Re-folded structure of syn-orogenic granitoids(Padrón dome, nW Iberia):Assessing rheological evolution of cooling continental crust in a collisional setting

This contribution discusses about the rheological, kinematic and dynamic frameworks necessary to produce recumbent and upright folds from syn-orogenic granitic massifs that were formed during an early stage of magma genesis related to the onset of a migmatitic dome. Syn-kinematic granitoids occurring within the high-grade infrastructure of the Padron migmatitic dome(nW Iberia) are deformed into largescale recumbent folds(D_2) that are later affected by upright folds(D_3). Petrostructural analysis of a selected area of this dome reveals that after a period of crustal thickening(D_1), nnW-directed extensional flow gave way to recumbent folds and penetrative axial plane foliation(S_2). Superimposed subhorizontal compression resulted in upright folds(D_3). A closer view into the dynamics of the dome allows exploring the factors that may condition the nucleation of folds with contrasting geometries during progressive deformation of molten continental crust. The formation of folds affecting syn-kinematic granitoids suggests a cooling metamorphic path in migmatitic domes. Active and passive folding mechanisms require a crystallizing(cooling) magma to nucleate folds. A more competent metamorphic host inhibits fold nucleation from much less competent magmas. As it crystallizes, magma becomes more rigid(competent),and approaches viscosity values of its host. Passive folding is favored when no significant competence contrast exists between magma and host, so this folding mechanism is more likely shortly after magma genesis and emplacement. In such conditions, and under dominant subhorizontal flow accompanied by flattening(D_2),passive folding would produce isoclinal recumbent geometries. Further magma cooling introduces a shift into the rheological behavior of partially molten crust. BACKGROUnD Colorectal cancer(CRC) is the second most common cause of cancer death worldwide. It is broadly described that cyclooxygenase-2(COX-2) is mainly overexpressed in CRC but less is known regarding post-translational modifications of this enzyme that may regulate its activity, intracellular localization and stability. Since metabolic and proteomic profile analysis is essential for cancer prognosis and diagnosis, our hypothesis is that the analysis of correlations between these specific parameters and COX-2 state in tumors of a high number of CRC patients could be useful for the understanding of the basis of this cancer in *** To analyze COX-2 regulation in colorectal cancer and to perform a detailed analysis of their metabolic and proteomic *** Biopsies from both healthy and pathological colorectal tissues were taken under informed consent from patients during standard colonoscopy procedure in the University Hospital of Bellvitge(Barcelona, Spain) and Germans Trias i Pujol University Hospital(Campus Can Ruti)(Barcelona, Spain). Western blot analysis was used to determine COX-2 levels. Deglycosylation assays were performed in both cells and tumor samples incubating each sample with peptide n-glycosidase F(PnGase F). Prostaglandin E2(PGE2) levels were determined using a specific ELISA. 1 H high resolution magic angle spinning(HRMAS) analysis was performed using a Bruker AVIII 500 MHz spectrometer and proteomic analysis was performed in a nano-liquid chromatography-tandem mass spectrometer(nano LC-MS/MS) using a QExactive HF orbitrap *** Our data show that COX-2 has a differential expression profile in tumor tissue of CRC patients vs the adjacent non-tumor area, which correspond to a glycosylated and less active state of the protein. Microstructural evolution during mechanical alloying of Ag and Zn, and subsequent heat treatments were investigated. The mechanical alloying was carried out in a SPEX 8000D miller. The microstructural characterization was obtained by X-ray diffraction （XRD）, transmission electron microscopy （TEM） and scanning electron microscopy （SEM）. The thermal behavior was studied using differential scanning calorimetry （DSC）. Based on the results obtained, it can be concluded that at the early stages of milling was possible to detect the ε, β, β′, α solid solutions and remaining Zn. Later, the ε, β, β′ and Zn phases disappeared while the Zn concentration of the α solid solution was strongly increased. After 7.2 ks of milling, the mechanical alloying process reached a steady state. During this period, both the composition and crystallite size of the α solid solution remained practically unchanged. On the other hand, subsequent heat treatments of milled powders showed that the α solid solution could also be obtained by the combination of mechanical alloying and heat treatment. Finally, the evolution of the microstructure during milling and annealing was combined to propose an optimal processing route in order to obtain a α solid solution.

Carbapenemases: A worldwide threat to antimicrobial therapy

Carbapenems are potent β-lactams with activity against extended-spectrum cephalosporinases and β-lactamases. These antibiotics, derived from thienamycn, a carbapenem produced by the environmental bacterium Streptomyces cattleya, were initially used as last-resort treatments forsevere Gram-negative bacterial infections presenting resistance to most β-lactams but have become an empirical option in countries with high prevalence of Extended Spectrum β-lactamase-producing bacterial infections. Imipenem, the first commercially available carbapenem, was approved for clinical use in 1985. Since then, a wide variety of carbapenem-resistant bacteria has appeared, primarily Enterobacteriaceae such as escherichia coli or Klebsiella pneumoniae(K. pneumoniae), Pseudomonas aeruginosa and Acinetobacter baumannii, presenting different resistance mechanisms. The most relevant mechanism is the production of carbapenem-hydrolyzing β-lactamases, also known as carbapenemases. These enzymes also inactivate all known β-lactams, and some of these enzymes can be acquired through horizontal gene transfer. Moreover, plasmids, transposons and integrons harboring these genes typically carry other resistance determinants, rendering the recipient bacteria resistant to almost all currently used antimicrobials, as is the case for K. pneumoniae carbapenemase- or new Delhi metallo-β-lactamases-type enzymes. The recent advent of these enzymes in the health landscape presents a serious challenge. First, the emergence of carbapenemases limits the currently available treatment options; second, these enzymes pose a risk to patients, as some studies have demonstrated high mortality associated with carbapenemase-producing bacterial infections; and third, these circumstances require an extra cost to sanitary systems, which are particularly cumbersome in developing countries. We previously showed that the increase in nitric oxide (nO) levels and nO synthase (nOS) expression correlate with the progression of reactive astrocytosis and demyelination in the brains of 6-month-old taiep rats. Increased levels of nO can result in high concentration of peroxynitrite and thus cause tissue damage, which consists of lipoperoxidation of the cytoplasmic membrane, such as the myelin, and of apoptotic and necrotic cell-death. On this basis, we studied whether the increased nO production is associated with lipoperoxidation and cell death in the cerebellum and brainstem over the age (1, 3, 6, and 8 months) of taiep rats. The results were compared with those obtained in matched Sprague-Dawley (SD) rats. We measured the levels of nitrites (nO production), malonyldialdehyde, and 4-hydroxyalkenal (lipoperoxidation) in brain tissue homogenates. The three nOS isoforms and cleaved caspase-3 were evaluated by using ELISA and immunostaining techniques. Our results showed that nO production and lipoperoxidation increased in the cerebellum and brainstem as the age of the taiep rats increased compared to SD rats. The overexpression of nnOS and inOS were in the Purkinje cells, magnocellular neurons, and in oligodendrocytes, whereas the glial cells showed strong cleaved-caspase-3 immunoreactivity. In summary our results suggest that nO plays a role in the demyelination and cell death in the taiep rat.
