Unfolded p53 in non-neuronal cells supports bacterial etiology of Alzheimer’s disease

Alzheimer’s disease has proven to be largely intractable to treatment,despite years of research,and numerous trials of therapies that target the hallmarks of the disease-amyloid plaques and neurofibrillary *** etiology of Alzheimer’s disease remains *** is a growing body of evidence for an infectious trigger of Alzheimer’s disease,and,in particular,the focus has been on the oral pathogen Porphyromonas gingivalis(***).Reports of the expression of a misfolded form of p53 in non-neuronal cells(fibroblasts,peripheral blood mononuclear cells,and B cells)and serum,which appears several years before clinical symptoms manifest,may provide further support for the role of bacteria in general,and *** in particular,in the initiation of the *** review presents a model of the pathway from initial oral infection with *** to amyloid plaque formation and neuronal degeneration,via the steps of chronic periodontitis;secretion of the inflammagens lipopolysaccharide and gingipains into the bloodstream;induction of an inflammatory response in both peripheral cells and tissues;disruption of the blood-brain barrier,and entry into the central nervous system of the inflammagens and the *** bacteria *** this model,the misfolded p53(or“unfolded p53”;up53)is induced in non-neuronal cells and upregulated in serum as a result of oxidative stress due to lipopolysaccharide from ***53 is therefore a potential biomarker for early diagnosis of the presence of a causative agent of Alzheimer’s *** dental hygiene and aggressive antibiotic treatment may prevent the patient progressing to clinical Alzheimer’s disease if serum up53 is detected at this pre-symptomatic stage.

The epididymal β-defensins have evolved by repeated gene duplication and divergence to encode a family of proteins that provide direct protection against pathogens and also support the male reproductive tract in its primary function. Male tract defensins also facilitate recovery from pathogen attack. The β-defensins possess ancient conserved sequence and structural features widespread in multi-cellular organisms, suggesting fundamental roles in species survival. Primate SPAG11, the functional fusion of two ancestrally independent β-defensin genes, produces a large family of alternatively spliced transcripts that are expressed according to tissue-specific and species-specific constraints. The complexity of SPAG11 varies in different branches of mammalian evolution. Interactions of human SPAG11D with host proteins indicate involvement in multiple signaling pathways. （Asian J Andro12007 July; 9： 453- 462）

This paper proposes a novel approach for generating 3-dimensional complex geological facies models based on deep generative *** can reproduce a wide range of conceptual geological models while possessing the flexibility necessary to honor constraints such as well *** with existing geostatistics-based modeling methods,our approach produces realistic subsurface facies architecture in 3D using a state-of-the-art deep learning method called generative adversarial networks(GANs).GANs couple a generator with a discriminator,and each uses a deep convolutional neural *** networks are trained in an adversarial manner until the generator can create "fake" images that the discriminator cannot distinguish from "real" *** extend the original GAN approach to 3D geological modeling at the reservoir *** GANs are trained using a library of 3D facies *** the GANs have been trained,they can generate a variety of geologically realistic facies models constrained by well data *** geomodelling approach using GANs has been tested on models of both complex fluvial depositional systems and carbonate reservoirs that exhibit progradational and aggradational *** results demonstrate that this deep learning-driven modeling approach can capture more realistic facies architectures and associations than existing geostatistical modeling methods,which often fail to reproduce heterogeneous nonstationary sedimentary facies with apparent depositional trend.

Immunobiology of congenital cytomegalovirus infection of the central nervous system--the murine cytomegalovirus model

Congenital human cytomegalovirus infection is a leading infectious cause of long-term neurodevelopmental sequelae, including mental retardation and hearing defects. Strict species specificity of cytomegaloviruses has restricted the scope of studies of cytomegalovirus infection in animal models. To investigate the pathogenesis of congenital human cytomegalovirus infection, we developed a mouse cytomegalovirus model that recapitulates the major characteristics of central nervous system infection in human infants, including the route of neuroinvasion and neuropathological findings. Following intraperitoneal inoculation of newborn animals with mouse cytomegalovirus, the virus disseminates to the central nervous system during high-level viremia and replicates in the brain parenchyma, resulting in a focal but widespread, non-necrotizing encephalitis. Central nervous system infection is coupled with the recruitment of resident and peripheral immune cells as well as the expression of a large number of pro-inflammatory cytokines. Although infiltration of cellular constituents of the innate immune response characterizes the early immune response in the central nervous system, resolution of productive infection requires virus-specific CD8＋ T cells. Perinatal mouse cytomegalovirus infection results in profoundly altered postnatal development of the mouse central nervous system and long-term motor and sensory disabilities. Based on an enhanced understanding of the pathogenesis of this infection, prospects for novel intervention strategies aimed to improve the outcome of congenital human cytomegalovirus infection are proposed.
