Antimalarial activity of the aqueous extract of Euphorbia cordifolia Elliot in Plasmodium berghei-infected mice

Objective:To evaluate the antimalarial activity of the aqueous extract of Euphorbia(E.)cordifolia Elliot against Plasmodium(P.)berghei-infected ***:Thirty healthy Swiss mice were intraperitoneally inoculated with 200μL of *** parasitized-erythrocytes and divided into five groups,and then daily treated for 5 d with single dose of 10 mL/kg of distilled water for malaria control,10 mg/kg of chloroquine for the chloroquine control and 100,200 and 400 mg/kg of the aqueous extract of *** for the three test *** was monitored by *** the end of the treatment,animals were sacrificed,and blood was collected for haematological and biochemical *** were collected for biochemical and histopathological *** significance(Pvaluated by analysis of variance followed by the Tukey post-test using Graphpad prism ***:*** extract decreased the parasite load to 2.46%,with an effective dose(ED50)of 113.07 mg/kg compared to the malaria group where the parasite load increased to(46.46±10.28)%.*** extract prevented hypoglycaemia,anaemia,leucocytosis and thrombocytopenia,attenuated the increase of transaminases activities,bilirubin and creatinine rate,and improved catalase and superoxide dismutase activities,while reducing malondialdehyde contents in the liver and *** extract significantly prevented histological damages observed in the malaria control *** acute toxicity sign was observed in mice with plant extract at the dose up to 5000 mg/***:*** extract at 200 and 400 mg/kg showed significant antimalarial *** results support its traditional use in the treatment of malaria.

Antibacterial Screening of Fifteen Cameroonian Medicinal Plants against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and Salmonella typhi

Multi-drug resistant microbial infections caused by Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa and Salmonella typhi represent a critical public health issue worldwide. Plant-derived agents are credible sources for search and development of alternative antimicrobials to fight these infections. This study describes the in-vitro antibacterial activity of plants extracts from 15 medicinal plants of the Cameroonian pharmacopeia against Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Salmonella typhi. Plants selected based on their ethnopharmacological uses were extracted by either maceration or decoction using distilled water, ethanol, methanol and ethylacetate. The afforded extracts were phytochemically screened for bioactive secondary metabolites. The microdilution method was used to assess the antibacterial activity. The two promising extracts were tested in combination using the cbeckboard technique. Eighty one extracts showed antibacterial activity with eight having minimum inhibitory concentrations below 2.5 mg/mL. results from combination assays with extracts from Terminalia catappa and Gnidia glauca leaves showed synergistic effect against Escherichia coli, Pseudomonas aeruginosa and Salmonella typhi and additive effect against Staphylococcus aureus. These extracts mainly contained saponins, tannins, glucosides, phenols, triterpenes and anthraquinones. These results support the ethnobotanical claims and indicate further directions for the investigation of plants extracts to develop alternative drugs against multi-resistant bacteria.

Two new hydralazine hydrochloride-derived Schiff bases: (E)-1-(Phthalazin- 1-yl)-1-[(Pyridin-2-yl)Ethylidene]Hydralazine (PPEH), and 1-[2-(1-(pyridine- 3-yl)ethylidene)hydrazinyl]phthalazine (PEHP), were synthesized and partially characterized by spectroscopic and crystallographic methods including IR and X-ray. The single-crystal X-ray diffraction (SCXRD) analysis of PEHP indicates that the hydralazine moiety of both ligands possesses the exocyclic C=N bond. Both, PPEH and PEHP were tested as antimicrobials and antiparasites. Just PEHP could be considered as slightly antiplasmodial and antibacterial agent. In effect, PPEH showed low antimicrobial activity against one bacterial strain with Minimum Inhibitory Concentration (MIC) value of 250 μg/ml while PEHP showed very interesting activity against 18 out of 19 bacterial strains with MIC of 31.25 - 250 μg/ml compared to the standard drug, amoxicillin. PPEH and PEHP showed higher reducing activity on ferric ions compared to Vitamin C. Two novel Schiff bases, 3-[1-(2-(phthalazin-1-yl)hydrazono)ethyl)-1,3-oxa- zinane (PHEO) and 2-[(2-(phthalazin-1-yl)hydrazono)methyl]phenol (PHMP), derived from hydralazine hydrochloride, an effective drug against hypertension, were synthesized and characterized by spectroscopic methods, Infrared (IR), Proton Nuclear Magnetic resonance (1H NMR) and Carbon-13 Nuclear Magnetic resonance (13C NMR). PHEO showed low antimicrobial activity against one bacterial strain with MIC value of 250 μg/ml while PHMP showed interesting activity against 4 bacterial strains with MIC of 31.25 - 250 μg/ml compared to the standard drug, amoxicillin. PHEO and PHMP showed higher reducing activity on ferric ions compared to Vitamin C. On lipid peroxidation, PHEO showed higher inhibition while PHMP showed lower inhibition compared to Vitamin C. Both compounds presented lower stimulating effect and lower catalase activity compared to the standard Vitamin C. PHEO and PHMP showed less than 80% inhibition in the preliminary antiplasmodial assay and so were not considered for the dose-response studies.
