The Sertoli cell tight junction （T J） is the key component of the blood-testis barrier, where it sequesters developing germ cells undergoing spermatogenesis within the seminiferous tubules. Hormonally regulated claudin-11 is a critical transmembrane protein involved in barrier function and its murine knockout results in infertility. We aimed to assess quantitatively the significance of the contribution of claudin-11 to TJ function, in vitro, using siRNA-mediated gene silencing. We also conducted an analysis of the contribution of occludin, another intrinsic transmembrane protein of the TJ. Silencing of claudin-11 and/or occludin was conducted using siRNA in an immature rat Sertoli cell culture model. Transepithelial electrical resistance was used to assess quantitatively TJ function throughout the culture. Two days after siRNA treatment, cells were fixed for immunocytochemical localization of junction proteins or lyzed for RT-PCR assessment of mRNA expression. Silencing of claudin-11, occludin, or both resulted in significant decreases in TJ function of 55% （P 〈 0.01）, 51% （P 〈 0.01）, and 62% （P 〈 0.01）, respectively. Data were concomitant with significant decreases in mRNA expression and marked reductions in the localization of targeted proteins to the Sertoli cell TJ. We provide quantitative evidence that claudin-11 contributes significantly （P 〈 0.01） to Sertoli cell TJ function in vitro. Interestingly, occludin, which is hormonally regulated but not implicated in infertility until late adulthood, is also a significant （P 〈 0.01） contributor to barrier function. Our data are consistent with in vivo studies that clearly demonstrate a role for these proteins in maintaining normal TJ barrier structure and function.

Over-fertilization has caused significant phosphorus(P)accumulation in Chinese greenhouse vegetable production(GVP)*** study,for the first time,quantified profile P accumulation directly from soil P measurements,as well as subsoil P immobilization,in three alkaline coarse-textured GVP soil profiles with 5(S5),15(S15),and 30(S30)years of cultivation in Tongshan,Southeast *** each profile,soil samples were collected at depths of 0-10(topsoil),10-20,20-40,40-60,60-80,and 80-100 *** accumulation was estimated from the difference in P contents between topsoil and parent material(60-100 cm subsoil).Phosphorus mobility was assessed from measurements of water-soluble P concentration(P_(Sol)).Finally,P sorption isotherms were produced using a batch sorption experiment and fitted using a modified Langmuir *** total P contents of 1980(S5),3190(S15),and 2330(S30)mg kg^(-1) were measured in the topsoils versus lower total P content of approximately 600 mg kg^(-1) in the 80-100 cm ***,topsoil PSol values were very high,varying from 6.4 to 17.0 mg L^(-1).The estimated annual P accumulations in the topsoils were 397(S5),212(S15),and 78(S30)kg ha^(-1) year^(-1).Sorption isotherms demonstrated the dominance of P desorption in highly P-saturated topsoils,whereas the amount of adsorbed P increased in the 80-100 cm subsoils with slightly larger P adsorption *** total P adsorption capacity of the 80-100 cm subsoils at a solution P concentration of0.5 mg L^(-1) was 15.7(S5),8.7(S15),and 6.5(S30)kg ha^(-1),demonstrating that subsoils were unable to secure P concentrations in leaching water below 0.5 mg L^(-1) because of their insufficient P-binding capacity.

Echo-enhanced ultrasound with pulse inversion imaging: A new imaging modality for the differentiation of cystic pancreatic tumours

AIm： To describe and discuss echo-enhanced sonography in the differential diagnosis of cystic pancreatic lesions. mETHODS： The pulse inversion technique （with intravenous injection of 2.4 mL SonoVue） or the power-Doppler mode under the conditions of the 2^nd harmonic imaging （with intravenous injection of 4 g Levovist） was used for echo-enhanced sonography. RESULTS： Cystadenomas frequently showed many vessels along fibrotic strands. On the other hand, cystadenocarcinomas were poorly and chaotically vascularized. ＂Young pseudocysts＂ were frequently found to have a highly vascularised wall. However, the wall of the ＂old pseudocysts＂ was poorly vascularized. Data from prospective studies demonstrated that based on these imaging criteria the sensitivities and specificities of echoenhanced sonography in the differentiation of cystic pancreatic masses were 〉 90%. CONCLUSION： Cystic pancreatic masses have a different vascularization pattern at echo-enhanced sonography. These characteristics are useful for their differential diagnosis, but histology is still the gold standard.

Three independent ethylene insensitive mutants were selected from an EmS- mutagenized population of Lotus japonicus mG-20 （miyakojima）. The mutants, called ＇Enigma＇, were mutated in the LjEIN2a gene from Lotus chromosome 1, sharing significant homology with Arabidopsis EIN2 （ethylene-insensitive2）. All three alleles showed classical ethylene insensitivity phenotypes （e.g., Triple Response）, but lacked the increased nodulation phenotype commonly associated with ethylene insensitivity. Indeed, all showed a marginal reduction in nodule number per plant, a phenotype that is enigmatic to sickle, an ethylene- insensitive EIN2 mutant in medicago truncatula. In contrast to wild type, but similar to an ETRI-1 ethylene ethylene-insensitive transgenic of L. japonicus, enigma mutants formed nodules in between the protoxylem poles, demonstrating the influence of ethylene on radial positioning. Suppression of nodule numbers by nitrate and colonisation by mycorrhizal fungi in the enigma-1 mutant were indistin- guishable from the wild-type mG-20. However, reflecting endogenous ethylene feedback, the enigma-1 mutant released more than twice the wild-type amount of ethylene, enigma-1 had a moderate reduction in growth, greater root mass （and lateral root formation）, delayed flowering and ripening, smaller pods and seeds. Expression analysis of ethylene-regulated genes, such as ETR1, NRL1 （neverripe-like 1）, and ElL3 in shoots and roots of enigma-1 and mG-20 illustrated that the ethylene-insensitive mutation strongly affected transcriptional responses in the root. These mutants open the possibility that EIN2 in L. japonicus, a determinate nodulating legume, acts in a more complex fashion possibly through the presence of a duplicated copy of LjEIN2.

Aim: A standardized preparation of seeds and shells of selected sub-species of Rosa canina L, trade name Hyben Vital, reduces discomfort from osteoarthritis of the knee and hip. This study aims to investigate the impact of the same rose-hip powder (RHP) on discomfort and the consumption of rescue medication, in patients with osteoarthritis of the hand. methods: The double blind, placebo-controlled, crossover trial included 30 patients with osteoarthritis of the dominant hand. Patients were randomly allocated to treatment with either five gram encapsulated RHP or placebo, for three months (Phase 1), after which they switched to the corresponding treatment for a further three months period (Phase 2). Before entering the study, after 3 weeks and following three months of each of the study phases, scores for pain, stiffness and general feeling of discomfort were evaluated using a 10 step categorical scale, focusing on 16 different daily activities of the hand. The consumption of rescue medication was also calculated at the beginning and at the end of each study phase. Data are based on the intention to treat. Results: At the end of Phase 1, 90% of patients in the group receiving RHP first (group A), showed a reduction in pain, as compared to 36% in the group B initially given placebo (p 0.029). In line with this observation, stiffness and the general feeling of discomfort from the disease declined during RHP treatment (p 0.032). In group A, symptom reduction was still indicated by the study subjects 3 weeks after the switch to placebo. The consumption of rescue medication such as paracetamol, codeine and tramadol also declined significantly in group A when compared to group B (p 0.013). Conclusion: The present data suggest that administration of RHP, containing seeds and shells can reduce symptoms of osteoarthritis of the hand and consumption of rescue medication.

The global trends towards improving fuel efficiency and reducing CO;emissions are the key drivers for lightweight solutions. In sheet metal processing, this can be achieved by the use of materials with a supreme strength-toweight and stiffness-to-weight ratio. Besides monolithic materials such as high-strength or light metals, in particular metal–plastic composite sheets are able to provide outstanding mechanical properties. Thus, the adaption of conventional, wellestablished forming methods for the processing of hybrid sheet metals is a current challenge for the sheet metal working industry. In this work, the planning phase for a conventional sheet metal forming process is studied aiming at the forming of metal–plastic composite sheets. The single process steps like material characterization, FE analysis, tool design and development of robust process parameters are studied in detail and adapted to the specific properties of metal–plastic composites. In material characterization, the model of the hybrid laminate needs to represent not only the mechanical properties of the individual combined materials, but also needs to reflect the behaviour of the interface zone between *** on experience, there is a strong dependency on temperature as well as strain rate. While monolithic materials show a moderate anisotropic behaviour, loads on laminates in different directions generate different strain states and completely different failure modes. During the FE analysis, thermo-mechanic and thermo-dynamic effects influence the temperature distribution within tool and work pieces and subsequently the forming behaviour. During try out and production phase,those additional influencing factors are limiting the process window even more and therefore need to be considered for the design of a robust forming process. A roadmap for sheet metal forming adjusted to metal–plastic composites is presented in this paper.

AIm: To investigate the function of NOD2 in colonic epithelial cells (CEC). mETHODS: A combination of in vivo and in vitro analyses of epithelial cell turnover in the presence and absence of a functional NOD2 protein and, in response to enteric Salmonella typhimurium infection, were used. shRNA interference was also used to investigate the consequences of knocking down NOD2 gene expression on the growth and survival of colorectal carcinoma cell lines. RESULTS:In the colonic mucosa the highest levels of NOD2 expression were in proliferating crypt epithelial cells. muramyl dipeptide (mDP), that is recognized by NOD2, promoted CEC growth in vitro . By contrast,the growth of NOD2-deficient CECs was impaired. In vivo CEC proliferation was also reduced and apoptosis increased in Nod2-/- mice, which were also evident following enteric Salmonella infection. Furthermore, neutralization of NOD2 mRNA expression in human colonic carcinoma cells by shRNA interference resulted in decreased survival due to increased levels of apoptosis. CONCLUSION: These findings are consistent with the involvement of NOD2 protein in promoting CEC growth and survival. Defects in proliferation by CECs in cases of CD may contribute to the underlying pathology of disrupted intestinal homeostasis and excessive inflammation.
