Aim: The present study was aimed to investigate Integrin beta-like 1(ITGBL1) methylation pattern and its clinical relevance in patients with acute myeloid leukemia(AML). Method: Real-time methylation-specific PCR(RQ-MSP) and bisulfite sequencing PCR(BSP) were performed to detect the methylation of ITGBL1 promoter. Real-time quantiative PCR(RQ-PCR) was performed to analyze ITGBL1 transcript level. Results: ITGBL1 methylation level in 131 AML patients was significantly higher than 29 controls(Phere were no significant differences in sex, age, white blood cells, hemoglobin, platelet count, percentage of marrow blasts, karyotypes and ten gene mutations between ITGBL1 hypermethylated and hypomethylated patients(P>0.05). ITGBL1 hypermethylated patients had significantly shorter overall survival(OS) and leukemia-free survival(LFS) than ITGBL1 hypomethylated patients in whole AML cohort(P=0.009 and 0.043, respectively) and non-acute promyelocytic leukemia(APL) patients(P=0.023 and 0.039, respectively). Multivariate analysis confirmed that ITGBL1 methylation served as an independent prognostic factor in both whole-cohort AML patients(HR = 1.829, 95% CI = 1.059-3.158, P = 0.030) and non-APL patients(HR = 1.938, 95% CI =1.111-3.380, P = 0.020). Furthermore, ITGBL1 methylation level was significantly decreased in follow-up AML patients who achieved complete remission after induction therapy(P=0.001). ITGBL1 methylation negatively correlated with ITGBL1 expression in AML patients(R=-0.328, P=0.008).Moreover, demethylation of ITGBL1 could increase ITGBL1 expression in the K562 leukemic cell line(P<0.05). Conclusions: ITGBL1 hypermethylation is a potential biomarker for predicting prognosis and monitoring disease status in AML patients.

Background: DOK-1 and DOK-2(DOK1/2) are closely related members of downstream of tyrosine kinase(DOK) family genes, which are found to be frequently rearranged in several hematopoietic cancers. However, the clinical implications of DOK1/2 in acute myeloid leukemia(AML) remain largely unknown. Methods: To investigate the clinical significance, real-time quantitative PCR was carried out to detect DOK1/2 expressions in 125 de novo AML patients and 28 healthy controls. Real-time quantitative methylation-specific PCR(RQ-MSP) and bisulfite sequencing PCR(BSP) were applied to detect DOK1/2 methylation level and density. Results: DOK1/2 expressions were significantly down-regulated in AML patients. The promoters of DOK1/2 were highly hypermethylated and negatively correlated with DOK1/2 expressions in AML patients. In addition, we also confirmed that DOK1/2 expressions could be restored by DOK1/2 demethylation using 5-aza-2′-deoxycytidine in leukemia cell line THP-1. Survival analyses showed that low-expressed DOK1/2 were associated with markedly shorter overall survival and leukemia free survival in both whole-cohort AML and non-M3 AML patients, but not in cytogenetically normal AML patients. Multivariate analyses further revealed that DOK1/2 were act as independent prognostic factors in AML patients. Conclusion: These findings indicate that decreased DOK1/2 expressions associated with promoter hypermethylations predict adverse prognosis in AML.

BACKGROUND: Altered expression of the BCL-2 family member MCL-1 has been linked to the progression and outcome of various malignancies. However, MCL-1 expression pattern in acute myeloid leukemia(AML) and its impact on prognosis remain poorly defined. METHODS: Real-time quantitative PCR was carried out to detect the pattern of MCL-1 expression in AML patients. RESULTS: MCL-1 expression was significantly up-regulated in AML compared with controls(P=0.042). High MCL-1 expressed patients had a higher incidence of NPM1 mutation(P=0.030). Moreover, MCL-1 overexpression correlated with lower complete remission(CR) rate and shorter overall survival(OS) time. Meanwhile, Cox regression analyses revealed that overexpression of MCL-1 acted as an independent risk factor not only in non-APL patients but also in CN-AML patients(P=0.011 and P=0.045, respectively). Furthermore, gene expression profiling(GEP) data also confirmed the adverse prognostic impact of MCL-1 high-expression in CN-AML patients. In follow-up patients, MCL-1 expression level decreased after CR compared with newly diagnosis time(P=0.020) and increased after relapse(P=0.004). CONCLUSION: Our findings suggest that MCL-1 overexpression predicts poor prognosis and can be used for disease monitoring.

We investigate the properties of the spectral function of the fermionic operator in the field theory which is dual to a 4-dimensional massive gravity. We first study the Fermi surface and the dispersion relation in the dual boundary theory. We find that as the massive parameters is decreased, the Fermi momentum becomes lower and the low energy excitation near Fermi surface behaves more like non-Fermi liquid. Then, we introduce a dipole coupling in the bulk theory and explore the emergence of a gap in the fermionic spectral function. It is found that larger critical dipole coupling is needed to open the gap than that in Einstein gravity. Accordingly,in the field theory dual to massive gravity, it requires stronger negative dipole coupling to generate the marginal Fermi liquid.

A sensitive flow injection chemiluminescence method has been developed for the detection of resveratrol in red wine based on the fact that resveratrol can greatly enhance chemiluminescence reaction between KMnO4 and HCHO in sulfuric acid medium. Analytes were pre-concentrated on solid sorbents （C18 solid-phase extraction cartridges）. Under the optimum conditions, the proposed method allows the measurement of resveratrol over the range of 1.32 × 10^-8 to 1.32 × 10^-5 mol/L with a detection limit of 3.30 ×10^-9 mol/L, and the relative standard deviation for 1.32 ×10^-5 mol/L resveratrol （n = 11） is 3.8%. This method has been successfully applied for the determination of the resveratrol in red wine. Furthermore, the possible reaction mechanism was also discussed.

To provide precise prediction of tunnelling-induced deformation of the surrounding geomaterials,a framework for derivation of rigorous large-strain solutions of unified spherical and cylindrical cavity contraction is presented for description of confinement-convergence responses for deep tunnels in *** the tunnelling-induced large deformation,logarithmic strains are adopted for cavity contraction analyses in linearly elastic,non-associated Mohr-Coulomb,and brittle Hoek-Brown *** with approximate solutions,the approximation error indicates the importance of release of small-strain restrictions for estimating tunnel convergence profiles,especially in terms of the scenarios with high stress condition and stiffness degradation under large *** ground reaction curve is therefore predicted to describe the volume loss and stress relaxation around the tunnel *** stiffness of circular lining is calculated from the geometry and equivalent modulus of the supporting structure,and a lining installation factor is thus introduced to indicate the time of lining installation based on the prediction of spherical cavity contraction around the tunnel opening *** study also provides a general approach for solutions using other sophisticated geomaterial models,and serves as benchmarks for analytical developments in consideration of nonlinear large-deformation behaviour and for numerical analyses of underground excavation.

Aims Root and heterotrophic respiration may respond differently to environmental variability,but little evidence is available from largescale *** we aimed to examine variations of root and heterotrophic respiration across broad geographic,climatic,soil and biotic *** We conducted a synthesis of 59 field measurements on root and heterotrophic respiration across China’s *** Findings Root and heterotrophic respiration varied differently with forest types,of which evergreen broadleaf forest was significantly different from those in other forest types on heterotrophic respiration but without statistically significant differences on root *** results also indicated that root and heterotrophic respiration exhibited similar trends along gradients of precipitation,soil organic carbon and satellite-indicated vegetation ***,they exhibited different relationships with temperature:root respiration exhibited bimodal patterns along the temperature gradient,while heterotrophic respiration increased monotonically with ***,they showed different relationships with MOD17 GPP,with increasing trend observed for root respiration whereas insignificant change for heterotrophic *** addition,root and heterotrophic respiration exhibited different changes along the age sequence,with insignificant change for root respiration and decreasing trend for heterotrophic ***,these results suggest that root and heterotrophic respiration may respond differently to environmental *** findings could advance our understanding on the different environmental controls of root and heterotrophic respiration and also improve our ability to predict soil CO_(2) flux under a changing environment.

Annealing Effects on Properties of Bi—Substituted Garnet Films with Perpendicular Anisotropy Prepared by a Sol—Gel Method

Bi-substituted garnet films with perpendicular anisotropy have been prepared by a sol-gel method. Annealing effects on magneto-optical properties and structures of the films are studied. The results indicate that the Kerr hysteresis loops of garnet films have good squareness at annealing temperatures above 650°C, the coercive force Hc of these films increases from 1070 to 1570 Oe with the increasing temperature from 650°C to 700°C. The Kerr rotation of the films was changed with the increasing annealing temperature, and there is a maximum value (0.82°) at 675°C. It was found that the structure of the films is a single phase of garnet without preferring orientation when the annealing temperature is about 650°C, but a new phase (Bi2Al4O9) can be formed when the annealing temperature reaches 675°C, another new phase (Bi2Fe4O9) appears when the annealing temperature is about 700°C.

Au Nanoparticle-DNAzyme Dual Catalyst System for Sensitively Colorimetric Detection of Thrombin

A novel colorimetric aptasensor was developed for thrombin detection with high sensitivity and specificity The assay takes the advantage of Au nanoparticles-DNAzyme as a dual catalytic system for signal amplification. Au nanparticles were modified with peroxidase mimicking DNAzyme sequence as well as thrombin binding aptamer. And then the thrombin binding aptamer hybridized with its complementary sequence which was immobilized on the surface of the magnetic nanoparticles to construct the colorimetric aptasensor. In the presence of thrombin, the tar- get-induced displacement takes place, resulting in the dissociation of the aptasensor. The DNAzyme functioned Au NPs are released due to the combine ability of thrombin binding aptamer with thrombin. The released Au NPs are capable of catalyzing the colorless 2,2＇-azino-bis（3-ethylbenzothiazoline）-6-sulfonic acid（ABTS） conversion into a blue-green product by H202-mediated oxidation, thus can amplify the colorimetric readout signals of thrombin detec- tion. Such a device can serve as a novel selectivity sensor for thrombin with a detection limit of 0.6 nmol/L.
