Brain targeted delivery of paclitaxel using endogenous ligand

The objective of the present investigation was to formulate nanoparticles constructed using PLGA polymer for the effective targeted delivery to brain via nasal route. The PLGA nanoparticles were optimized using novel design of experiment technique by 23 full factorial design. Drug: polymer ratio（X1）, surfactant concentration（X2） and stirring speed（X3） were identified as critical process parameters, and its impact on particle size（Y1） and % entrapment efficiency（Y2） was studied. The optimized nanoparticle formulation was conjugated with glutathione as an endogenous ligand by using carbodiimide chemistry using(1-Ethyl-3-（3-dimethylaminopropyl）carbodiimide)（EDAC） as linker molecule. From Ellman's assay,it was found that a total of 691.27 ± 151 units of glutathione were conjugated upon each PLGA nanoparticle. The in vitro release studies as well as ex vivo studies revealed biphasic pattern of drug release with initial burst release followed by slow exponential release of drug over a period of 24 h. The in vivo biodistribution studies were conducted on rat following nasal administration of the nanoparticle formulation（conjugated and unconjugated）and were compared with plain paclitaxel suspension. The results clearly demonstrated that the brain targeting efficiency was enhanced with the glutathione conjugated formulation（387.474%） as compared to the unconjugated nanoparticle formulation（224.327%）. Further,the in vitro in vivo correlation studies revealed good relationship（r2> 0.95） as obtained from the levy plot. Glutathione proves to be an efficient vector for the successful transport of poor bioavailable drug to the brain.

Enhanced saturation magnetization in La^(3+) doped NiCo ferrites prepared by sol-gel auto-combustion method

Lanthanum doped nickel-cobalt nano ferrites with chemical formula Ni_(0.5)Co_(0.5)LaxFe_(2-x)O_(4)(x=0.05,0,10,0.15 and 0,20) were prepared using a simple sol-gel auto combustion *** basic structural properties were determined by X-ray diffraction method and the formation of single phased spinel ferrite was *** crystalline size decreased from 25 to 11 nm and lattice parameter a increases with increase of La *** surface morphology of these ferrites was observed by field-emission scanning electron microscopy(FESEM) and agglomerated irregular grains are observed with increase of the rare earth element La ***-dispersive X-ray spectroscopy(EDX) result confirms the presence of the required *** Fourier transform infrared spectroscopy(FTIr) spectrum indicates the formation of the spinel ferrite structure with M-O *** direct band measurements from ultraviolet-visible spectroscopy(Uv-vis) spectroscopy indicate that the direct band gap decreases from 1.39 to 1.19 ev for x=0.05 to x=0.15,then increases to 1.28 ev for x=*** room temperature magnetic properties of these ferrites were studied by a vibrating sample magnetometer(vSM).The enhanced saturation magnetization of 49.73 emu/g is observed for x=0.10 and then saturation magnetizations are gradually decreased for x=0.15 and x=*** the remanent magnetization and coercivity also follow the same trend.

The early Archean oceans were marked by significant redox changes which have subsequently shaped the Earth’s *** chemical sediments of banded Iron and Manganese formations provide important geochemical proxies for these historical shifts in the redox conditions and to trace the ancient sedimentation patterns and *** this study,we investigate the proto-ore of the Archean Mn-formations of the Sandur,Chitradurga and Shimoga greenstone belts of Dharwar Craton of southern Peninsular India,which is geochemically characterised as quartz arenites,Mn-arenites,Fe-arenites,Mn-argillites and *** geochemical systematics suggest their deposition in shallow to deeper shelf in the Archean *** detrital zircon U-Pb systematics of Mn arenites and argillites indicate their maximum depositional age of 3230±52 Ma representing the oldest onset of sedimentation during the Paleo-Mesoarchean timeframe in the Chitradurga Group of Dharwar *** detrital influx proxies suggest variations in sedimentation rates associated with the Archean transgressive-regressive cycles and fluctuating hydrodynamic conditions,together reflecting an increasing trend in the contributions of recycled sediment from Sandur to Chitradurga and Shimoga greenstone *** available detrital zircon ages of the Mn arenites and argillites from these greenstone belts indicate a~600 Ma prolonged period of Mn deposition for which high-T hydrothermal fluids from Archean mid-oceanic ridges supplied the *** trace element compositions of the concordant detrital zircons suggest 3.3-3.1 Ga Dharwar basement TTG/granitoid source which is corroborated by the zircon crystallization temperatures of 690-820℃.The source-normalisedα-dose rates of the detrital zircons signify greater degrees of sediment transport and multi-cycle nature which correspond to the earliest episode of crustal growth in the Indian sub-continent associated with the Mesoarchean Ur supercontinent.T

Background: Duodenoscope-related multidrug-resistant organism(MDrO) infections raise concerns. Disposable duodenoscopes have been recently introduced in the market and approved by regulatory agencies with the aim to reduce the risk of endoscopic retrograde cholangiopancreatography(ErCP) associated infections. The aim of this study was to evaluate the outcome of procedures performed with single-use duodenoscopes in patients with clinical indications to single-operator cholangiopancreatoscopy. Methods: This is a multicenter international, retrospective study combining all patients who underwent complex biliopancreatic interventions using the combination of a single-use duodenoscope and a single-use cholangioscope. The primary outcome was technical success defined as ErCP completion for the intended clinical indication. Secondary outcomes were procedural duration, rate of cross-over to reusable duodenoscope, operator-reported satisfaction score(1 to 10) on performance rating of the single-use duodenoscope, and adverse event(AE) rate. results: A total of 66 patients(26, 39.4% female) were included in the study. ErCP was categorized according to ASGE ErCP grading system as 47(71.2%) grade 3 and 19(28.8%) grade 4. The technical success rate was 98.5%(65/66). Procedural duration was 64(interquartile range 15-189) min, cross-over rate to reusable duodenoscope was 1/66(1.5%). The satisfaction score of the single-use duodenoscope classified by the operators was 8.6 ± 1.3 points. Four patients(6.1%) experienced AEs not directly related to the single-use duodenoscope, namely 2 post-ErCP pancreatitis(PEP), 1 cholangitis and 1 ***: Single-use duodenoscope is effective, reliable and safe even in technically challenging procedures with a non-inferiority to reusable duodenoscope, making these devices a viable alternative to standard reusable equipment.

Stability of stratified shear flows in channels with variable cross sections

For the instability problem of density stratified shear flows in sea straits with variable cross sections, a new semielliptical instability region is found. rurthermore, the instability of the bounded shear layer is studied in two cases：（i） the density which takes two different constant values in two layers and （ii） the density which takes three different constant values in three layers. In both cases, the dispersion relation is found to be a quartic equation in the complex phase velocity. It is found that there are two unstable modes in a range of the wave numbers in the first case, whereas there is only one unstable mode in the second case.

ZrB_(2)-SiC based composites for thermal protection by reaction sintering of ZrO_(2)+B_(4)C+Si

Synthesis and sintering of ZrB_(2)-SiC based composites have been carried out in a single-step pressureless reaction sintering(PLrS)of ZrO_(2),B_(4)C,and Si.Y_(2)O_(3) and Al_(2)O_(3) were used as sintering *** effect of ratios of ZrO_(2)/B_(4)C,ZrO_(2)/Si,and sintering additives(Y_(2)O_(3) and Al_(2)O_(3)),was studied by sintering at different temperatures between 1500 and 1680℃in argon ***_(2),SiC,and YAG phases were identified in the sintered *** as high as 4.2 g/cm^(3),micro hardness of 12.7 GPa,and flexural strength of 117.6 MPa were obtained for PLrS *** material was also prepared by PLrS for tungsten inert gas(TIG)welding of the ZrB_(2)-SiC based *** shear strength of the weld was 63.5 *** PLrS ZrB_(2)-SiC composites exhibited:(i)resistance to oxidation and thermal shock upon exposure to plasma flame at 2700℃for 600 s,(ii)thermal protection for Cf-SiC composites upon exposure to oxy-propane flame at 2300℃for 600 s.

Filler materials of(ZrB_2-SiC-B_4C-YAG) composite were developed for gas tungsten arc welding(GTAW) of the ZrB_2-SiC and Cf-SiC based composites to themselves and to each other. reaction with filler material,porosity and cracks were not observed at weld interfaces of all the joints. Penetration of filler material in to voids and pores existing in the Cf-SiC composites was observed. Average shear strength of 25.7 MPa was achieved for joints of Cf-SiC composites. By incorporation of Cf-SiC(CvD) ground short fibre reinforcement the(ZrB_2-SiC-B_4C-YAG) composite was machinable with tungsten carbide tool. The joint and machined composites were resistance to oxidation and thermal shock when exposed to the oxy-propane flame at 2300℃ for 300s. The combination of(ZrB_2-SiC-B_4C-YAG) and Cf-SiC based composites can be used for making parts like thermal protection system or nozzles for high temperature applications.

Gas tungsten arc welding of ZrB_2-SiC based ultra high temperature ceramic composites

The difficulty in fabricating the large size or complex shape limits the application of ZrB2-SiC composites. Joining them by fusion welding without or with preheating, controlled cooling under protective gas shield leads to thermal shock failure or porosity at the weld interface. In the present work, a filler material of(ZrB2-SiC-B4C-YAG) composite with oxidation resistance and thermal shock resistance was produced in the form of welding wire. Using the filler, gas tungsten arc welding(GTAW) was performed without employing preheating, post controlled cooling and extraneous protective gas shield to join hot pressed ZrB2-SiC(ZS), and pressureless sintered ZrB2-SiC-B4C-YAG(ZSBY) composites to themselves. The fusion welding resulted in cracking and non-uniform joining without any filler material. The weld interfaces of the composites were very clean and coherent. The vickers micro-hardness across the weld interface was found to increase due to the increase in the volume % of both SiC and B4C in the filler material. The shear strength of the weld was about 50% of the flextural strength of the parent composite.
