Crystal engineering concept has been utilized to modify the physico-chemical parameters of a naturally occurring alkaloid, quinine sulphate, by exploring its H-bond interactions to generate different forms. Quinine sulphate is found to exist in four different crystal forms. The Forms I and II depict endo/exo events suggesting conversion of metastable low melting forms to higher melting and stable form indicated by sharp melting endotherms. The low melting form IL is found to be monotropically related to high melting Form IH while low melting Form IIL is enantiotropically related to high melting Form IIH. The Form III and IV showed broad endotherms accompanied by mass loss in TGA prior to melting indicating the existence of solvatomorphism. The solvent molecules are tightly bound in the crystal lattice of the drug molecules which is shown by high values of the binding energies of the solvents in these two forms. The enthalpy of solution was found to be endothermic for all the forms which followed the order: Form O > Form II > Form III > Form I > Form IV and is further related to the lattice energy suggesting Form II to be least crystalline. The solubility for Form II was found to be highest with maximum release rate in dissolution studies. The effectiveness of new polymorphic forms was confirmed by performing in vivo antimalarial activity against P. berghei infection. The studies have shown an increase in antimalarial activity of Form IV concluding a successful development of new polymorphic form.

A superionic mixed metal iodide glass system [(PbI2 –CuI ) - Ag2O- V2O5- B2O3] has been prepared by splat quenching technique. The prepared glass samples are characterized by X-ray diffraction (XRD) and differential scanning calo- rimetry (DsC). The effect of mixed metal iodide salts concentration on electrical properties has been investigated by the complex impedance spectroscopy (CIs). AC conductivity analysis is carried out in the frequency range of 2MHz-1Hz at different temperatures and an enhancement in the conductivity with iodide salts is the main feature of the observed re- sults with an anomaly at 20 mole % of PbI2 –CuI.

Aims: Obesity is the major contributor of the metabolic syndrome (MetS), but a unique phenotype of obesity known as metabolically healthy obese (MHO) shows healthier metabolic profile; however understanding of their biochemical correlates is poorly understood. Obesity is defined by Body mass index (BMI), but controversy exists regarding ethnic-specific BMI cut-offs. The present study used the Asian Indian BMI cut-offs to assess relationships of MHO phenotypes with oxidative stress (OS) and inflammation. Methods: In this case-control study, 299 metabolically-healthy (MH) respondents were divided into four groups as per Asian criteria for obesity: MH non-obese (MHNO), MH overweightspan> (MHOW), MHO and MH severely obese (MHSO). Their oxidative stress and pro-inflammatory markers were measured. Obesity (Ob) is an accepted major risk factor for the metabolic syndrome (Mets), a combination of at least three of five risk factors, which predispose to high oxidative stress (Os), but all obese do not show symptoms of Mets. There is dearth of data comparing Os homeostasis of severely obese adults with and without Mets, and need for biomarkers to help in differential diagnosis. Erythrocytic lipid and protein damage markers, malondialdehyde (MDA) and protein carbonyl (PCO), antioxidant enzymes erythrocytic superoxide dismutase(sOD), catalase (CAT), plasma glutathione peroxidase (GPX), and total antioxidant capacity (TAC) as ferric-reducing-ability-of-plasma (FRAP) were compared to understand Os homeostasis among 102 severely Ob (body mass index > 30), 102 Ob with severe (z-score > 2) Mets as per National Cholesterol Education Program-Adult Treatment Panel III guidelines and 100 healthy non-obese Controls. MDA/PCO and all antioxidant enzymes were lowest for ObMets, followed by Ob, indicating greater damage to protein moieties of the erythrocytic membrane. Multiple regression analysis confirmed z-scores > 2 as significant predictor of lowered enzymes and TAC. Receiver Operator Curve analysis predicted that TAC was the most potential biomarker for the diagnosis and prognosis of Mets with an Odds Ratio of 88.5 indicating the high probability that FRAP would be low for ObMets (z-score > 2) than for Ob with BMI > 30, but z-scores s qualified as the most effective biomarker to distinguish between severely obese respondents with and without metabolic syndrome, and as a useful candidate for study of homeostatic breakdown in metabolic syndrome and the importance of z-score in assessment of Mets in obese respondents.

Development and Validation of Bioanalytical Method for Determination of Telmisartan and Hydrochlorothiazide Using HPTLC in Human Plasma

A simple, sensitive, rapid and economic high performance thin layer chromatographic method has been developed for determination of telmisartan and hydrochlorothiazide in human plasma using paracetamol as an internal standard. The plasma sample was extracted using mixture of methanol-acetonitrile (3.0:0.1, v/v). A concentration range from 200, 400, 600, 800, 1000, 1200 ng/spots were used for calibration curve of hydrochlorothiazide and telmisartan respectively. The percent recovery of telmisartan and hydrochlorothiazide was found to be75.98 and 81.91%. The mobile phase consists of chloroform: methanol: toluene (8:2:4 v/v/v). Densitometric analysis was carried out at wavelength 278 nm. The Rf values for hydrochlorothiazide, paracetamol and telmisartan were 0.28 ± 0.05, 0.50 ± 0.05, 0.66 ± 0.05 respectively. The stability of telmisartan and hydrochlorothiazide in plasma were confirmed during three freeze-thaw cycles (?20?C), on bench during 24 hours and post preparative during 48 hours. The proposed method was validated statistically and by performing recovery study for determination of telmisartan and hydrochlorothiazide in human plasma.

A Novel Method to Fix Type C3 Distal Femur Fractures with Bone Defect Loss Using “Harms Cage”

A case of compound C3 distal femur fracture with 12 cm bone defect. After thorough debridement, the Harms cage was used to reconstruct the medial column and the fracture was fixed with the locking distal femur plate laterally. The Harms cage prevents the cantilever effect and eliminates the need for medial plating. This avoids another medial incision and preserves medial biology. At 18 months’ follow-up, plain radiographs demonstrated full reconstruction of the medial column with good range of motion at the knee joint. This technique avoids the need of dual plating in these fractures with good results.

Film of PEO containg AgCF3sO3 were prepared by the solution casting technique. Fourier transform infrared (FTIR) spectroscopy have been carried out on a series of complexes containing poly (ethylene oxide) PEO and silver triflate (AgCF3sO3) salt. spectral analysis of all the samples has revealed the complexation of silver ions with oxygen in PEO. The ac conductivity and electrical modulus of the samples prepared have been analyzed. The ac conductivity was observed to obey the Universal power law. The temperature dependence of the power law exponent n is reasonably interpreted by the overlapping large polaron tunneling (OLPT) model. The imaginary part, M” of electrical modulus shows the formation of dispersion peak. The plot of log conductivity relaxation times and log sigma as a function of salt concentration was in accordance with each other.

The electrical properties of proton ion beam irradiated glass samples are carried out by impedance spectroscopy in the frequency range from 10 Hz to 32 MHz. The ion beam of energy 3 MeV and fluence of of 1014 particles cm-2 was chosen for irradiation. The conductivity of the super ionic glass samples increases after irradiation and other electrical parameters like dielectric constant, dielectric loss and modulus of the proton irradiated glass samples as a function of glass composition and temperature are observed to change. The dielectric constant and the dielectric losses are increased after irradiation and the modulus parameters confirm the non-Debye nature for irradiated samples also.
