Effects of interleukin-10 treated macrophages on bone marrow mesenchymal stem cells via signal transducer and activator of transcription 3 pathway

BACKGROUND Alveolar bone defects caused by inflammation are an urgent issue in oral implant surgery that must be solved.Regulating the various phenotypes of macrophages to enhance the inflammatory environment can significantly affect the progression of diseases and tissue engineering repair process.AIM To assess the influence of interleukin-10(IL-10)on the osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs)following their interaction with macrophages in an inflammatory environment.METHODS IL-10 modulates the differentiation of peritoneal macrophages in Wistar rats in an inflammatory environment.In this study,we investigated its impact on the proliferation,migration,and osteogenesis of BMSCs.The expression levels of signal transducer and activator of transcription 3(STAT3)and its activated form,phos-phorylated-STAT3,were examined in IL-10-stimulated macrophages.Subsequently,a specific STAT3 signaling inhibitor was used to impede STAT3 signal activation to further investigate the role of STAT3 signaling.RESULTS IL-10-stimulated macrophages underwent polarization to the M2 type through substitution,and these M2 macrophages actively facilitated the osteogenic differentiation of BMSCs.Mechanistically,STAT3 signaling plays a crucial role in the process by which IL-10 influences macrophages.Specifically,IL-10 stimulated the activation of the STAT3 signaling pathway and reduced the macrophage inflammatory response,as evidenced by its diminished impact on the osteogenic differentiation of BMSCs.CONCLUSION Stimulating macrophages with IL-10 proved effective in improving the inflammatory environment and promoting the osteogenic differentiation of BMSCs.The IL-10/STAT3 signaling pathway has emerged as a key regulator in the macrophage-mediated control of BMSCs’osteogenic differentiation.

Data-driven discovery of partial differential equations(PDEs)has recently made tremendous progress,and many canonical PDEs have been discovered successfully for proof of concept.However,determining the most proper PDE without prior references remains challenging in terms of practical applications.In this work,a physics-informed information criterion(PIC)is proposed to measure the parsimony and precision of the discovered PDE synthetically.The proposed PIC achieves satisfactory robustness to highly noisy and sparse data on 7 canonical PDEs from different physical scenes,which confirms its ability to handle difficult situations.The PIC is also employed to discover unrevealed macroscale governing equations from microscopic simulation data in an actual physical scene.The results show that the discovered macroscale PDE is precise and parsimonious and satisfies underlying symmetries,which facilitates understanding and simulation of the physical process.The proposition of the PIC enables practical applications of PDE discovery in discovering unrevealed governing equations in broader physical scenes.

Micro-mechanism Study on the Different Antioxidants Protective Effect of Vegetable Oil by ReaxFF Molecular Dynamics

The aging problem of vegetable oil has severely restricted the popularization and application of vegetable oil transformers.Adding antioxidants is the most recommended solution.At present,there is insufficient research on the micromechanism application of the protective effect of antioxidants on vegetable oil.In this paper,multiple vegetable oil models with different types and concentrations of antioxidants are established.Then the molecular dynamics simulation based on ReaxFF is carried out at different temperatures and oxygen content.Results show that the protective mechanism of antioxidants is to release H and combine this with free radicals generated by the decomposition of unsaturated fatty acids(UFA)in the oil.In addition,the protective effects of four antioxidants show different results.The stronger ones are tert-butyl hydroquinone(TBHQ)and butylated hydroxytoluene(BHT),while the weaker ones are butylated hydroxyanisole(BHA)and propyl gallate(PG).TBHQ has a better protective effect at lower concentrations,but the decomposition of UFA is promoted at higher concentrations.When the temperature rises,the decomposition of UFA is promoted.With the addition of oxygen,smaller molecular compounds are easily oxidized,and the decomposition of UFA is accelerated.The above research could reveal the microscopic mechanism of antioxidant protection on vegetable oil,providing theoretical references for further exploration of effective vegetable oil aging protection technology.

Paclitaxel,a tetracyclic diterpenoid,has garnered attention for its potent anti-cancer properties and intricate molecular structure,making it a significant target for chemical synthesis and biosynthesis[1].However,its natural sources are extremely limited,as it is derived exclusively from the bark of endangered genus Taxus plants,which contain paclitaxel in very low concentrations(0.01%−0.05%)[2-3].Recent advances in synthetic biology present promising opportunities to enhance paclitaxel levels in Taxus cell cultures or to enable the reconstitution of its production in heterologous hosts,such as yeast or tobacco(Nicotiana benthamiana).

Scenario Modeling-Aided AP Placement Optimization Method for Indoor Localization and Network Access

Owing to the ubiquity of wireless networks and the popularity of WiFi infrastructures,received signal strength(RSS)-based indoor localization systems have received much attention.The placement of access points(APs)significantly influences localization accuracy and network access.However,the indoor scenario and network access are not fully considered in previous AP placement optimization methods.This study proposes a practical scenario modelingaided AP placement optimization method for improving localization accuracy and network access.In order to reduce the gap between simulation-based and field measurement-based AP placement optimization methods,we introduce an indoor scenario modeling and Gaussian process-based RSS prediction method.After that,the localization and network access metrics are implemented in the multiple objective particle swarm optimization(MOPSO)solution,Pareto front criterion and virtual repulsion force are applied to determine the optimal AP placement.Finally,field experiments demonstrate the effectiveness of the proposed indoor scenario modeling method and RSS prediction model.A thorough comparison confirms the localization and network access improvement attributed to the proposed anchor placement method.

Coalbursts in China: Theory, practice and management

Coalburst is one of the most serious disasters that threaten the safe production of coal mines, and this disaster is particularly serious in China. This paper presents an overview of coalbursts in China since 1980s. From the "stress and energy" and "regional and local" perspectives, the achievements in the theory, practice and management of coalbursts in China are systematically summarized. A theoretical system of coalbursts has been formed to reveal the deformational behavior of coalbursts and explain the mechanism of coalbursts. The occurrence conditions of coalbursts are put forward and the critical stress is obtained. The stress index method for risk evaluation of coalbursts before mining is proposed, and the deformation localization prediction method of coalbursts is put forward. The relationship between energy release and absorption in the process of coalbursts is found, and the prevention and control methods of coalbursts, including the regional method, the local method and support, are presented. The safety evaluation index of coalburst prevention and control is put forward. The integrated prevention and control method for coal and gas outbursts is proposed. The prevention and control technology and equipment of coalbursts have also been developed. Amongst them, the distribution law of the critical stress in China coalburst mines is discovered. The technology and equipment for monitoring, prevention and control of coalbursts, as well as for integrated prevention and control of combined coalbursts and other disasters, have been developed. The energy-absorbing and coalburst-preventing support technology for roadways is invented, and key engineering parameters of coalburst prevention and control are pointed out. In China, coalburst prevention and control laws and standards have been developed. Technical standards for coalbursts are formulated, statute and regulations for coal mines are established, and regulatory documents are promoted.

A UWB/IMU-Assisted Fingerprinting Localization Framework with Low Human Efforts

With the rapid development of smart phone,the location-based services(LBS)have received great attention in the past decades.Owing to the widespread use of WiFi and Bluetooth devices,Received Signal Strength Indication(RSSI)fingerprintbased localization method has obtained much development in both academia and industries.In this work,we introduce an efficient way to reduce the labor-intensive site survey process,which uses an UWB/IMU-assisted fingerprint construction(UAFC)and localization framework based on the principle of Automatic radio map generation scheme(ARMGS)is proposed to replace the traditional manual measurement.To be specific,UWB devices are employed to estimate the coordinates when the collector is moved in a reference point(RP).An anchor self-localization method is investigated to further reduce manual measurement work in a wide and complex environment,which is also a grueling,time-consuming process that is lead to artificial errors.Moreover,the measurements of IMU are incorporated into the UWB localization algorithm and improve the label accuracy in fingerprint.In addition,the weighted k-nearest neighbor(WKNN)algorithm is applied to online localization phase.Finally,filed experiments are carried out and the results confirm the effectiveness of the proposed approach.

Prognostic analysis of related factors of adverse reactions to immunotherapy in advanced gastric cancer and establishment of a nomogram model

BACKGROUND Immunotherapy for advanced gastric cancer has attracted widespread attention in recent years.However,the adverse reactions of immunotherapy and its relationship with patient prognosis still need further study.In order to determine the association between adverse reaction factors and prognosis,the aim of this study was to conduct a systematic prognostic analysis.By comprehensively evaluating the clinical data of patients with advanced gastric cancer treated by immunotherapy,a nomogram model will be established to predict the survival status of patients more accurately.AIM To explore the characteristics and predictors of immune-related adverse reactions(irAEs)in advanced gastric cancer patients receiving immunotherapy with programmed death protein-1(PD-1)inhibitors and to analyze the correlation between irAEs and patient prognosis.METHODS A total of 140 patients with advanced gastric cancer who were treated with PD-1 inhibitors in our hospital from June 2021 to October 2023 were selected.Patients were divided into the irAEs group and the non-irAEs group according to whether or not irAEs occurred.Clinical features,manifestations,and prognosis of irAEs in the two groups were collected and analyzed.A multivariate logistic regression model was used to analyze the related factors affecting the occurrence of irAEs,and the prediction model of irAEs was established.The receiver operating characteristic(ROC)curve was used to evaluate the ability of different indicators to predict irAEs.A Kaplan-Meier survival curve was used to analyze the correlation between irAEs and prognosis.The Cox proportional risk model was used to analyze the related factors affecting the prognosis of patients.RESULTS A total of 132 patients were followed up,of whom 63(47.7%)developed irAEs.We looked at the two groups’clinical features and found that the two groups were statistically different in age≥65 years,Ki-67 index,white blood cell count,neutrophil count,and regulatory T cell(Treg)count(all P<0.05).Multivariate logistic regression an

A hybrid machine learning optimization algorithm for multivariable pore pressure prediction

Pore pressure is essential data in drilling design,and its accurate prediction is necessary to ensure drilling safety and improve drilling efficiency.Traditional methods for predicting pore pressure are limited when forming particular structures and lithology.In this paper,a machine learning algorithm and effective stress theorem are used to establish the transformation model between rock physical parameters and pore pressure.This study collects data from three wells.Well 1 had 881 data sets for model training,and Wells 2 and 3 had 538 and 464 data sets for model testing.In this paper,support vector machine(SVM),random forest(RF),extreme gradient boosting(XGB),and multilayer perceptron(MLP)are selected as the machine learning algorithms for pore pressure modeling.In addition,this paper uses the grey wolf optimization(GWO)algorithm,particle swarm optimization(PSO)algorithm,sparrow search algorithm(SSA),and bat algorithm(BA)to establish a hybrid machine learning optimization algorithm,and proposes an improved grey wolf optimization(IGWO)algorithm.The IGWO-MLP model obtained the minimum root mean square error(RMSE)by using the 5-fold cross-validation method for the training data.For the pore pressure data in Well 2 and Well 3,the coefficients of determination(R^(2))of SVM,RF,XGB,and MLP are 0.9930 and 0.9446,0.9943 and 0.9472,0.9945 and 0.9488,0.9949 and 0.9574.MLP achieves optimal performance on both training and test data,and the MLP model shows a high degree of generalization.It indicates that the IGWO-MLP is an excellent predictor of pore pressure and can be used to predict pore pressure.

The Al_(2)O_(3)and Mn/Al_(2)O_(3)sorbents highly utilized in destructive sorption of NF_(3)

NF_(3)is commonly used as an etching and cleaning gas in semiconductor industry,however it is a strongly greenhouse gas.Therefore,the destruction of disposal NF_(3)is an urgent task to migrate the greenhouse effect.Among the technologies for NF_(3)abatement,the destructive sorption of NF_(3)over metal oxides sorbents is an effective way.Thus,the search for a highly reactive and utilized sorbent for NF_(3)destruction is in great demand.In this work,AlOOH supported on carbon-sphere(AlOOH/CS)as precursors were synthesized hydrothermally and heat-treated to prepare the Al_(2)O_(3)sorbents.The influence of AlOOH/CS hydrothermal temperatures on the reactivity of derived Al_(2)O_(3)sorbents for NF_(3)destruction was investigated,and it is shown that the Al2O3 from AlOOH/CS hydro-thermalized at 120℃is superior to others.Subsequently,the optimized Al_(2)O_(3)was covered by Mn(OH)x to prepare Mn/Al_(2)O_(3)sorbents via changing hydrothermal temperatures and Mn loadings.The results show that the Mn/Al_(2)O_(3)sorbents are more utilized than bare Al_(2)O_(3)in NF_(3)destructive sorption due to the promotional effect of Mn_(2)O_(3)as surface layer on the fluorination of Al_(2)O_(3)as substrate,especially the optimal 5%Mn/Al2O3(160℃)exhibits a utilization percentage as high as 90.4%,and remarkably exceeds all the sorbents reported so far.These findings are beneficial to develop more efficient sorbents for the destruction of NF_(3).
