Many researchers have developed various methods for in-planta or floral dip transformation of Arabidopsis thaliana, one of the simple protocol and widely used to produce transgenic Arabidopsis. As the efficiency and ease of getting a transformant is very much time consuming effort and less number of the transformants people get, we have developed a little modified transformation protocol to avoid the disparities. Four types of inoculums (inoculum1, inoculum2, inoculum3 and inoculum4) were used to check the transformation efficiency out of which Inoculum3 showed the highest rate of transformation among the four types. 0.07% Twin-20 also acts in same manner as silwet L-77 to increase the rate of transformation efficiency and glucose instead of sucrose can be used in inoculum to transform Arabidopsis. After vacuum infiltration keeping the Agrobacterium infected plants for 7-8 hrs horizontally in low light at 280C temperature condition, considered best to get an increased number of transformed seeds. Modified protocol produced ~12-14% increase in transformants. Selection pots (kanamycin supplemented soil filled pots) in place of selection plates (Kanamycin supplemented Murashige and Skoog agar plates) proved beneficial as no MS medium and no aseptic condition is required for selection of transformed plants. This increase in transformation efficiency consequently increased the percentage of homozygous and single copied stable transgenic lines.

Na_(0.5)Bi_(0.5)TiO_(3)-based lead-free piezoelectrics are considered for potential replacement of the lead-based commercial piezoceramics in high-power transducer *** have examined the role of grain size in influencing the structural-polar inhomogeneity of stoichiometric and off-stoichiometric Na_(0.5)Bi_(0.5)TiO_(3)(NBT),and its morphotropic-phase-boundary(MpB)derivative 0.94Na_(0.5)Bi_(0.5)TiO_(3)-0.06BaTiO_(3)(NBT-6BT).Our study reveals that size effect comes into play in these systems on a very large length scale(on the scale of microns)considerably affecting its global structure and properties.

Microscopic Observation of Nuclear Track pores in polymeric Membranes

Nuclear track pores were develop and observed in polycarbonate and LR-115 membranes. The polymer membranes were irradiated by swift heavy ions (SHI) and etched chemically. The stopping range of heavy ion is larger than the thickness of the membrane. The fluence of ions was changes from 103 to 107 ions-cm-2. The etching time was increased till pore size reaches up to micron range. The etching conditions were set to develop the porous tracks. The tracks are revealed under optical microscope as the size reaches up to micron or sub-micron size. The size was measured using standard electron diffraction grid. Results show that the porous tracks having average size of 2.4 micron can be developed in polycarbonate, while 5 - 10 micron porous tracks can be developed in LR-115.

pEGylated-L-asparaginase(pEG-ASNase)is a chemotherapeutic agent used to treat pediatric acute lymphoblastic leukemia(ALL).Its use is avoided in adults due to its high risk of liver injury including hepatic steatosis,with obesity and older age considered risk factors of the *** study aims to elucidate the mechanism of pEG-ASNase-induced liver *** received 1500 U/kg of pEG-ASNase and were sacrificed 1,3,5,and 7 days after drug *** triglycerides were quantified,and plasma bilirubin,ALT,AST,and non-esterified fatty acids(NEFA)were *** mRNA and protein levels of genes involved in hepatic fatty acid synthesis,β-oxidation,very low-density lipoprotein(VLDL)secretion,and white adipose tissue(WAT)lipolysis were *** developed hepatic steatosis after pEG-ASNase,which associated with increases in bilirubin,ALT,and *** hepatic genes ppara,Lcad/Mcad,Hadhb,Apob100,and Mttp were upregulated,and Srebp-1 c and Fas were downregulated after *** plasma NEFA,WAT loss,and adipose tissue lipolysis were also observed after ***,we found that pEG-ASNase-induced liver injury was exacerbated in obese and aged mice,consistent with clinical studies of ASNase-induced liver *** data suggest that pEG-ASNase-induced liver injury is due to drug-induced lipolysis and lipid redistribution to the liver.

The regulation of gene expression in brain vicissitudes during aging is still not much known and explored. Differential gene expression and regulation is a key factor involved to identify the important landmarks within the brain transcriptome to study neuronal aging. Recentlypan>, transcriptomic studies are highly explored to understand and depict diseased versus normal as next generation sequencing enables to capture the complete biological context to the entire genome. Study of gene expression during aging compared to young flies provides a signature and scenario of gene expression and regulation during aging. In this study, we took advantage of NGS raw data of young and old flies head from SRA database of NCBI and decrypted the gene expression regulation during normal aging in drosophila model. We identified 350 genes with significant differential expression between young and old flies having 0.01% FDR. Various pathways in context to identified genes which are involved in aging include autophagy i.e. cell death and apoptosis, proteolysis, oxidativepan style="font-family:Verdana;"> stress, declination grey and white matter and neurotransmitter levels, mitochondrial discrepancy, electron transport chain, sugar degradation pathways, activation of transcription factors involved in epigenetic changes, regulators involved in negative and positive regulation WNT signaling pathways, G protein coupled receptor etc. as all these factors contribute to neurodegeneration and possibly dementia in normal aging. To perform a systematic review and meta-analysis on clinical outcomes of photodynamic therapy (pDT) in non-resectable *** studies compared outcomes with photodynamic therapy and biliary stenting (pDT group) vs biliary stenting only (BS group) in palliation of non-resectable cholangiocarcinoma. Articles were searched in MEDLINE, pubMed, and EMBASE. pooled proportions were calculated using fixed and random effects model. Heterogeneity among studies was assessed using the Ip>2p> *** studies (n = 402) that met inclusion criteria were included in this analysis. The p for χp>2p> heterogeneity for all the pooled accuracy estimates was > 0.10. pooled odds ratio for successful biliary drainage (decrease in bilirubin level > 50% within 7days after stenting) in pDT vs BS group was 4.39 (95%CI: 2.35-8.19). Survival period in pDT and BS groups were 413.04 d (95%CI: 349.54-476.54) and 183.41 (95%CI: 136.81-230.02) respectively. The change in Karnofsky performance scores after intervention in pDT and BS groups were +6.99 (95%CI: 4.15-9.82) and -3.93 (95%CI: -8.63-0.77) respectively. Odds ratio for post-intervention cholangitis in pDT vs BS group was 0.57 (95%CI: 0.35-0.94). In pDT group, 10.51% (95%CI: 6.94-14.72) had photosensitivity reactions that were self-limiting. Subgroup analysis of prospective studies showed similar results, except the incidence of cholangitis was comparable in both *** palliation of unresectable cholangiocarcinoma, pDT seems to be significantly superior to BS alone. pDT should be used as an adjunct to biliary stenting in these patients.
