Burn-Up Measurements on Dissolver Solution of Mixed Oxide Fuel Using HpLC-Mass Spectrometric Method

Burn-up measurement on an irradiated mixed oxide (MOX) test fuel pellet was carried out through measurements on the dissolver solution by HpLC-Thermal Ionization Mass Spectrometric (TIMS) technique. The studies carried out using HpLC as well as TIMS for quantification of burn-up value are described. While in one case, both the separation and determination of elements of interest (U, pu and Nd) were carried out by HpLC;in another case, TIMS technique was used to quantify them from the HpLC separated *** rapid separation procedures developed in our laboratory earlier were employed to isolate pure fractions of the desired elements. The individual lanthanide fission products (La to Eu) were separated from each other using dynamic ion-exchange chromatographic technique whereas uranium and plutonium were separated from each other using reversed phase chromatographic technique. The pure fractions of U, pu and Nd obtained after HpLC separation procedure for “spiked” and “unspiked” dissolver solutions were used in TIMS measurements for the first time in our laboratory. In TIMS analysis, isotopic abundances of uranium, plutonium and neodymium fractions obtained from HpLC separation procedure on an “unspiked” fuel sample were measured. For the determination of U, pu and Nd by isotopic dilution mass spectrometric technique (IDMS), known quantities of tracers enriched in 238U, 240pu and 142Nd were added to the pre-weighed dissolver solution and subjected to HpLC separation procedures. The isotope ratios viz. 142Nd/(145Nd +146Nd), 238U/233U and 240pu/239pu in the pertinent “spiked” fractions were subsequently measured by TIMS. The spikes were pre-standardized in our laboratory employing reverse isotopic dilution technique against the standard solutions available in our laboratory (for 238U, 239pu and 142Nd, standard solutions of 233U, 239pu (of higher abundance than in the sample) and 150Nd were employed as spikes). The over extraction of groundwater from the coastal aquifers, result in reduction of groundwater resource and lowering of water level. In general, the depletion of groundwater level enhances the landward migration of saltwater wedge. pondicherry is one such region with recent alluvium as the major formation. Since the study area forms a part of the coastal aquifer system this behaves as a fragile ecosystem. The present study has been attempted to calculate the extraction of water and to estimate the amount of recharge into this alluvial aquifer by using groundwater level variations. The monthly water level fluctuation was observed during the study period (2000-2002) in eighteen locations. The maximum rise in groundwater level observed during 2000 was considered as the initial water level for the study and the subsequent decline in water level (draw down) was monitored monthly until the rising trend was noted. This indicates the fall in water level due to extraction. Later keeping the deepest draw down as the initial value increasing water level trend was studied until there was a notice of decline in groundwater level. This indicates as the rise in water level due to recharge. This method of observation carried out at a single location was adopted for all eighteen locations. The spatial representation of these data for eighteen locations were carried out by using GIS and the area occupied by different groundwater level contours were calculated and the amount of water withdrawn/re- charged was estimated. The maximum recharge was noted in the central and the northern part of the study area when compared to the other regions. Similarly, the maximum discharge was noted in the northern and the southern part of the study area during the study period.

Supercritical Fluid Extraction (SFE) is emerging as a powerful technique in the extraction of metal ions. In the present study, the extraction of nitrates of uranium and thorium was carried out using supercritical carbon dioxide (Sc-CO2) modified with various organophosphorous compounds such as dialkylalkyl phosphonates, trialkyl phosphates and trialkyl phosphine oxides in the presence of co-solvents such as methanol, dichlormethane and n-hexane. The influence of ligand and co-solvent on the extraction of the metal nitrates was studied in detail. These studies have established that co-solvent plays an important role in the extraction as well as fractionation of uranium and thorium nitrates. polar co-solvent, methanol provided faster extraction without fractionation whereas the non-polar solvent, e.g. n-hexane provided some fractionation of metal nitrates though the extraction kinetics was slower.

由于会产生大量的固体废弃物,皮革生产过程中的鞣前操作总是一个具有挑战性的工作。清洁生产是消除现有制革污染问题的新途径。在清洁皮革加工技术背景下,酶是传统化学品的最好替代者。目前研究在鞣前操作中使用酶,蛋白酶用于脱毛,淀粉酶用于除去纤维间质。蛋白酶和淀粉酶可以复配和单独用于传统的浸灰和脱灰工序。在转鼓中复合酶用量3.5%最佳。脱毛效果和纤维松散程度通过孔隙尺寸和污染负荷来评价,形貌特征通过染色和扫描电镜观察分析评价,也评价了坯革的物理力学性能、感官性能和视觉效果。就脱毛和一般外观而言,复合酶的脱毛效果与传统脱灰系统相当。酶脱毛后蓝湿革收缩温度约为112℃,成革的力学强度符合联合国工发组织(UNIDO)规范。扫描显微电镜证实纤维未损伤变形。酶处理过程中,纤维紧实、松散及分布均匀。单一的酶处理步骤降低了污染负荷。本研究揭示了制革工业中酶的作用,并对现有的清洁生产系统提供了一个新的生物资源利用途径。

Infectious hepatitis viral diseases are major health problems in both developing and developed countries. Hepatitis E virus（HEV） is an important cause of severe hepatitis in humans and is responsible for high rate of mortality in pregnant women. We developed a recombinant HEV vaccine based on virus-like particles（VLps）, denoted as p239, which had been demonstrated with high efficacy against Hepatitis E and good safety profile in a large-scale, randomized, double-blind placebo-controlled phase 3 trial. At present, our HEV239 vaccine Hecolin has been licensed for use in China. To understand the functional moieties in p239, we did a series of truncations of p239 to determine the minimal structural requirements for dominant epitopes and solved the crystal structure of its minimum dimerization domain（E2s） at 2 A resolution [1]. With respect to other structures of HEV VLps, this domain protrudes from the viral surface. Functional studies illustrated that this domain formed an intimate homodimer and that this dimerization is essential for HEV–host interactions. In addition, we also showed that the neutralizing antibody recognition site of HEV is located on the E2 s domain. Furthermore, we determined the complex crystal structure of 8C11 Fab, a neutralizing antibody, with HEV E2s（I） domain at 1.9 A resolution. The 8C11 epitopes on E2 s were identified at Asp496-Thr499, Val510-Leu514 and Asn573-Arg578. The mutagenesis and cell model assay identified Arg512 as the most crucial residue for 8C11 interaction and for the neutralization of HEV [2]. For assembly mechanism, a highly conserved 28-aa region（aa368-395） of p239 was found to be critical for particle assembly [3]. Interestingly, p239 can form particles even in 4 M urea from its homodimer precursor（as E2 forms） with a salt trigger. Three hydrophobic residues Leu372, Leu375, and Leu395 located on N-terminal of p239 were shown to play key roles in the assembly of p239 via hydrophobic interaction [4]. Thus, structural tailor

OBJECTIVE platinum compounds such as cisplatin and carboplatin are frequently used as the first-line chemotherapy for the treatment of the head and neck squamous cell carcinoma(HNSCC).In the present study,we investigated whether garcinol,apolyisoprenylated benzophenone can chemosensitize HNSCC to *** The effect of garcinol and cisplatin on HNSCC was assessed by MTT,Western blotting,real time pCR,FACS,immunohistochemistry,DNA binding assay and xenograft mouse *** We found that garcinol inhibited the viability of a panel of diverse HNSCC cell lines,enhanced the apoptotic effect of cisplatin,suppressed constitutive as well as cisplatin-induced NF-κB activation,and downregulated the expression of various oncogenic gene products(cyclin D1,Bcl-2,survivin and VEGF).In vivo study showed that administration of garcinol alone(0.5 mg·kg-1,ip five times/week)significantly suppressed the growth of the tumor,and this effect was further increased by *** the markers of proliferation index(Ki-67)and microvessel density(CD31)were downregulated in tumor tissues by the combination of cisplatin and *** pharmacokinetic results of garcinol indicated that good systemic exposure was achievable after ip administration of garcinol at 0.5and 2mg·kg-1 with mean peak concentration(cmax)of 1825.4 and 6635.7nmol·L-1 in the mouse serum,*** Overall,our results suggest that garcinol can indeed potentiate the effects of cisplatin by negative regulation of various inflammatory and proliferative biomarkers.
