Canine parvovirus 2(CPV-2)is a highly contagious virus in dogs that typically causes hemorrhagic enteritis and a high mortality rate in unvaccinated *** genetic variability and antigenic diversity of CPV-2 hinder its efective prevention of infection by *** investigate the epidemiology and genetic characteristics of CPV-2 in China,rectal swabs from afected dogs were collected from diferent animal clinics in Kunshan from 2022 to *** detection and capsid gene sequencing of CPV-2 were performed using previously described primers and *** overall detection rate for CPV-2 was 16.5%(33/200).A signifcant association was found between the CPV-2-positivity and clinical signs,age,breed and vaccination *** analysis revealed the presence of CPV-2c genotypes in all positive samples,which were genetically similar to other Asian CPV-2c ***,four key mutations(A5G,F267Y,Y324I and Q370r)were detected in all isolates,and one novel mutation(I447M)was detected in three CPV-2 *** mutations in the CPV-2 strains could impact vaccine efcacy and the efectiveness of the virus immune ***,no recombination events were observed between the identifed CPV-2c strains and reference strains from *** data revealed that amino acid residues 324,426 and 440 of VP2 may under strong selection *** pattern of genetic variation in the CPV-2 lineage warrants continuous laboratory-based surveillance programs in other parts of China to better understand the pattern of seasonal distribution and association between emerging genotypes and the intensity of disease severity.

Cross-species hybridization of woodchuck hepatitis virus-induced hepatocellular carcinoma using human oligonucleotide microarrays

AIM： To demonstrate the feasibility of using woodchuck samples on human microarrays, to provide insight into pathways involving positron emission tomography （pet） imaging tracers and to identify genes that could be potential molecular imaging targets for woodchuck hepatocellular carcinoma. METHODS： Labeled crNA from woodchuck tissue samples were hybridized to Affymetrix U133 plus 2.0 GeneChips. Ten genes were selected for validation using quantitative rT-PCr and literature review was made. rESULTS： Testis enhanced gene transcript （BAX Inhibitor 1）, alpha-fetoprotein, isocitrate dehydrogenase 3 （NAD＋） beta, acetyI-CoA synthetase 2, carnitine palmitoyltransferase 2, and N-myc2 were up-regulated and spermidine/spermine N1-acetyltransferase was down-regulated in the woodchuck HCC. We also found previously published results supporting 8 of the 10 most up-regulated genes and all 10 of the 10 most downregulated genes. CONCLUSION： Many of our microarray results were validated using rT-PCr or literature search. Hence, we believe that woodchuck HCC and non-cancerous liver samples can be used on human microarrays to yield meaningful results.

通过对下载的16株小反刍兽疫病毒(PPrV)N基因片段进行序列分析,合成了PPrV N基因。用NP1和NP2引物扩增。纯化的PPrV N基因在T4DNA连接酶的作用下,和pet32-a载体进行连接反应构建重组质粒pet-a-PPrV-N。经诱导表达和纯化后,得到了大量的PPrV N蛋白。用PPrV N蛋白制备免疫血清,经国际标准血清标化后作为参考阳性血清,同时以PPrV N蛋白为抗原建立间接ELISA(I-ELISA)方法。对山东省的467份羊血清检测,使用S/P(样品值/阳性值)平均值加3倍标准差的方法,计算出其临界点为0.28。应用间接ELISA和针对H蛋白的单抗为基础的C-ELISA检测来自于疫区的69份血清,结果两者的符合率为79.7%。

DNA repair capacity correlates with standardized uptake values from 18F-fluorodeoxyglucose positron emission tomography/CT in patients with advanced non-small-cell lung cancer

Objective: The DNA repair capacity (DrC) of tumor cells is an important contributor to resistance to radiation and platinum-based drugs. Because DrC may be affected by tumor cell metabolism, we measured DrC in lymphocytes from patients with non-small-cell lung cancer (NSCLC) and compared the findings with the maximum standardized uptake value (SUVmax) on 18 F-fluorodeoxyglucose positron emission tomography (FDG pet) after (chemo)radiation therapy. Methods: This study included 151 patients with stage IA-IV NSCLC who had FDG pet at a single institution and donated blood samples before chemotherapy. We assessed the correlation of DrC, measured in peripheral T lymphocytes by a host-cell reac-tivation assay with SUVmax and their associations with overall survival (OS) time by hazards ratios calculated with a Cox pro-portional hazards regression model. results: SUVmax of the primary tumor at diagnosis was inversely associated with lymphocyte DrC (r＝－0.175, P＝0.032), particularly among patients with advanced disease (r ＝－0.218, P ＝ 0.015). However, △SUVmax of primary tumor was not significantly associated with DrC (r＝0.005, P＝0.968). SUVmax of regional lymph nodes at diagnosis (r＝－0.307, P＝0.0008) and after (chemo)radiation treatment (r＝－0.329, P＝0.034) and SUVmax of the primary tumor after (chemo)radiation treatment (r＝－0.253, P＝0.045) were also inversely associated with OS time. Conclusion: DrC was inversely associated with primary tumor SUVmax before treatment but not with △SUVmax after (chemo)radiation.

An Investigation of the required Mr Bone Attenuation Correction for Quantitative Whole-Body pet/Mr Imaging Using Clinical NaF pet/CT Studies

Tissue-classification-based attenuation correction strategies have been previously proposed to correct for bone attenuation in pet/Mr imaging and simulated using computed tomography. However, the complication of voxel averaging uniquely associated with bone has not been considered explicitly in the past. This study investigated the effect of voxel averaging between bone and soft tissue in attenuation images and determined how accurately bone must be detected in Mr images in order to perform acceptable attenuation correction of pet data by using CT-simulated attenuation correction. We found out that treating bone as soft tissue caused a mean quantification difference of -9.9% ± 5.5% in all 119 bone lesions. There were no significant differences between lesions in the pelvis and the vertebrae. The nominal difference in lesions in the ribs was significantly lower, likely due to the spatial misregistration between the emission and attenuation images. Interestingly, a non-monotonic relationship between the bone imaging ability and the absolute pet quantification accuracy was observed, with the minimal quantification difference achieved at a BVF around 40% for skull lesions (2.6% ± 2.1%), and 30% for non-skull lesions (1.4% ± 1.1%) and all lesions (1.5% ± 1.3%). This study established that a bone classification sensitivity of approximately 30% BVF is required in order for Mr-based attenuation correction methods to achieve optimal quantification in whole-body pet/Mr studies. For this purpose, higher bone imaging ability of Mr may not be necessary.

Metastatic peritoneal sarcomatosis from radiation-induced osteosarcoma in a patient with previously treated pelvic Ewing’s sarcoma: report of a case

Metastatic peritoneal sarcomatosis most commonly occurs from primary soft tissue sarcomas arising either within the abdomen or extremities. Metastatic peritoneal sarcomatosis from an osteosarcoma is extremely rare;only six cases have previously been reported. We report the first case of metastatic peritoneal sarcomatosis originating from radiation-induced osteoblastic osteosarcoma in a 22-year-old woman who had previously been treated for pelvic Ewing’s sarcoma. Abdominal computed tomography, bone scintigraphy and FDG pet/CT demonstrated extensive finely nodular disseminated peritoneal lesions. Histopathologic examination of these peritoneal lesions revealed osteosarcomatosis. In summary, we describe an unusual case of metastatic peritoneal sar comatosis from secondary osteosarcoma arising in a previously irradiated pelvicEwing’s sarcoma.

Background. The role of combined positron emission tomography and computed tomography (pet- CT) in the diagnosis of recurrent ovarian mucinous adenocarcinoma is uncertain because of previous reports that pet has limited sensitivity in the detection of mucinous neoplasms. Case. A 71- year- old white woman presented with complaints of right lower quadrant pain and a palpable adnexal mass. Physical examination and transvaginal ultrasonography revealed a 12 × 13 cm cystic mass in the left side of the pelvis. Exploratory laparotomy, optimal tumor- reductive surgery, total abdominal hysterectomy, bilateral salpingo- oophorectomy, appendectomy, and omentectomy were performed. The diagnosis was FIGO stage IIIC welldifferentiated ovarian mucinous adenocarcinoma. The patient was treated with 6 cycles of carboplatin and paclitaxel. Four months after completing chemotherapy, the patient was noted to have an elevated serum CA125 level (72 U/mL), being otherwise asymptomatic. Findings on abdominal and pelvic CT were compatible with postsurgical changes. pet-CT was performed and revealed increased metabolism along the posterior aspect of the right rectus abdominis muscle and abutting the anterior wall of an adjacent loop of bowel. Conclusion. pet-CT may identify clinically occult recurrent ovarian mucinous adenocarcinoma.
