. Conclusions: 1) Higher BMI is an independent predictor of LAE. 2) Presence of LV enlargement and LV systolic dysfunction in these patients suggests that in the face of higher BMI, cardiac workload may exceed the compensatory LV hypertrophy and 3) LAE may be a consequence of the greater force developed by the LA to fill the LV.

Design of a Low Loss RF Mixer in Ku-Band (12 - 18 GHz)

The goal of this paper is to design and develop low conversion loss mixers at Ku Band (12 - 18 GHz) in microstrip line configuration. The Ku-band (Kurtz-under band) is primarily used for satellite Communications, particularly for editing and broadcasting satellite television. CAD tools (Agilent’s Advance Design System software) have been used for simulation and optimization of the circuits. 180 degree balanced mixer configuration has been adopted due to better RF to LO isolation requirement. The circuit is designed on microstrip line using RT Duroid (Dielectric Constant = 2.22). Initially a rat race coupler (180 degree hybrid) was designed and simulated and optimized for required performance successfully.

Immune response is known to develop against malignant tumours. Malignant tumours express newer antigen on their cell surface membrane which elicit immunological reaction in and around tumoural tissue. In early part of immune reaction neutrophil, eosinophil migrates followed by monocyte-macrophage cell. Tumor associated tissue eosinophilia (TATE) is believed to play a significant role in the biological behavior of the carcinoma. Eosinophil infiltrate in association with the head and neck squamous cell carcinoma (SCC) have been reviewed from time-to-time. The significance of such an association has been variably thought to be either a potential diagnostic tool for stromal invasion or as a prognostic indicator. The aim of this study was to investigate and evaluate the possible role of the tumour associated tissue eosinophilia (TATE) as a predictive indicator for the grading and establishing prognosis of the upper aerodigestive tract squamous cell carcinoma (SCC).
