Effect of tim.-of-day and day-of-the-week on congestion duration and breakdown:A case study at a bottleneck in Salem.NH

This work uses regression m.dels to analyze two characteristics of recurrent congestion： breakdown, the transition from.freely flowing conditions to a congested state, and duration, the tim. between the onset and clearance of recurrent congestion. First, we apply a binary logistic regression m.del where a continuous m.asurem.nt for traffic flow and a dichoto- m.us categorical variable for tim.-of-day （Am. or Pm.rush hours） is used to predict the probability of breakdown. Second, we apply an ordinary least squares regression m.del where categorical variables for tim.-of-day （Am. or Pm.rush hours） and day-of-the-week （m.nday-Thursday or Friday） are used to predict recurrent congestion duration. m.dels are fitted to data collected from.a bottleneck on 1-93 in Salem. NH, over a period of 9 m.nths. Results from.the breakdown m.del, predict probabilities of recurrent congestion, are consistent with observed traffic and illustrate an upshift in breakdown probabilities between the Am. and Pm.rush periods. Results from.the regression m.del for congestion duration reveal the presences of significant interaction between tim.-of-day and day-of-the-week. Thus, the effect of tim.-of-day on congestion duration depends on the day-of-the-week. This work provides a sim.lification of recurrent congestion and recovery, very noisy processes. Sim.lification, conveying com.lex relationships with sim.le statistical sum.aries-facts, is a practical and powerful tool for traffic adm.nistrators to use in the decision-m.king process.

Binding specificity and affinity analysis of an anti-protective antigen peptide reagent using capillary electrophoresis

Peptide biosensor reagents are em.rging as an alternative to typical antibody-based detection m.thods. Peptides can be rapidly isolated using bacterial display m.thods for new and em.rging biothreats and can be chem.cally synthesized for rapid, large-scale production. With the em.rgence of peptide biosensor reagents, there is a growing need to develop m.thods for characterizing binding interactions. Capillary electrophoresis (CE) is a free-solution separation m.thod that is able to determ.ne target and analyte binding association (Kb) and dissociation constants (Kd). In this study, the Kb, Kd, and peptide specificity of an isolated peptide binding reagent to protective antigen (PA) of Bacillus anthracis were evaluated using capillary electrophoresis at 10 and 20 kV. The relative binding specificity was rapidly assessed by m.asuring the peptide relative m.bility shift at 20 kV at nonequilibrium.using bovine serum.album.n (BSA), horseradish peroxidase (HRP), and an anti-PA m.noclonal antibody (m.b). The αPA peptide was shown to be highly specific for PA, with a Kd = 177 nm.m.asured at 20 kV and Kd = 312 nm.m.asured at 10 kV. These results show that peptides from.bacterial display libraries can be rapidly tested for specificity and binding affinity, in solution, for use as a potential biosensor reagent against new and em.rging biothreats.

Is the whole greater than the sum.of the parts? Self-rated health and transdisciplinarity

Self-rated health (SRH)—a person’s subjective evaluation of his general health—is a m.re valid and powerful predictor of m.rbidity and m.rtality than any other com.ination of objective and self-reported m.asures. However, current theoretical fram.works fail to explain this association. Here, we sought to investigate SRH in relation to health outcom.s from.a transdisciplinary perspective. Using a selective review of epidem.ological, clinical and qualitative SRH literature, we analyzed the relationships between this global subjective self-perception of health (the whole) and its directly m.asurable constituents (the parts). Although SRH often predicts m.jor health outcom.s, its underpinnings vary from.person to person. Factors influencing individual’s health interact in com.lex ways evade reductionist m.thods assessing the parts, and m.y be best captured by global self-perceptions of health. The study of SRH from.a transdisciplinary perspective exem.lifies the notion that the whole is greater than the sum.of its parts. Insight into individual’s experience of “health”, their association with physiological processes, and im.act on the health/disease continuum.m.y contribute to the developm.nt of individualized strategies for health care and prom.tion with aging. In particular, this should be m.st valuable for addressing non-com.unicable health conditions where cross-talk between health dom.ins (biological, psychological, social, behavioral, spiritual) m.y significantly contribute to pathophysiology.

Polyam.nes are sm.ll aliphatic am.nes found in alm.st all organism.,ranging from.bacteria to plants and *** m.st plants,putrescine,the m.tabolic precursor for longer polyam.nes,such as sperm.dine and sperm.ne,is produced from.arginine,with either agm.tine or ornithine as *** we show that Arabidopsis thaliana(Arabidopsis)arginine de-carboxylase 1(ADC1),one of the two known arginine decarboxylases in Arabidopsis,not only synthesizes agm.tine from.arginine,but also converts *** to *** analyses indicate that duplication and neofunctionalization of ADC1 and NATA1,the enzym.s that synthesize Nδ-acetylorm.thine in Arabidopsis,co-occur in a sm.ll num.er of related species in the *** ADC2,which is localized in the chloroplasts,ADC1 is in the endoplasm.c reticulum.together with NATA1,an indication that these two enzym.s have access to the sam. substrate ***,these results are con-sistent with a m.del whereby NATA1 and ADC1 to-gether provide a pathway for the synthesis of N-acetylputrescine in Arabidopsis.

Effect of processing routes on m.crostructure and m.chanical properties of 2014 Al alloy processed by equal channel angular pressing

Al-Cu alloy was deform.d through equal channel angular pressing(ECAP) by routes A,Ba,Bc and C up to 5 *** was done using a 90° die for three different conditions,nam.ly 1) as received,2) solutionised at 768 K for 1 h and 3) solutionised at 768 K for 1 h + aged at 468 K for 5 *** m.crostructure,m.crohardness and tensile strength were studied for all the three conditions and four *** im.rovem.nt in hardness(HV 184 after five passes) and strength(602 m.a after three passes) was observed in solutionised and aged 2014 Al alloy deform.d through route *** evolution was reasonably equiaxed in route Bc with aspect ratio of *** and aged 2014 Al alloy deform.d through route Bc was identified to have better m.crostructure and m.chanical property than the other processing routes and conditions.

Im.act of Expressing p-Coum.ryl Transferase in m.dicago sativa L. on Cell Wall Chem.stry and Digestibility

The addition of p-coum.ric acid (pCA) to lignin m.lecules is frequently found in m.m.ers of the grass fam.ly. The role of this addition is not clearly understood, but is thought to potentially aid in the form.tion of syringyl-type lignin. This is because the incorporation is as a conjugate of pCA ester linked to sinapyl alcohol, a m.jor com.onent of lignin. The forage legum. alfalfa (m.dicago sativa L.) does not contain appreciable levels of pCA in its m.re heavily lignified stem.tissues. The m.ize p-coum.ryltransferase (pCAT) gene was used to transform.alfalfa to determ.ne its im.act upon lignin com.osition and its potential to alter cell wall digestibility. A constitutive expression vector using the cassava vein m.saic virus (CsVm.) prom.ter was used to drive expression of m.ize pCAT in alfalfa. Expression of the pCAT transgene was detected in both leaves and stem.. Though there was a range of pCAconcentration in transform.d alfalfa stem. (0.2 - 1.79 m.crogram. (μg)), this was a clear increase over bound pCA in control stem. (0.15 - 0.2 m.an = 0.17 m.crogram. (μg)). This did not lead to consistent responses concerning total lignin in the stem.tissues. Leaf tissue, on the other hand, already has a relatively high level of pCA (0.85 - 1.2, m.an = 0.99 m.crogram. (μg)) and those expressing pCAT gene showed on average a sm.ll increase, but there is a wide range of values am.ng the transform.nts (0.38 - 1.55, m.an = 1.06 m.crogram. (μg)). Lignin in leaves did not appear to be significantly im.acted. However, incorporation of pCA into the wall appears to cause a shift in lignin com.osition. Testing the pCAT expressing stem.cell walls for digestibility using a rum.n in vitro system.showed there was no change in the digestibility of the stem.com.ared to em.ty vectors and control alfalfa stem.. Although expression of pCAT gene in alfalfa changes the am.unt of wall bound pCA, it does not appear to change lignin levels or im.act digestibility.

Nurses work within a m.ltidim.nsional scope of practice, which includes a reactive approach of treating health problem., a proactive stance of health prom.tion, and disease prevention. The purpose of this paper is to discuss the contribution of the intersectionality fram.work in building nursing knowledge related to the Pm.CT of HIV and show how the intersectionality fram.work can contribute to the im.rovem.nt of nursing practice, policy, and research related to Pm.CT of HIV. A com.rehensive literature review was conducted to be able to discuss the contribution of intersectionality fram.work in nursing knowledge. Intersectionality is a nice perspective that gives insight into different identities that link and work together to provide inequalities. Intersectionality is a well-suited approach in a nursing profession where we strive to offer efficient and better care to our patients or clients;an excellent tool to explore m.re on the roots of inequality, oppression, and discrim.nation.
