A convenient one-pot three-component method for the preparation of tetra-substituted thiophene derivatives has been developed. Reaction ofacetyl acetone 1, phenyl isothiocynate 2 and 2-chloromethyI derivatives 3a-3c in the presence of potassium carbonate afforded the target compounds, namely ethyl 2-（4-acetyl-3-methyl-5-（phenylamino）thiophen-2-yl）-2-oxoacetate derivatives 4a-4e, ethyl 3-（4- acetyl-3-methyl-5-（phenylamino）thiophen-2-yl）-3-oxopropanoate derivatives 4f-4i, di（（4-acetyl-3- methyl-5-phenylamino）thiophen-2-yl）ketone derivatives 4j-4n in reasonable overall yields. The synthesized compounds were screened for antirnicrobial activity. The detailed synthesis, spectroscopic data and antimicrobial activities of synthesized compounds were reported.

A simplified tool for building layout design based on thermal comfort simulations

Thermal comfort aspects of indoor spaces are crucial during the design stages of building layout planning. This study presents a simplified tool based on thermal comfort using predicted mean vote （PMV） index. Thermal comfort simulations were performed for 14 different possible room Layouts based on window configurations. ECOTECT 12 was used to determine the PMV of these rooms for one full year, leading to 17,808 simulations. Simulations were performed for three different climatic zones in India and were validated using in-situ measurements from one of these climatic zones. For moderate climates, rooms with window openings on the south facade exhibited the best thermal comfort conditions for nights, with comfort conditions prevailing for approximately 79.25% of the time annually. For operation during the day, windows on the north facade are favored, with thermal comfort conditions prevailing for approximately 77.74% of the time annually. Similar results for day and night time operation for other two climatic zones are presented. Such an output is essential in deciding the layout of buildings on the basis of functionality of the different rooms （living room, bedroom, kitchen） corresponding to different operation times of the day.

The present review is intended to provide information on botany,ethnomedicinal uses,phytochemistry and biological activities of various parts of Euphorbia neriifolia(E. neriifolia). E. neriifolia has several ethnomedicinal uses. The latex of E. neriifolia is used as laxative,purgative,rubefacient,carminative and expectorant as well as in treatment of whooping cough,gonorrhea,leprosy,asthma,dyspepsia,jaundice,enlargement of the spleen,tumors,stone in the bladder,abdominal troubles and leucoderma. Leaves are brittle,heating,carminative,and good for improving the appetite and treatment of tumors,pains,inflammations,abdominal swellings and bronchial infections. Roots are used as symptomatic treatment of snake bite,scorpion sting and antispasmodic. Various plant parts or whole E. neriifolia extract and its isolates have been reported scientifically using various in-vivo and in-vitro experimental methods for anaesthetic,analgesic,anti-anxiety,anti-convulsant,anti-psychotic,anti-arthritis,anti-carcinogenic,antidiabetic,anti-diarrhoeal,anti-inflammatory,anti-thrombotic,antimicrobial,antioxidant,antiulcer,cytotoxic,death-receptor expression enhancing,dermal irritation,diuretic,hemolytic,immunomodulatory,radioprotective,scorpion venom and wound healing properties. It is reported to have chemical constituents like,neriifolin-S,neriifolin,neriifoliene,euphol,neriifolione,cycloartenol,nerifoliol,lectin,euphonerins A–G,3-O-acetyl-8-O-tigloylingol,taraxerol,antiquorin,etc. Identified chemical constituents are still required to be explored for their advanced isolation techniques and biological activities.

Objective:To report the indigenous people's uses of plants from a multidimensional perspective in a remote area where strong ethnobotanical cultural practices prevail. Methods:An ethnobotanical survey was conducted in the field during 2014-2016. Ethnobotanical data were gathered from 182 informants through oral interviews and semi-structured questionnaires. The distribution of plants was explored using both descriptive and graphical methods. Further,a Multinomial Logit Specification was applied to find out the probability of the occurrence of diverse utilization of plants in multipurpose domains. Results:The study identified 202 plant species distributed among 71 families and 156 genera. Ethnobotanical data indicate that there are more medicinal(36.96%) uses of plants as compared to all other use categories. The output from the Multinomial Logit Specifications(MLS) model reveals that perennial and non-woody plants are exploited more for medicinal and food uses than annual and woody plants. In the context of ethnomedicinal uses,aerial plant parts particularly leaves are more extensively used for the preparation of herbal recipes as compared to underground parts. Conclusions:The results of the study emphasize the need to create awareness among the local communities about the conservation status of plant species in order to maintain a sustainable resource of plant-derived materials into the future. The novel econometric approach employed in this study adds a new insightful methodology to the existing body of literature in the field of ethnobotany. We strongly recommend conservation measures,alongside phytochemical and pharmacological studies on the useful plant species identified in this study in order to ensure their sustainable and effective utilization.

The intention of current review is to make available up-to-date information on morphology, ecological biodiversity, medicinal uses, phytochemistry and pharmacolog-ical activities on different parts of Euphorbia tirucalli (E. tirucalli). This plant has a number of medicinal uses. Latex of E. tirucalli is vesicant and rubefacient which is used for rheumatism, warts, cough, asthma, ear-ache, tooth-ache and neuralgia. It acts as a purgative in small doses while in big doses it is bitter irritant and emetic. Milky juice is alexiteric, carminative and purgative. It is useful in whooping cough, gonorrhea, asthma, leprosy, dropsy, dyspepsia, enlargement of spleen, colic, jaundice and stone in bladder. The fresh milky juice is good alternative in syphilis and a good application in neuralgia. A decoction of branches is used in gastralgia and colic. Bark is used in treatment of fractures. Poultices prepared from the stem are useful to repair the broken bones. Boiled root liquid acts as an emetic in cases of snake-bite and for infertility in women. The wood is used for rafters, toys and veneering purposes. It is also useful against leprosy and foot paralysis subsequent to childbirth. E. tirucalli is reported to have euphol, β-sitosterol, euphorbol hexacosonate, cycloeuphordenol, cyclotirucanenol, tirucalicine, tri-methyl ellagic acid, gallic acids, terpenic alcohol, isoeuphorol, taraxasterol, tirucallol, euphor-one, euphorcinol, euphorbins, 12-deoxy-4β-hydroxyphorbol-13-phenyl acetate-20-acetate, 12, 20-dideoxyphorbol-13-isobutyrate, glut-5-en-3-β-ol, 3,3′-di-O-methylellagic acid, euphorbin-A (polyphenol), tirucallin-A (7) (tannin), tirucallin-B (11), euphorbin-F (14) (dimers), cycloartenol, 24-methylenecycloartenol, ingenol triacetate, 12-deoxy-4β-hydroxyphorbol-13-phenyl acetate-20-acetate, taraxerone, euphorginol, taraxerol, cam-pesterol, stigmasterol, palmitic acid, linoleic acid,β-amyrin, etc. active phytoconstituents. Development and validation of multicentre study on novel Artificial Intelligence-based Cardiovascular Risk Score(AICVD)

Objective Cardiovascular diseases(CVD)are one of the most prevalent diseases in India amounting for nearly 30%of total deaths.A dearth of research on CVD risk scores in Indian population,limited performance of conventional risk scores and inability to reproduce the initial accuracies in randomised clinical trials has led to this study on large-scale patient *** objective is to develop an Artificial Intelligence-based Risk Score(AICVD)to predict CVD event(eg,acute myocardial infarction/acute coronary syndrome)in the next 10 years and compare the model with the Framingham Heart Risk Score(FHRS)and *** Our study included 31599 participants aged 18–91 years from 2009 to 2018 in six Apollo Hospitals in India.A multistep risk factors selection process using Spearman correlation coefficient and propensity score matching yielded 21 risk factors.A deep learning hazards model was built on risk factors to predict event occurrence(classification)and time to event(hazards model)using multilayered neural ***,the model was validated with independent retrospective cohorts of participants from India and the Netherlands and compared with FHRS and *** The deep learning hazards model had a good performance(area under the curve(AUC)0.853).Validation and comparative results showed AUCs between 0.84 and 0.92 with better positive likelihood ratio(AICVD−6.16 to FHRS−2.24 and QRisk3−1.16)and accuracy(AICVD−80.15%to FHRS 59.71%and QRisk351.57%).In the Netherlands cohort,AICVD also outperformed the Framingham Heart Risk Model(AUC−0.737 vs 0.707).Conclusions This study concludes that the novel AI-based CVD Risk Score has a higher predictive performance for cardiac events than conventional risk scores in Indian population.

Healthy cartilage is a water-filled super lubricious *** type II provides it structural stability,and proteoglycans absorb water to keep the cartilage in a swollen condition,providing it the ability to creep and provide weeping ***(OA)is a degenerative and debilitating disorder of diarthrodial joints,where articular cartilage damage originates from enzymatic degradation and mechanical damage(wear).The objective of this research is to observe the level of cartilage damage present in knee arthroplasty patients and to understand the friction and creep behavior of enzymatically degraded bovine cartilage in ***(Lat)and medial(Med)condylar cartilages from OA patients undergoing total knee arthroplasty showed signs of enzymatic degradation and mechanical *** cartilages were exposed to collagenase III and chondroitinase ABC to degrade collagen and proteoglycans,*** loss of proteoglycans or collagen network and morphological changes were observed through histology and the atomic force microscope(AFM),respectively.A significant effect on creep due to enzymatic treatment was not *** the enzymatic treatment was found to significantly decrease the coefficient of friction(COF)at 4 N,while higher COF was shown from chondroitinase ABC degraded cartilage at 40 *** III treatment leads to the release of intact proteoglycans at the sliding interface,while chondroitinase ABC treatment leads to the loss of chondroitin sulfate(CS)from the *** ABC-digested bovine cartilage mimicked patient samples the best because of the similar distributions of proteoglycans,collagen network,and friction behavior.

This study was conducted to evaluate the effects of proline and glutamine on in vitro callus induction and subsequent regeneration and to develop a reproducible and highly efficient plant regeneration protocol in four rice genotypes, viz. Pawana, jaya, Indrayani and Ambemohar. Considerable variation in response to plant growth regulators and amino acid supplements used was observed in all the four genotypes. Medium supplemented with proline and glutamine was shown to be superior to medium without proline and glutamine. The best callusing from mature embryo was observed on Murashige and Skoog （MS） medium supplemented with 2.0 mg/L 2,4-dichlorophenoxyacetic acid （2,4-D）, 500 mg/L proline and 500 mg/L glutamine. Shoot induction was higher in the callus obtained from medium supplemented with 500 mg/L proline and 500 mg/L glutamine. The highest shoot regeneration frequency （83.2%） was observed on MS medium with 2.0 mg/L benzylaminopurine, 0.5 mg/L 1-naphthaleneacetic acid, 500 mg/L proline, and 500 mg/L glutamine in the callus obtained from MS medium supplemented with 2.0 mg/L 2,4-D, 500 mg/L proline and 500 mg/L glutamine. Among the four genotypes, Pawana has the highest regeneration efficiency （83.2%）, whereas the regeneration efficiency of the rest three rice genotypes was in the range of 32.0% to 72.3%. This optimized regeneration protocol can be efficiently used for Agrobacterium mediated genetic transformation in rice.
