Simple LC Isocratic methods Development, Validation, and Application in the Analysis of Poor Quality Antimalarial medicines

Liquid chromatographic methods in isocratic mode for the analysis of poor quality medicines are privileged due to their simplicity and facility in methods development. They are generally fast;do not need to be re-equilibrated between sample injections;have larger flexibility with acceptable changes on different column dimensions;and are applicable to LC systems equipped with simple or high developed pumps. In this study, we focused on developing simple isocratic methods using classical mobile phase composed by methanol and ammonium formate buffer for the analysis of most common antimalarial medicines marketed in malaria endemic countries and susceptible of being counterfeit/falsified, substandard and degraded. The selected medicines were quinine and related cinchona alkaloids in tablets and injectable forms;artemether/lumefantrine tablets;and artemisinin compounds (arteether, artemether, and artesunate) in injectable forms. The current methods were developed thanks to simple methodological approach consisting in sequential isocratic runs through adjustment or adaptation of existing methods to obtain optimal analytical conditions without complex design of experiments that might be long and costly. Then, the new methods presented shorter analysis time;allowed increase of sample analysis throughput;and obviously consumed little mobile phase solvents on classical analytical columns: 50 - 250 mm of length (L), 4.6 mm of internal diameter (I.D.), and 3.5 - 5.0 μm of particle size (dp).

Strawberry(Fragaria×ananassa)fruits are an excellent source of L-ascorbic acid(AsA),a powerful antioxidant for plants and *** the genetic components underlying AsA accumulation is crucial for enhancing strawberry nutritional ***,we unravel the genetic architecture of AsA accumulation using an F1 population derived from parental lines‘Candonga’and‘Senga Sengana’,adapted to distinct Southern and Northern European *** account for environmental effects,the F1 and parental lines were grown and phenotyped in five locations across Europe(France,Germany,Italy,Poland and Spain).Fruit AsA content displayed normal distribution typical of quantitative traits and ranged five-fold,with significant differences among genotypes and *** content in each country and the average in all of them was used in combination with 6,974 markers for quantitative trait locus(QTL)*** stable QTLs for AsA content were detected in linkage group(LG)3A,LG 5A,LG 5B,LG 6B and LG *** genes were identified within stable QTL intervals and expression analysis in lines with contrasting AsA content suggested that GDP-L-Galactose Phosphorylase FaGGP(3A),and the chloroplast-located AsA transporter gene FaPHT4;4(7C)might be the underlying genetic factors for QTLs on LG 3A and 7C,*** show that recessive alleles of FaGGP(3A)inherited from both parental lines increase fruit AsA ***,expression of FaGGP(3A)was two-fold higher in lines with high *** here identified represent a useful resource for efficient selection of new strawberry cultivars with increased AsA content.

Understanding the immune responses elicited by SARS-CoV-2 infection is critical in terms of protection against reinfection and,thus,for public health policy and vaccine development for *** this study,using either live SARS-CoV-2 particles or retroviruses pseudotyped with the SARS-CoV-2 S viral surface protein(Spike),we studied the neutralizing antibody(nAb)response in serum samples from a cohort of 140 SARS-CoV-2 qPCR-confirmed infections,including patients with mild symptoms and also more severe forms,including those that required intensive *** show that nAb titers correlated strongly with disease severity and with anti-spike IgG ***,patients from intensive care units exhibited high nAb titers;conversely,patients with milder disease symptoms had heterogeneous nAb titers,and asymptomatic or exclusive outpatient-care patients had no or low *** found that nAb activity in SARS-CoV-2-infected patients displayed a relatively rapid decline after recovery compared to individuals infected with other ***,we found an absence of cross-neutralization between endemic coronaviruses and SARS-CoV-2,indicating that previous infection by human coronaviruses may not generate protective nAbs against ***,we found that the D614G mutation in the spike protein,which has recently been identified as the current major variant in Europe,does not allow neutralization ***,our results contribute to our understanding of the immune correlates of SARS-CoV-2-induced disease,and rapid evaluation of the role of the humoral response in the pathogenesis of SARS-CoV-2 is warranted.

Inventory of Estuarine and Lagoonal Ecosystems Subjected to Sand-mining Activities in Southern Benin (West Africa)

Sand is a useful resource for the development of any society. As such, the issues related to its extraction are everywhere present in Benin, especially in the southern part of the country. For the purposes of eventually characterizing those issues, the present study is undertaken with a focus on inventorying estuarine and lagoonal ecosystems which are subjected to sand mining in southern Benin. The study area covers both the Eastern Complex and the Western Complex. The Eastern Complex includes the Porto-Novo Lagoon and the Ouémé River delta;while the Western Complex includes the Coastal Lagoon, the Aheme Lake along with its channels, and the mono Basin. The method used in this study is a systematic inventorying of all known and documented sites as kept on record with relevant state offices in charge of the management of those resources. Then, the data were completed by combing the whole area for direct census observations and field interviews with stakeholders. All inventoried sites have been geo-referenced using a GARmIN GPS and their surface areas have been established. To date, 43 sand-mining sites have been identified, of which 29 are located in the Oueme Delta and in the Porto-Novo Lagoon, 8 are in the Coastal Lagoon, and 6 are in the Aheme Lake and its channels as well as in the mono River basin. The areas are small in the Oueme Delta, where extraction is mostly manual, but get to be more than 89-ha wide in other places such as the Coastal Lagoon where the process is rather mechanized.

Distortions and Deformations of metaled meso-Substituted and Unsubstituted Porphyrins and Derivatives in Crystal Structures

Experimental crystallographic structural parameters of a range of metaled meso-substituted and unsubstituted porphyrins were reviewed to show how far the meso-substitution by any functional group and the insertion of metal in the porphyrins core macrocycle may affect the geometry. The analysis of twists and angles has shown two kinds of distortions: external [T(Cβ-Cα-Cmeso-Xn) and T(Cβ-Cα-Cmeso-Cα)] and internal [T(Nm-Cα-Cmeso-Xn) and T(Nn-Cα-Cmeso-Cα)] with averages of [+6°and –6°] and [–5°and +5°], respectively. In the meso-substituted case, the external and internal twists Cβ-Cα-Cmeso-X and N-Cα-Cmeso-X, respectively are oppositely orientated. Similar effect is observed in meso-unsubstituted of Cβ-Cα- Cmeso-H and N-Cα-Cmeso-H. However, the external distortions are more significant than internal. Considering the same order, the limit of distortions is [97°and 132°(–48°)] for external and [91°(–89°) and 52°] for internal. In the two cases, the substituents have opposite directions of distortions. The meso-substituted porphyrins have a high limit of twisting than usubstituted one, depending of the weight of substituents. The average of the bond angular deformations is 168°, almost planar. However, the limit of angular deformation is 94°.

Pelagic fish,including sardines and anchovies(Order Clupeiformes),are the most common species taken by artisanal marine fisheries along the Togolese *** investigated fisher involvement as well as fish captures over a period of 10 years,particularly of the European anchovy(Engraulis encrasicolus).Our results indicate that 60%of fishers operated from the Loméfishing harbour,most fishers being Ghanaians working seasonally in Togo.63.7%of all the fishers used canoes with outboards,a higher percentage compared to the previous *** fishing gear type were identified,with bottom gillnet and surface gillnet being the most commonly ***,in the most important fishing camp in the country in terms of fish production(Loméfishing harbour),all fishers used shark ***,fisheries catches did not change significantly across years,but anchovy fishing effort and catch per unit of effort(CPUE)declined over the study period,suggesting some depletion of the species stocks on the Togolese coast and a demotivation of fishers.

Nanomechanical heterogeneity is expected to have an effect on elasticity, injury and bone remodelling. In normal bone, we have two types of cells (osteoclasts and osteoblasts) working together to maintain existing bone. Bone cancers can produce factors that make the osteoclasts work harder. This means that more bone is destroyed than rebuilt, and leads to weakening of the affected bone. We report here the first demonstration of the nanoscale stiffness distribution in bone metastases before and after treatment of animals with the bisphosphonate Risedronate, a drug which is currently used for the treatment of bone metastases in patients with advanced cancers. The strategy used here is applicable to a wide class of biological tissues and may serve as a new reflection for biologically inspired scaffolds technologies.

The phylogeny of the Acarosporaceae(Lecanoromycetes,Acarosporomycetidae,Acarosporales)is investigated using data from three molecular markers;nuclear ITS-LSU rDNA,mitochondrial SSU andβ-*** is shown to be constituted by six main clades;myriospora,Timdalia,Pleopsidium,a clade composed by BAcarospora^rhizobola and BA.^terricola,the poorly supported Sarcogyne clade(including several Polysporina and Acarospora species)and the Acarospora clade(including the type of Polysporina,***,and several other Polysporina species).The common ancestor of the Acarosporaceae did not produce strongly black pigmented(carbonized or melanized)ascomata,but this trait has arisen secondarily and independently numerous times in the evolution of the *** number of changes in character states of both carbonized epihymenium and carbonized exciple are considerably more than the minimum *** genera Sarcogyne and Polysporina—largely circumscribed based on the presence of black pigmented ascomata—are shown to be distinctly *** presence of green algae in the ascoma margin(lecanorine or lecideine ascomata)may vary even within single species.
