Nitric oxide(NO)is an essential reactive oxygen species and a signal molecule in plants.Although several studies have proposed the occurrence of oxidative NO production,only reductive routes for NO production,such as the nitrate(NO_(3)^(-))-upper-reductase pathway,have been evidenced to date in land plants.However,plants grown axenically with ammonium as the sole source of nitrogen exhibit contents of nitrite and NO3−,evidencing the existence of a metabolic pathway for oxidative production of NO.We hypothesized that oximes,such as indole-3-acetaldoxime(IAOx),a precursor to indole-3-acetic acid,are intermediate oxidation products in NO synthesis.We detected the production of NO from IAOx and other oximes catalyzed by peroxidase(POD)enzyme using both 4-amino-5-methylamino-2′,7′-difluorescein fluorescence and chemiluminescence.flavins stimulated the reaction,while superoxide dismutase inhibited it.Interestingly,mouse NO synthase can also use IAOx to produce NO at a lower rate than POD.We provided a full mechanism for POD-dependent NO production from IAOx consistent with the experimental data and supported by density functional theory calculations.We showed that the addition of IAOx to extracts from Medicago truncatula increased the in vitro production of NO,while in vivo supplementation of IAOx and other oximes increased the number of lateral roots,as shown for NO donors,and a more than 10-fold increase in IAOx dehydratase expression.furthermore,we found that in vivo supplementation of IAOx increased NO production in Arabidopsis thaliana wild-type plants,while prx33-34 mutant plants,defective in POD33-34,had reduced production.Our data show that the release of NO by IAOx,as well as its auxinic effect,explain the superroot phenotype.Collectively,our study reveals that plants produce NO utilizing diverse molecules such as oximes,POD,and flavins,which are widely distributed in the plant kingdom,thus introducing a long-awaited oxidative pathway to NO production in plants.This knowledge has essential implications for und

Despite ample literature,the influence of the individual soil properties and covers on the hydrological response of burned soils of forests has not clearly identified.A clear understanding of the surface runoff and erosion rates altered by wildfires and prescribed fires is beneficial to identify the most suitable post-fire treatment This study has carried out a combined analysis of the hydrological response of soil and its driving factors in burned forests of Central-Eastern Spain.The pine stands of these forests were subjected to both prescribed fire and wildfire,and,in the latter case,to post-fire treatment with mulching.Moreover,simple multi-regression models are proposed to predict runoff and erosion in the experi-mental conditions.In the case of the prescribed burning,the fire had a limited impact on runoff and erosion compared to the unburned areas,due to the limited changes in soil parameters.In contrast,the wildfire increased many-fold the runoff and erosion rates,but the mulching reduced the hydrological response of the burned soils,particularly for the first two-three rainfalls after the fire.The increase in runoff and erosion after the wildfire was associated to the removal of the vegetation cover,soil water repellency,and ash left by fire;the changes in water infiltration played a minor role on runoff and erosion.The multi-regression models developed for the prescribed fire were accurate to predict the post-fire runoff coefficients.However,these models were less reliable for predictions of the mean erosion rates.The predictions of erosion after wildfire and mulching were excellent,while those of runoff were not satisfactory(except for the mean values).These results are useful to better understand the relations among the hydrological effects of fire on one side and the main soil properties and covers on the other side.Moreover,the proposed prediction models are useful to support the planning activities of forest managers and hydrologists towards a more effective conservation of forest soils.

The detection of genes that show similar profiles under different experimental conditions is often an initial step in inferring the biological significance of such genes. Visualization tools are used to identify genes with similar profiles in microarray studies. Given the large number of genes recorded in microarray experiments, gene expression data are generally displayed on a low dimensional plot, based on linear methods. However, microarray data show nonlinearity, due to high-order terms of interaction between genes, so alternative approaches, such as kernel methods, may be more appropriate. We introduce a technique that combines kernel principal component analysis （KPCA） and Biplot to visualize gene expression profiles. Our approach relies on the singular value decomposition of the input matrix and incorporates an additional step that involves KPCA. The main properties of our method are the extraction of nonlinear features and the preservation of the input variables （genes） in the output display. We apply this algorithm to colon tumor, leukemia and lymphoma datasets. Our approach reveals the underlying structure of the gene expression profiles and provides a more intuitive understanding of the gene and sample association.

The exponential growth of mobile applications and services during the last years has challenged the existing network infrastructures.Consequently,the arrival of multiple management solutions to cope with this explosion along the end-to-end network chain has increased the complexity in the coordinated orchestration of different segments composing the whole infrastructure.The Zero-touch Network and Service Management(ZSM)concept has recently emerged to automatically orchestrate and manage network resources while assuring the Quality of Experience(QoE)demanded by users.Machine Learning(ML)is one of the key enabling technologies that many ZSM frameworks are adopting to bring intelligent decision making to the network management system.This paper presents a comprehensive survey of the state-of-the-art application of ML-based techniques to improve ZSM performance.To this end,the main related standardization activities and the aligned international projects and research efforts are deeply examined.from this dissection,the skyrocketing growth of the ZSM paradigm can be observed.Concretely,different standardization bodies have already designed reference architectures to set the foundations of novel automatic network management functions and resource orchestration.Aligned with these advances,diverse ML techniques are being currently exploited to build further ZSM developments in different aspects,including multi-tenancy management,traffic monitoring,and architecture coordination,among others.However,different challenges,such as the complexity,scalability,and security of ML mechanisms,are also identified,and future research guidelines are provided to accomplish a firm development of the ZSM ecosystem.

It is increasingly common to find alarming news related to tragic events occurring in schools around the world.Being able to deal with these situations without getting hurt is a task not suitable for everyone.In general,teachers are the ones who must deal with this type of situation in addition to other daily problems that appear in any classroom that make the level of stress to which they are subjected can become dangerous.This research aims to know the current situation of teachers in a region of southeastern Spain in terms of their level of work stress,resilience,and other associated variables,for which an ex post facto quantitative approach study was designed.for this purpose,an ad hoc sociodemographic questionnaire,and questions based on the Teaching and Learning International Survey of the Organization for Economic Co-Operation and Development,the Maslach Burnout Inventory,and the Brief Resilient Coping Scale were applied.The participating sample consisted of 470 teachers.The main findings were that 6%of teachers had burnout(high emotional exhaustion,high cynicism,and low professional effectiveness simultaneously).Resilience correlated inversely with stress,emotional exhaustion,cynicism,excessive teaching,and difficulty maintaining classroom discipline.In conclusion,knowing the needs of teachers allows us to reflect on what kind of prevention and intervention programs are necessary to improve the well-being of teachers and thus improve the quality of education.

Heusler compounds attract a great deal of attention from researchers thanks to a wealth of interesting properties,among which is superconductivity.Here we perform an extensive study of the superconducting and elastic properties of the cubic(full-)Heusler family using a mixture of ab initio methods,as well as interpretable and predictive machine-learning models.By analyzing the statistical distributions of these properties and comparing them to anti-perovskites,we recognize universal behaviors that should be common to all conventional superconductors while others turn out to be specific to the material family.In total,we discover a total of eight hypothetical materials with critical temperatures above 10 K to be compared with the current record of T_(c)=4.7 K in this family.furthermore,we expect most of these materials to be highly ductile,making them potential candidates for the manufacture of wires and tapes for superconducting magnets.

fecal microbiota transplantation(fMT)is a successful method for treating recurrent Clostridioides difficile(C.difficile)infection(rCDI)with around 90%efficacy.Due to the relative simplicity of this approach,it is being widely used and currently,thousands of patients have been treated with fMT worldwide.Nonetheless,the mechanisms underlying its effects are just beginning to be understood.Data indicate that fMT effectiveness is due to a combination of microbiological direct mechanisms against C.difficile,but also through indirect mechanisms including the production of microbiota-derived metabolites as secondary bile acids and short chain fatty acids.Moreover,the modulation of the strong inflammatory response triggered by C.difficile after fMT seems to rely on a pivotal role of regulatory T cells,which would be responsible for the reduction of several cells and soluble inflammatory mediators,ensuing normalization of the intestinal mucosal immune system.In this minireview,we analyze recent advances in these immunological aspects associated with the efficacy of fMT.

Obesity is associated with several diseases,including mental health.Adipose tissue is distributed around the internal organs,acting in the regulation of metabolism by storing and releasing fatty acids and adipokine in the tissues.Excessive nutritional intake results in hypertrophy and proliferation of adipocytes,leading to local hypoxia in adipose tissue and changes in these adipokine releases.This leads to the recruitment of immune cells to adipose tissue and the release of pro-inflammatory cytokines.The presence of high levels of free fatty acids and inflammatory molecules interfere with intracellular insulin signaling,which can generate a neuroinflammatory process.In this review,we provide an up-to-date discussion of how excessive obesity can lead to possible cognitive dysfunction.We also address the idea that obesity-associated systemic inflammation leads to neuroinflammation in the brain,particularly the hypothalamus and hippocampus,and that this is partially responsible for these negative cognitive outcomes.In addition,we discuss some clinical models and animal studies for obesity and clarify the mechanism of action of anti-obesity drugs in the central nervous system.
