Alzheimer’s disease has proven to be largely intractable to treatment,despite years of research,and numerous trials of therapies that target the hallmarks of the disease-amyloid plaques and neurofibrillary *** etiology of Alzheimer’s disease remains *** is a growing body of evidence for an infectious trigger of Alzheimer’s disease,and,in particular,the focus has been on the oral pathogen Porphyromonas gingivalis(***).Reports of the expression of a misfolded form of p53 in non-neuronal cells(fibroblasts,peripheral blood mononuclear cells,and B cells)and serum,which appears several years before clinical symptoms manifest,may provide further support for the role of bacteria in general,and *** in particular,in the initiation of the *** review presents a model of the pathway from initial oral infection with *** to amyloid plaque formation and neuronal degeneration,via the steps of chronic periodontitis;secretion of the inflammagens lipopolysaccharide and gingipains into the bloodstream;induction of an inflammatory response in both peripheral cells and tissues;disruption of the blood-brain barrier,and entry into the central nervous system of the inflammagens and the *** bacteria *** this model,the misfolded p53(or“unfolded p53”;up53)is induced in non-neuronal cells and upregulated in serum as a result of oxidative stress due to lipopolysaccharide from ***53 is therefore a potential biomarker for early diagnosis of the presence of a causative agent of Alzheimer’s *** dental hygiene and aggressive antibiotic treatment may prevent the patient progressing to clinical Alzheimer’s disease if serum up53 is detected at this pre-symptomatic stage.

This paper proposes a novel approach for generating 3-dimensional complex geological facies models based on deep generative *** can reproduce a wide range of conceptual geological models while possessing the flexibility necessary to honor constraints such as well *** with existing geostatistics-based modeling methods,our approach produces realistic subsurface facies architecture in 3D using a state-of-the-art deep learning method called generative adversarial networks(GANs).GANs couple a generator with a discriminator,and each uses a deep convolutional neural *** networks are trained in an adversarial manner until the generator can create "fake" images that the discriminator cannot distinguish from "real" *** extend the original GAN approach to 3D geological modeling at the reservoir *** GANs are trained using a library of 3D facies *** the GANs have been trained,they can generate a variety of geologically realistic facies models constrained by well data *** geomodelling approach using GANs has been tested on models of both complex fluvial depositional systems and carbonate reservoirs that exhibit progradational and aggradational *** results demonstrate that this deep learning-driven modeling approach can capture more realistic facies architectures and associations than existing geostatistical modeling methods,which often fail to reproduce heterogeneous nonstationary sedimentary facies with apparent depositional trend.

To evaluate the role of tissue factor (TF) and protease activated receptor (PAR)-2 in liver fibrosis. METHODSUsing CCl4 administration for eight weeks, we induced hepatic fibrosis in wild-type C57BL/6 mice and in mice with deletion of the cytoplasmic signalling domain of TF (TF§CT/§CT), deletion of PAR-2 (PAR-2-/-) and combined deletion of TF signalling domain and PAR-2 (TF§CT/§CT/PAR-2-/-). Hepatic fibrosis area was assessed by quantitative imaging of picrosirius red staining. Hepatic collagen content was assessed by hydroxyproline levels. Hepatic stellate cells (αSMA positive) and hepatic macrophages (CD68 positive) were identified by immunohistochemistry. Hepatic gene expression was determined by PCR and liver TGFβ1 content by ELISA. RESULTSCCl4 treated mice with deletion of the PAR-2 gene (PAR-2-/-) and the cytoplasmic domain of TF (TF§CT/§CT) developed significantly less hepatic fibrosis, characterised by reduced liver fibrosis area and hydroxyproline content, compared to control wildtype mice treated with CCl4. The observed reduction in histological fibrosis was accompanied by a significant decrease in the hepatic content of TGFβ, the prototypic fibrogenic cytokine, as well as fewer activated hepatic stellate cells and hepatic macrophages. Deletion of the TF cytoplasmic signalling domain reduced hepatic fibrosis to levels similar to those observed in mice lacking PAR-2 signalling but combined deletion provided no added protection against fibrosis indicating a lack of mutual modulating effects that have been observed in other contexts such as angiogenic responses. CONCLUSIONTissue factor cytoplasmic domain is involved in TF-PAR-2 signalling initiating hepatic fibrosis and is a potential therapeutic target, as its deletion would not impact coagulation.
