Filoviruses cause severe and fatal viral hemorrhagic fever in humans. Filovirus research has been extensive since the 2014 Ebola outbreak. Due to their high pathogenicity and mortality, live filoviruses require Biosafety Level-4(BSL-4) facilities, which have restricted the development of anti-filovirus vaccines and *** HIV-based pseudovirus cell infection assay is widely used for viral entry studies in BSL-2 conditions. Here,we successfully constructed nine in vitro pseudo-filovirus models covering all filovirus genera and three in vivo pseudo-filovirus-infection mouse models using Ebola virus, Marburg virus, and Lloviu virus as representative viruses. The pseudo-filovirus-infected mice showed visualizing bioluminescence in a dose-dependent manner. A bioluminescence peak in mice was reached on day 5 post-infection for Ebola virus and Marburg virus and on day4 post-infection for Lloviu virus. Two known filovirus entry inhibitors, clomiphene and toremiphene, were used to validate the model. Collectively, our study shows that all genera of filoviruses can be well-pseudotyped and are infectious in vitro. The pseudo-filovirus-infection mouse models can be used for in vivo activity evaluation of anti-filovirus drugs. This sequential in vitro and in vivo evaluation system of filovirus entry inhibitors provides a secure and efficient platform for screening and assessing anti-filovirus agents in BSL-2 facilities.

The development of spaceborne Synthetic Aperture Radar(SAR)technology declares that the golden era of SAR remote sensing in archeology is approaching;however,nowadays its methodology framework is still lacking due to the inadequate case studies validated by *** this study,we investigated the crop marks using multi-temporal Cosmo-SkyMed data acquired in 2013 by applying a twostep decision-tree classifier in conjunction with a spatial analysis in an area of archeological interest nearby the archeological site of Han-Wei capital city(1900–1500 BP),in Luoyang,*** time-series backscattering anomalies related to the wheat growth cycle were identified and then further validated in two zones by geophysical investigations(Ground Penetration Radar and electrical measurements)and in a third zone by archeological excavations made after the SAR data *** study provides a new approach for the relic detection,shallowly buried and covered by the crop vegetation,by temporal crop marks on spaceborne SAR *** also emphasize the necessity to establish a satellite-to-ground methodology framework for the promotion of remote-sensing technology in archeology.

Overlapping gravity accumulation bodies were formed on the northwestern steep slope of the Shuangyang Formation in the Moliqing fault depression of northeast *** study analyzed in detail the spatial distribution of the lithofacies and lithofacies associations of these accumulation bodies based on more than 600 m of core sections,and summarized 12 major types of lithofacies and three types of lithofacies associations：（1） the proximal zone consists of gravelly debris flows dominated by alluvial channel conglomerates;（2） the middle zone is dominated by various gravity flow deposits and traction flow deposits;and（3） the distal zone is dominated by mudstones with intercalations of sandy debris and *** with the grain size cumulative probability curves analysis,we determined the transformation of debris flows to sandy debris flows and to turbidity currents in the slope zone of the basin margin,and further proposed a lacustrine slope apron model that is characterized by（1） an inconstant multiple source（line source）,（2） an alternation of gravity flow deposits and traction flow deposits dominated by periodical changes in a source flood flow system,and（3） the transformation of sandy debris flow deposits into distal turbidity current *** sedimentary model may be applicable to other fault depressions for predicting reservoir distribution.

Composite lymphoma(CL)involving B-cell lymphoma and T-cell lymphoma is extremely ***,we report three such cases using immunohistochemistry,flow cytometry,and the next-generation sequencing(NGS)to identify the pathological and molecular characteristics of *** the first case,the patient was admitted to hospital for generalized pruritic maculopapular rash over the whole *** excisional biopsy of the skin lesions showed T-cell *** the same time,the staging bone marrow(BM)biopsy revealed a diffuse large B-cell lymphoma(DLBCL).After R-CHOP(rituximab,cyclophosphamide,doxorubicin,vincristine,and prednisone)therapies,the patient produced a good response with substantial dissipation of the rashes and relief of *** other two patients were admitted to hospital due to lymphadenopathy and were diagnosed with DLBCL and follicular lymphoma(FL)after core needle biopsy of lymph nodes,BM biopsy,BM aspiration,and flow *** R-CHOP and R-COP(rituximab,cyclophosphamide,vincristine,and prednisone)therapies,they achieved complete remission unconfirmed(CRu)and complete remission(CR).However,one or two years later,they suffered a relapse of *** shocking fact was that re-biopsy of lymphadenopathy revealed peripheral T-cell lymphoma(PTCL)and angioimmunoblastic T-cell lymphoma(AITL).NGS findings identified DNA methyltransferase 3a(DNMT3a),isocitrate dehydrogenase 2(IDH2),Ras homolog gene family,member A(RHOA),splicing factor 3B subunit 1(SF3B1),and tumor protein p53(TP53)*** immunochemotherapy,these patients achieved CRu and CR ***,they suffered a second relapse of T-cell ***,they died due to progression of *** found that the occurrence of CL is associated with Epstein-Barr virus infection and DNMT3a,IDH2,and TP53 mutations,and the prognosis of the disease is closely related to the T-cell lymphoma components.

Mitochondrial genome of Chthamalus challengeri(Crustacea:Sessilia): gene order comparison within Chthamalidae and phylogenetic consideration within Balanomorpha

Acorn barnacles are important model species in researches on intertidal ecology, larval development and biofouling. At present, with the development of mitochondrial genomics, it is helpful to understand the phylogenetic relationship from the mitogenomic level. The complete mitochondrial genome of Chthamalus challengeri was presented. The genome is a circular molecule of 15 358 bp. Compared with other species in Balanomorpha, the non-coding region is longer, while the length of the genes is similar to the other species. The overall A+T content of the mitochondrial genome of C. challengeri is 70.5%. There are variations of initiation and stop codons in the known Balanomorpha mitochondrial genomes. The C. challengeri and C. antennatus within the same genus share the identical gene arrangement. However, the gene arrangement of different genera in Chthamalidae is different, as there is a translocation between two tRNA genes and an inversion involving a large gene block. In particular, both srRNA and lrRNA of the two species in Chthamalus are encoded in the heavy strand, differing from the former Balanomorpha species. The topology and gene arrangement in Chthamalidae support each other. Phylogenetic analysis indicates that the Chthamalidae is monophyletic, while the Balanidae and Archaeobalanidae are polyphyletic.

Lipidomics reveals carnitine palmitoyltransferase 1C protects cancer cells from lipotoxicity and senescence

Lipotoxicity, caused by intracellular lipid accumulation, accelerates the degenerative process of cellular senescence, which has implications in cancer development and therapy. Previously, carnitine palmitoyltransferase 1C(CPT1C), a mitochondrial enzyme that catalyzes carnitinylation of fatty acids, was found to be a critical regulator of cancer cell senescence. However, whether loss of CPT1C could induce senescence as a result of lipotoxicity remains unknown. An LC/MS-based lipidomic analysis of PANC-1,MDA-MB-231, HCT-116 and A549 cancer cells was conducted after siRNA depletion of CPT1C. Cellular lipotoxicity was further confirmed by lipotoxicity assays. Significant changes were found in the lipidome of CPT1C-depleted cells, including major alterations in fatty acid, diacylglycerol, triacylglycerol, oxidative lipids, cardiolipin, phosphatidylglycerol, phosphatidylcholine/phosphatidylethanolamine ratio and sphingomyelin. This was coincident with changes in expressions of mRNAs involved in *** and biochemical analyses revealed higher lipid accumulation and increased malondialdehyde and reactive oxygen species, signatures of lipid peroxidation and oxidative stress. Reduction of ATP synthesis, loss of mitochondrial transmembrane potential and down-regulation of expression of mitochondriogenesis gene m RNAs indicated mitochondrial dysfunction induced by lipotoxicity, which could further result in cellular senescence. Taken together, this study demonstrated CPT1C plays a critical role in the regulation of cancer cell lipotoxicity and cell senescence, suggesting that inhibition of CPT1C may serve as a new therapeutic strategy through induction of tumor lipotoxicity and senescence.

Autophagy is the basic catabolic progress involved in cell degradation of unnecessary or dysfunctional cellular *** has been proven that autophagy could be utilized for cell survival under ***-preconditioning(HPC)could reduce apoptosis induced by ischemia and hypoxia/serum deprivation(H/SD)in bone marrow-derived mesenchymal stem cells(BMSCs).Previous studies have shown that both leptin signaling and autophagy activation were involved in the protection against apoptosis induced by various stress,including ***,it has never been fully understood how leptin was involved in the protective effects conferred by *** the present study,we demonstrated that HPC can induce autophagy in BMSCs by increased LC3-II/LC3-I ratio and autophagosome ***,similar effects were also observed when BMSCs were pretreated with *** beneficial effects offered by HPC were absent when BMSCs were incubated with autophagy inhibitor,3-methyladenine(3-MA).In addition,down-regulated leptin expression by leptin-shRNA also attenuated HPC-induced autophagy in BMSCs,which in turn was associated with increased apoptosis after exposed to sustained H/***,increased AMP-activated protein kinase phosphorylation and decreased mammalian target of rapamycin phosphorylation that were observed in HPC-treated BMSCs can also be attenuated by down-regulation of leptin *** data suggests that leptin has impact on HPC-induced autophagy in BMSCs which confers protection against apoptosis under H/SD,possibly through modulating both AMPK and mTOR pathway.

In situ synthesis and unidirectional insertion of membrane proteins in liposome-immobilized silica stationary phase for rapid preparation of microaffinity chromatography

Cell membrane affinity chromatography has been widely applied in membrane protein(MP)-targeted drug screening and interaction ***,in current methods,the MP sources are derived from cell lines or recombinant protein expression,which are time-consuming for cell culture or purification,and also difficult to ensure the purity and consistent orientation of MPs in the chromatographic stationary *** this study,a novel in situ synthesis membrane protein affinity chromatography(iSMAC)method was developed utilizing cell-free protein expression(CFE)and covalent immobilized affinity chromatography,which achieved efficient in situ synthesis and unidirectional insertion of MPs into liposomes in the stationary *** advantages of iSMAC are:1)There is no need to culture cells or prepare recombinant proteins;2)Specific and purified MPs with stable and controllable content can be obtained within 2 h;3)MPs maintain the transmembrane structure and a consistent orientation in the chromatographic stationary phase;4)The flexible and personalized construction of cDNAs makes it possible to analyze drug binding *** was successfully applied to screen PDGFRβinhibitors from Salvia miltiorrhiza and Schisandra *** columns prepared by in-situ synthesis maintain satisfactory analysis activity within 72 *** new PDGFRβinhibitors,salvianolic acid B and gomisin D,were screened out with KD values of 13.44 and 7.39μmol/L,*** vitro experiments confirmed that the two compounds decreasedα-SMA and collagen Ⅰ mRNA levels raised by TGF-βin HSC-T6 cells through regulating the phosphorylation of p38,AKT and *** vivo,Sal B could also attenuate CCl_(4)-induced liver fibrosis by downregulating PDGFRβdownstream related protein *** iSMAC method can be applied to other general MPs,and provides a practical approach for the rapid preparation of MP-immobilized or other biological solid-phase materials.
