Inter Simple Sequence Repeats （ISSR） markers were used to assess genetic diversity within and among populations of dwarf mountain pine （Pinus rnugo Turra） growing in the Tatra National Park （UNESCO Biosphere Reserve） in Southern Poland （Central Europe）. The analyzed population belongs to two different geobotanical sub-districts： the Western and High Tatras. The level of genetic diversity assessed in this study for P. mugo is generally comparable to that reported for the other pine species in the Pinaceae family assessed by ISSR markers, especially with respect to Nei＇s genetic diversity and the percentage of polymorphic bands. Bayesian analysis clustered the analyzed populations into two groups, corresponding to their geobotanical locations in the Tatras. Significant divergence between the two genetical clusters was supported by the results of Analysis of Molecular Variance （AMOVA）. According to the Mantel test, there was no correlation between the genetic distance and the geographical distance. The present study confirms the existence of two genetically distinct dusters of P. mugo populations in the Tatra Mountains. The observed high population-genetic differentiation of *** in the Tatras could be attributed to several genetic, environmental and historical factors occurring in this mountain area.

Background： Grain legumes represent a valuable energy source in pig diets due to their high starch content. The present study was conducted to determine the content and apparent ileal digestibility（AID） of starch in different grain legume cultivars for pigs by means of both a polarimetric and enzymatic method for starch ***： Three experiments were conducted with six barrows each which were fitted with ileal T-cannulas. In total, 18 diets including six different cultivars of faba beans（Vicia faba L.） and peas（Pisum sativum L.）, five different cultivars of lupins（Lupinus luteus L., Lupinus angustifolius L.）, and one diet with a soybean meal（SBM） were ***： The starch content of faba beans and peas was greater（P ＆lt; 0.05） when determined polarimetrically than enzymatically（438 vs. 345 g/kg dry matter（DM） in faba beans and 509 vs. 390 g/kg DM in peas, respectively）.Considerable lower starch contents were obtained in lupins and SBM, with 82 and 48 g/kg DM（analyzed polarimetrically） and ＆lt;1.1 and 3 g/kg DM（analyzed enzymatically）, respectively. Mean values for contents of neutral detergent fiber（NDF） and acid detergent fiber（ADF） in grain legumes ranged from 111 and 79 g/kg DM in peas to248 and 207 g/kg DM in lupins, respectively. Contents of condensed tannins in the colored flowered faba bean cultivars ranged from 2.1 to 7.4 g/kg DM. The AID of starch was greater（P ＆lt; 0.05） in pea than in faba bean cultivars,and using the polarimetric starch determination method resulted in greater（P ＆lt; 0.05） digestibility values than using enzymatic starch analysis（84 vs. 80% in faba beans and 86 vs. 83% in peas）. Moreover, AID of starch differed（P ＆lt; 0.05）within pea cultivars and starch digestibility in faba beans decreased linearly（P ＆lt; 0.05） as the content of condensed tannins increased. This study presents a methodology for assessment of the condition of hiking trails(HTs) and their impact zones in the central part of the Low Tatra mountain range, based on comparison of two complex data sets gathered in 33 years apart. The first field investigation was performed in 1980 and 1981, and the second in 2013 and 2014. The main goal was to perform a landscape typology in order to assess the susceptibility of landscape complexes to occurrence of anthropogenic and natural destruction processes and to assess the condition and prospects of HTs and their impacts on the adjacent alpine environment. Landscape complexes were characterised by selected abiotic, biotic and technical parameters of HTs and their impact zones. Due to the high variability of these parameters over the length of the HTs, we had to decide on how to map them. This was done using a square grid with 100 m-sized cells. For each cell with HT, the parameters were assigned the typical value within that cell. In total, 26.3 km of HT were studied, stretching over 266 grid cells. On comparison of the two data sets, it was seen that, 64%(171 grid cells) display a generally positive condition, with 54%(143 grid cells) even exhibiting significant improvement or continuously positive state of their condition. 36%(95 grid cells) were in bad condition, including 3%(9 grid cells) whose state had deteriorated, and 2%(6 grid cells) whose state had significantly deteriorated, in the time between the two assessments.

The objective of this study was to investigate whether medical therapy can reduce sperm aneuploidy levels and irfiprove the results of intracytoplasmic sperm injection （ICSI） in patients with severe idiopathic oligoasthenoteratospermia （OAT）. Thirty-three infertile couples requiring ICSI because of severe idiopathic OAT after at least one unsuccessful ICSI cycle were considered. Semen parameters （concentration, motility and morphology）, the percentage of aneuploid sperm and the results of ICSI （the number of oocytes fertilized, embryos transferred, biochemical pregnancies, clinical pregnancies and live births） were compared before and after a 3-month course of treatment with L-carnitine 1 g given twice per day＋acetyI-L-carnitine 500 mg given twice per day＋one 30-mg cinnoxicam tablet every 4 days. Aneuploidy was assessed using fluorescent in situ hybridisation （FISH） performed on chromosomes X, Y, 13, 15, 16, 17, 18, 21 and 22. The results showed that 22 of the 33 patients had a reduced frequency of aneuploid sperm and improved sperm morphology after treatment （group 1）, and 11 showed no change （group 2）. The numbers of biochemical pregnancies, clinical pregnancies and live births were significantly higher in group I than in group 2. No significant difference was found between the groups regarding the numbers of oocytes fertilized and embryos transferred. The side effects were negligible. The numbers of ICSI pregnancies and live births in severe idiopathic OAT patients improved with a course of L-carnitine, acetyI-L-carnitine and cinnoxicam.

Oligosaccharide Mass Profiling （OLIMP） allows a fast and sensitive assessment of cell wall polymer structure when coupled with Matrix Assisted Laser Desorption Ionisation Time Of Flight Mass Spectrometry （MALDI-TOF MS）. The short time required for sample preparation and analysis makes possible the study of a wide range of plant organs, revealing a high degree of heterogeneity in the substitution pattern of wall polymers such as the cross-linking glycan xyloglucan and the pectic polysaccharide homogalacturonan. The high sensitivity of MALDI-TOF allows the use of small amounts of samples, thus making it possible to investigate the wall structure of single cell types when material is collected by such methods as laser micro-dissection. As an example, the analysis of the xyloglucan structure in the leaf cell types outer epidermis layer, entire epidermis cell layer, palisade mesophyll cells, and vascular bundles were investigated. OLIMP is amenable to in situ wall analysis, where wall polymers are analyzed on unprepared plant tissue itself without first isolating cell walls. In addition, OLIMP enables analysis of wall polymers in Golgi-enriched fractions, the location of nascent matrix polysaccharide biosynthesis, enabling separation of the processes of wall biosynthesis versus post-deposition apoplastic metabolism. These new tools will make possible a semi-quantitative analysis of the cell wall at an unprecedented level.
