Molecules with multiple stereocenters are widely present in biologically active natural products and pharmaceuticals. These molecules exhibit great three-dimensional structural diversity, which can affect the strength and selectivity of protein-ligand interactions[1]. Therefore, the precise synthesis of each stereoisomer is very important in medicinal chemistry. In the past 40 years, asymmetric catalysis has developed rapidly, and a variety of methods has been developed to construct chiral compounds containing single or adjacent stereocenters[2].

Atherosclerosis(AS)is characterized by impairment and apoptosis of endothelial cells,continuous systemic and focal inflammation and dysfunction of vascular smooth muscle cells,which is documented as the traditional cellular paradigm.However,the mechanisms appear much more complicated than we thought since a bulk of studies on efferocytosis,transdifferentiation and novel cell death forms such as ferroptosis,pyroptosis,and extracellular trap were reported.Discovery of novel pathological cellular landscapes provides a large number of therapeutic targets.On the other side,the unsatisfactory therapeutic effects of current treatment with lipid-lowering drugs as the cornerstone also restricts the efforts to reduce global AS burden.Stem cell-or nanoparticle-based strategies spurred a lot of attention due to the attractive therapeutic effects and minimized adverse effects.Given the complexity of pathological changes of AS,attempts to develop an almighty medicine based on single mechanisms could be theoretically challenging.In this review,the top stories in the cellular landscapes during the initiation and progression of AS and the therapies were summarized in an integrated perspective to facilitate efforts to develop a multi-targets strategy and fill the gap between mechanism research and clinical translation.The future challenges and improvements were also discussed.

Drug-induced liver injury(DILI)is a common adverse drug reaction,which can even result in liver failure[1,2].The Chinese Medical Association issued the Guidelines for the Diagnosis and Treatment of DILI based on the Roussel Uclaf Causality Assessment Method(RUCAM)in 2015[3].A previous study reported that traditional Chinese medicines(TCMs),herbal and dietary supplements,and antituberculosis drugs were the main causes of DILI in China[4].Herb-induced liver injury(HILI)refers to liver injury caused by TCMs,natural drugs,and their related preparations[5].

BACKGROUND Endoscopic retrograde cholangiopancreatography(ERCP)is an accurate diagnostic method for choledocholithiasis and treatment option for stone removal.Additionally,ursodeoxycholic acid(UDCA)can dissolve cholesterol stones and prevent their development and reappearance by lowering the cholesterol concen-tration in bile.Despite these treatment options,there are still patients who experience stone recurrence.The clinical data of 100 patients with choledochal stones who were hospitalized at the Yixing People’s Hospital and underwent ERCP for successful stone extraction between June 2020 and December 2022 were retrospectively collected.According to the post-ERCP treatment plan,100 patients were classified into UDCA(n=47)and control(n=53)groups.We aimed to assess the clinical efficacy and rate of relapse in the two patient populations.We then collected information(basic demographic data,clinical characteristics,and serum biochemical indicators)and determined the factors contributing to relapse using logistic regression analysis.Our secondary goal was to determine the effects of UDCA on liver function after ERCP.Compared to the control group,the UDCA group demonstrated a higher clinical effectiveness rate of 92.45%vs 78.72%(P<0.05).No significant differences were observed in liver function indices,including total bilirubin,direct bilirubin,gamma-glutamyl transpeptidase,alanine aminotransferase,alkaline phosphatase,and aspartate aminotransferase,between the two groups before treatment.After treatment,all liver function indices were significantly reduced.Comparing the control vs UDCA groups,the UDCA group exhibited significantly lower levels of all indices(55.39±6.53 vs 77.31±8.52,32.10±4.62 vs 45.39±5.69,142.32±14.21 vs 189.63±16.87,112.52±14.25 vs 149.36±15.36,122.61±16.00 vs 171.33±22.09,96.98±10.44 vs 121.35±11.57,respectively,all P<0.05).The stone recurrence rate was lower in the UDCA group(13.21%)in contrast with the control group(44.68%).Periampullary diverticula(OR:6.00,95%CI:1.69-21.30),maximum stone diam

Diaphorina citri is a global citrus pest.As a vector insect,it can transmit the causative agents of citrus huanglongbing,causing irreversible losses to the citrus industry.The acquisition of genomic information can provide a molecular genetic basis for effective control of D.citri.Here,the DNBSEQ™,Oxford Nanopore Technologies,and Hi-C technologies are applied to generate a high-quality chromosome-level genome of D.citri.The genome size of D.citri was 523.78 Mb with a scaffold N50 of 47.05 Mb distributed on 13 chromosomes.A total of 250.64 Mb(47.85%)repeat sequences and 24048 protein-coding genes were predicted.Genome resequencing of female and male individuals indicated that the sex chromosome system of D.citri is XO.Phylogenetic analysis demonstrated that D.citri and Pachypsylla venusta,which separated from their most recent common ancestor about 336.62 million years ago,were the most closely related.Additionally,we identified genes potentially involved in detoxification metabolism,pathogen transmission,and honeydew secretion for further investigation.The high-quality genome provides an important reference for developing effective management strategies of D.citri.

Tuning Charge Transport of Oligopeptide Junctions via Interfacial Amino Acids

Comprehensive Summary,The charge transport through peptides can imitate the corresponding processes in more complicated proteins,enabling us to develop high-performance bioelectronic devices and to understand the mechanisms of biomolecular recognition and information transfer.While charge transport modulation through individual peptides has been achieved via various covalent strategies,the intermolecular modulation is still very challenging,which may capture the charge transport between proteins.To tackle this challenge,we used well-defined self-assembled monolayers(SAMs)of oligopeptides as a model to imitate the interface of proteins and explored an interfacial amino acid strategy for charge transport modulation.We showed that non-covalently interfaced charged amino acids(e.g.,arginine)effectively attenuated the charge transport of glutamic acid terminated polyglycine peptide SAMs.By analyzing the relationship of the charge transport with the molecular frontier orbital relative to the Fermi energy level of the electrode,the molecule-electrodes coupling(Γ),and the trends in skewness and kurtosis with voltage and the dielectric constant(εr),we showed that the attenuation was from the decreasedΓand the reduced polarizability.We present an efficient strategy to modulate the charge transport of oligopeptide-SAM junctions by intermolecular interactions,which will advance our understanding of charge transport in biological systems and facilitate developing future electronics.

Many evolved biomolecular functions such as ion pumping or redox catalysis rely on controlled charge transport through the polypeptide matrix, which can be regulated by shifts in molecular protonation states and dependent supramolecular packing modes in response to environmental cues. However, the exact roles of such dynamic, non-covalent interactions in peptide charge transport have remained elusive. To tackle this challenge, here we report the modulation of charge transport in a series of lysine(Lys)-substituted hepta-glycine(Gly) peptide self-assembled monolayers(SAMs) on template-striped gold(Au^(TS)) bottom electrodes with eutectic gallium-indium(EGaIn) liquid metal top electrodes. We demonstrate systematic modulation of hydrogen bonding and more general electrostatic interactions by shifting the position of the charged Lys-residue and creating different protonation patterns by changing the environmental pH in the Au^(TS)/peptide//Ga Ox/EGa In junctions. The effective modulation is evidenced by current density-voltage(J-V) measurements combined with SAM characterization using ultraviolet photoelectron spectroscopy(UPS) and angle-resolved X-ray photoelectron spectroscopy(ARXPS), polarization modulation-infrared reflection-absorption spectroscopy(PM-IRRAS), and molecular dynamics(MD) simulations. Decreasing the hydrogen bonding inside the peptide SAMs and increasing the electrostatic interactions by environmental counterions amplifies the charge transport differently with Lys-position, which means that the sensitive electrical response of peptide SAMs can be tuned by the peptide sequence. Our results provide insights into the relationship between molecular design and in situ modulation of charge transport properties for the development of bionanoelectronics.

分析180 kW大功率直流充电桩在不同工况下的气动噪声特性,进行整桩的降噪优化设计.采用计算流体力学(CFD)与计算气动声学(CAA)分步耦合仿真的方法,分析整桩的噪声频谱和声场分布规律;开展整桩试验测试,验证仿真的精确性;对比、分析不同降噪方案的降噪性能,并进行方案的试验验证.结果显示,整桩噪声主频为830 Hz,整桩噪声次频为650 Hz,分别与电源模块风扇以及系统风扇的叶片通过频率重合;在电源模块进风口布置吸音棉能够有效消除整桩噪声第三、第四峰值频率幅值;在系统风扇出风口布置吸音棉能够使整桩A计权声压级总值降低约1.2 dB;相同布置位置,在保证散热的同时增加吸音棉厚度的降噪效果不显著,整桩A计权声压级总值降低不足1.0 dB.

Soft robotics has been receiving increasing attention due to its flexibility and adaptability offered by embodied intelligence. A soft robot may undergo complex motions including stretching,contraction,bending,twisting,and their intricate combinations.Among these basic motions,bending plays a central role when a robot accomplishes tasks such as locomotion,grasping and manipulation. Although a rich repertoire of bending mechanisms has been reported,a systematic and rational design framework is still lack. In this paper,we provide a novel design strategy for soft bending actuators which allows integral modeling and design optimization. Nowadays,metamaterials are emerging as a new tool for soft robots,by encoding the desired complex motions directly within the material architectures,leading to conformable monolithic systems. We combine pneumatic actuators and flexible metamaterials to provide an alternative solution to soft bending actuators,with advantages of compact design,large bending motion,and convenient fabrication. A regular pneumatic chamber is embedded inside auxetic and non-auxetic metamaterials,and bending is generated when inflated. We carry out dimensionless analysis to identify the key design variables. To provide insight into design optimization,we develop a computation framework by modeling metamaterial structures with beam elements and the inner chamber with shell elements as an integral part,allowing efficient simulation of the coupled system. We systematically investigate how the bending angle varies with the key design variables and find the optimal design parameters.The experimental results are well in line with the simulation,and a remarkable bending motion of 0.43°/mm is achieved.

[目的]由于化学测定方法费时费力,很多历史土壤资料缺少阳离子交换量(CEC)信息。本研究旨在基于易获取的变量建立安徽省土壤CEC的预测模型。[方法]利用覆盖安徽全省区域的711个土样的有机质含量、颗粒组成和土壤pH的信息,采用逐步多元线性回归方法建立CEC预测模型;并按土壤层次、土壤类型、母质、土地利用方式、质地和石灰性六种分组方式检验了其对模型预测精度的影响。[结果](1)利用全省未分组数据建立的预测模型精度较低,调整R^(2)仅为0.33;(2)按土壤类型、土地利用方式和石灰性进行分组,整体上可提高模型的预测精度,调整R2提升至0.44~0.93;但按土壤层次、母质和质地进行分组,模型精度未得到明显改善甚至有所下降;(3)预测安徽省土壤CEC的参数重要性依次为土壤黏粒含量、土壤有机质含量和土壤pH。[结论]基于未分组数据集上建立的安徽省土壤CEC模型精度很低,依据土壤类型、土地利用方式和石灰性进行分组可提高CEC的预测精度,对CEC预测最重要的变量是土壤黏粒含量,其次是土壤有机质含量和土壤pH。
