Huntington's disease(HD) is an inherited autosomal dominant neurodegenerative disorder characterized by the development of adult-onset motor dysfunction,psychiatric disturbances and intellectual *** is associated with an expansion of CAG repeat sequence in the huntingtin gene(Htt).Exon 1 of Htt normally contains between 6 to 35 CAG repeats,whereas in patients affected with HD it contains more than 40 trinucleotides.

Rural areas in the K?odzko Region were subject to a long-term depopulation,marginalization,and long-lasting socio-economic decline since the 19th *** recent years,however,the study area has been subject to a shift in socio-economic trends that show rural *** main goal of the research is to assess the actual role of tourism in socio-economic revival of rural areas in the K?odzko *** determine the dynamics of the tourism function development,statistical data were used and an inventory of tourism beds in accommodation facilities was *** results show that there was a substantial development of the tourism function in the study area and its most intense scale was noted in the depopulating villages,as evidenced by the increase in the numbers of tourism facilities and numbers of tourism ***,it can be concluded that the development of tourism contributed to the limitation of negative socioeconomic processes in the area and,in the case of some villages,even to the reversal of these trends,which might testify to the rural ***,the improvement of the socio-economic situation is visible also in the villages that do not have significant tourist values but are in the neighbourhood of tourism *** observation might be important for forecasting future development and planning of rural areas.

Solid phase microextraction(SPME)in combination with high-resolution mass spectrometry was employed for the determination of metabolomic profile of mouse melanoma growth within in vitro 2D,in vitro 3D,and in vivo *** multi-model approach had never been investigated *** to the low-invasiveness of SPME,it was possible to perform time-course analysis,which allowed building time profile of biochemical reactions in the studied *** approach does not require the multiplication of samples as subsequent analyses are performed from the very same cell culture or from the same *** already reduces the number of animals required for experiment;therefore,it is with good concordance with the 3Rs rule(replacement,reduction,and refinement).Among tested models,the largest number of compounds was found within the in vitro 2D cell culture model,while in vivo and in vitro 3D models had the lowest number of detected *** results may be connected with a higher metabolic rate,as well as lower integrity of the in vitro 2D model compared to the in vitro 3D model resulting in a lower number of compounds released into medium in the latter *** terms of in vitro-in vivo extrapolation,the in vitro 2D model performed more similar to in vivo model compared to in vitro 3D model;however,it might have been due to the fact that only compounds secreted to medium were ***,in further experiments to obtain full metabolome information,the intraspheroidal assessment or spheroid dissociation would be necessary.

Objective:To analyze the effects of Ageratum conyzoides *** the monosodium iodoacetate induced osteoarthritis ***:Thin layer chromatography was performed to analyze the constituents of the babandotan extract *** male Sprague-Dawley rats used in this study were divided into 6 groups:normal control and negative control groups,both given0.5%carboxymethyl cellulose;the positive control group that was given glucosamine and chondroitin suspension(486 mg/200 g B.W.);the 3 dose variation extract groups including dose 1,2,and 3 that were given 40,80,and 160 mg/200 g *** on day 29 *** the groups were induced with 0.05 mL monosodium iodoacetate(20 mg/mL)on day1,except normal control induced by *** of edema volume of rat knees was performed on day 0,8,15,22,29,43,and *** data was measured at day 1,29and *** was collected at day 50 to evaluate TNF-α and MMP-9 by *** histopathology was evaluated by staining with H&E and Safranin-o-fast green staining on day ***:The babandotan leaves extract dose 2(80 mg/200 g B.W.)and dose 3(160mg/200 g B.W.)could decrease the edema volume,increase the area and thickness of articular cartilage,and increase proteoglycan ***,dose 3(160 mg/200 g B.W.)of extract babandotan leaves were able to significantly decrease the number of leukocytes,lymphocytes and udem volume,and decrease TNF alpha and MMP-9 ***:Babandotan leaves extract can recover inflammation and cartilages degradation by inhibiting TNF-αin inflammation processes and MMP-9 in the collagenase reaction in the cartilages.

Composition of inflammatory infiltrate and its correlation with HBV/HCV antigen expression

AIM： To study the composition of liver inflammatory infiltrate in biopsy material from patients chronically infected with hepatotropic viruses and to evaluate the correlation of inflammatory infiltrate with hepatitis B virus （HBV） and hepatitis C virus （HCV） viral antigen expression in chronic B and C hepatitis. METHODS： The phenotype of inflammatory cells was evaluated by the EnVision system, using a panel of monoclonal antibodies. HBV and HCV antigens were detected with the use of monoclonal anti-HBs, polyclonal anti-HBc and anti-HCV antibodies, respectively. RESULTS： The cellular composition of liver inflammatory infiltrate was similar in the patients with B and C hepatitis：～50%-60% of cells were T helper lymphocytes. Approximately 25% were T cytotoxic lymphocytes; B lymphocytes comprised 15% of inflammatory infiltrate; other cells, including NK, totalled 10%. Expression of HLA antigens paralleled inflammatory activity. Portal lymphadenoplasia was found more often in hepatitis C （54.5%） than in hepatitis B （30.6%）. Expression of HB-cAg was found more often in chronic B hepatitis of moderate or severe activity. Overall inflammatory activity in HBV-infected cases did not correlate with the intensity of HBsAg expression in hepatocytes. Inflammatory infiltrates accompanied the focal expression of HCV antigens. A direct correlation between antigen expression and inflammatory reaction in situ was noted more often in hepatitis C than B. CONCLUSION： Irrespective of the etiology and activity of hepatitis, components of the inflammatory infiltrate in liver were similar. Overall inflammatory activity did not correlate with the expression of HBsAg and HCVAg; HBcAg expression, however, accompanied chronic hepatitis 8 of moderate and severe activity.

Proline-, glutamic acid-, and leucine-rich protein 1 （PELP1） is a scaffolding protein involved in both genomic and nongenomic estrogen signal transduction pathways. To date, the role of PELP1 protein has yet to be characterized in human sperm and has not been associated with sperm parameters. To confirm the presence of PELP1 in human sperm, fresh semen samples were obtained from 178 donors. The study was designed to establish both mRNA and protein presence, and protein cellular localization. Additionally, the number of PELP1-positive spermatozoa was analyzed in men with normal and abnormal semen parameters. Sperm parameters were assessed according to the World Health Organization （WHO） 2010 standards. The presence of PELP1 in spermatozoa was investigated using four precise, independent techniques. The qualitative presence of transcripts and protein was assessed using reverse transcription-polymerase chain reaction （RT-PCR） and western blot protocols, respectively. The cellular localization of PELP1 was investigated by immunocytochemistry. Quantitative analysis of PELP1-positive cells was done by flow cytometry. PELP1 mRNA and protein was confirmed in spermatozoa. Immunocytochemical analysis identified the presence of PELP1 in the midpieces of human sperm irrespective of sperm parameters. Becton Dickinson fluorescence-activated cell sorting （FACSCalibur^Tm） analysis revealed a significantly lower number of PELP1-positive cells in males with normal semen parameters versus abnormal samples （42.78% ± 11.77% vs 61.05% ± 21.70%, respectively; P = 0.014）. The assessment of PELP1 may be a time-saving method used to obtain information about sperm quality. The results of our study suggest that PEPL1 may be utilized as an indicator of sperm quality; thereby, PELP1 may be an additional biomarker useful in the evaluation of male infertility.
