Helicobacter pylori vac A genotype is a predominant determinant of immune response to Helicobacter pylori CagA

To evaluate the frequency of Helicobacter pylori (H. pylori) CagA antibodies in H. pylori infected subjects and to identify potential histopathological and bacterial factors related to H. pylori CagA-immune response. METHODSSystematic data to H. pylori isolates, blood samples, gastric biopsies for histological and molecular analyses were available from 99 prospectively recruited subjects. Serological profile (anti-H. pylori, anti-CagA) was correlated with H. pylori isolates (cagA, EPIYA, vacA s/m genotype), histology (Sydney classification) and mucosal interleukin-8 (IL-8) mRNA and protein expression. Selected H. pylori strains were assessed for H. pylori CagA protein expression and IL-8 induction in co-cultivation model with AGS cells. RESULTSThirty point three percent of microbiologically confirmed H. pylori infected patients were seropositive for CagA. Majority of H. pylori isolates were cagA gene positive (93.9%) with following vacA polymorphisms: 42.4% vacA s1m1, 23.2% s1m2 and 34.3% s2m2. Anti-CagA-IgG seropositivity was strongly associated with atrophic gastritis, increased mucosal inflammation according to the Sydney score, IL-8 and cagA mRNA expression. VacA s and m polymorphisms were the major determinants for positive (vacA s1m1) or negative (vacA s2m2) anti-CagA serological immune response, which also correlated with the in vitro inflammatory potential in AGS cells. In vitro co-cultivation of representative H. pylori strains with AGS cells confirmed functional CagA translocation, which showed only partial correlation with CagA seropositivity in patients, supporting vacA as major co-determinant of the immune response. CONCLUSIONSerological immune response to H. pylori cagA+ strain in H. pylori infected patients is strongly associated with vacA polymorphism, suggesting the crucial role of bacterial factors in immune and clinical phenotype of the infection.

Accurate measurement of the average plasma parameters in the edge region,including the temperature and density of electrons and ions,is critical for understanding the characteristics of the scrape-off layer(SOL) and divertor plasma transport in magnetically confined fusion *** the J-TEXT tokamak,a multi-channel retarding field analyzer(RFA) probe has been developed to study average plasma parameters in the edge region under various poloidal divertor and island divertor *** edge radial profile of the ion-to-electron temperature ratio,τ_(i/e),has been determined,which gradually decreases as the SOL ion self-collisionality,v_(SOL)*,*** is broadly consistent with what has been observed previously from various tokamak ***,the comparison of experimental results under different configurations shows that in the poloidal divertor configuration,even under the same v_(SOL)*,τ_(i/e) in the SOL region becomes smaller as the distance from the X-point to the target plate *** the island divertor configuration,τ_(i/e) near the O-point is higher than that near the X-point at the same v_(SOL)*,and both are higher than those in the limiter *** results suggest that the magnetic configuration plays a critical role in the energy distributions between electrons and ions at the plasma boundary.

Y90-radioembolization via variant hepatic arteries: Is there a relevant risk for non-target embolization?

BACKGROUND The hepatic arterial anatomy is highly variable, with the two most common variants being a replaced right hepatic artery(RHA) originating from the superior mesenteric artery(SMA) and a left hepatic artery(LHA) originating from the left gastric artery(LGA). These anatomical variants could potentially increase the risk for non-target embolization during Y90-Radioembolization due to the close proximity between hepatic and enteric vessel *** To evaluate the safety of Yttrium-90radioembolization(90Y-RE) with resin microspheres in patients with a variant hepatic arterial *** In this retrospective single-center observational study, 11 patients who underwent RE with 90Y-resin microspheres via a LHA originating from the LGA,and 13 patients via a RHA originating from the SMA were included. Patient and treatment data were reviewed regarding clinical and imaging evidence of nontarget embolization of 90Y-resin microspheres to the GI tract. Positioning of the tip of the microcatheter in relationship to the last hepatoenteric side branch was retrospectively analyzed using angiographic images, cone-beam CT and preinterventional *** None of the 24 patients developed clinical symptoms indicating a potential nontarget embolization to the GI tract within the first month after 90Y-RE. On thepostinterventional 90Y-bremsstrahlung images and/or 90Y-positron emission tomographies, no evidence of extrahepatic 90Y-activity in the GI tract was noted in any of the patients. The mean distance between the tip of the microcatheter and the last enteric side branch during delivery of the 90Y microspheres was 3.2 cm(range: 1.9-5 cm) in patients with an aberrant LHA originating from a LGA. With the growing importance of the black soldier fly(Hermetia illucens)for both sustainable food production and waste management as well as for science,a great demand of understanding its immune system ***,we present the first description of the circulating larval hemocytes with special emphasis on uptake of microorganisms and distinguishing hemocyte *** histological,zymographic,and cytometric methods and with a set of hemocyte binding lectins and antibodies,the hemocytes of *** are identified as plasmatocytes,crystal cells,and putative *** hemocyte counts(THC)are determined,and methods for THC determination are *** 1100 hemocytes per microliter hemolymph are present in naive animals,while hemocyte density decreases dramatically shortly after wounding,indicating a role of hemocytes in response to wounding(and immune response in general).The determination of the relative abundance of each hemocyte type(differential hemocyte count,DHC)revealed that plasmatocytes are highly abundant,whereas prohemocytes and crystal cells make up only a small percentage of the circulating *** are not only the most abundant but also the professional phagocytes in *** rapidly engulf and take up bacteria both in vivo and in vitro,indicating a very potent cellular defense against invading *** bioparticles such as yeasts are also removed from circulation by phagocytosis,but slower than *** is the first analysis of the potent cellular immune response in the black soldier fly,and a first toolbox that helps to identify hemocyte(types)is presented.
