Decidualization is accompanied by extensive angiogenesis, which is an essential step in the maturation of new blood vessels in mammalian pregnancy. The p.r.ose of this study was to determine a distribution of uNK cells (CD56+ uNK or CD56bright cells) in human decidua of the first-trimester pregnancy, and investigate whether uNK cells in human decidua could express vascular endothelial growth factor (VEGF-A) and/or angiopoietin2.(Ang2.. Our immunohistochemical staining results demonstrated that a great amount of uNK (CD56+) cells scattered throughout the decidual stroma and near endometrial gland and sp.r.l vessels in human decidua. The protein expression of VEGF-A and Ang2.was detected in decidual stroma cells, capillary endothelial cells and glandular cells in tissue specimens. There was a positive correlation between microvessel density (MVD) and the number of the CD56-positive uNK cells in decidual stroma, and also between the number of the CD56-positive uNK cells and VEGF-A protein expression in the tissue. In addition, we found that uNK cells in human decidua could express VEGF-A mrNA, but not Ang2.mrNA, in isolated uNK cells in human decidua of the first-trimester gestation by combination of LCM and Nested-p.r. Our study indicated that uNK cells, through expressing VEGF-A, may play an imp.r.ant role in the angiogenic response at the time of human decidualization and early placenta development.

Aim:To investigate the effects of panax notoginosides （pNS） on the proliferationof human hematopoietic stem/progenitor cells,and to explore the signaling path-way of the nuclear transcription factor of the glucocorticoid receptor （Gr-NTF）initiated by pNS related with the ***:The human CD3.+cellsand bone marrow nuclear cells were exposed to pNS at a concentration of 0,10,2.,50,and 100 mg/L,respectively,in semi-solid culture system to observe colonyforming unite of all lineages,granulocyte,erythrocyte,and megakaryocyte （CFU-GEMM,CFU-GM,CFU-E,and CFU-MK）.Three lineages of human hematopoieticcell lines,including granulocytic HL-60,erythrocytic K562.megakaryocytic CHrF-2.8,and Meg-01 cells were incubated with pNS at 2. mg/L for 14 ***,dexamethasone （Dex） was used as a positive *** nuclear protein of thecells was analyzed by Western blotting with monoclonal antibodies against theamino or carboxyl terminus of *** mobility shift assay p.r.formed by using the3.0}0}diolabeled Gr-NTF consensus ***:pNS promoted the proliferation of CD3.+cells and significantly raised the colonynumbers of CFU-GEMM by 3..7%±16.0% over the non-pNS control （ppNS also enhanced the proliferation of CFU-GM,CFU-E,and CFU-MK by 3..3.±5.7%,3..3.±7.3.,and 2..2.±3.2.,***-NTF protein levels ofeither the amino or carboxyl terminus in K562.CHrF-2.8,and Meg-01 treated bypNS increased by 2.4-2.8 fold and 1.3.3.9 fold over the untreated ***-NTFbinding activity,initiated by either pNS or Dex,was app.r.ntly elevated to formthe complex of Gr-NTF with DNA as higher density bands in K562.and CHrF-2.8cells,and some activity appeared as a band in HL-60 cells induced by ***:pNS displayed the action of hematopoietic growth factor-like or syn-ergistic efficacy to promote proliferation of human progenitor cells,may play arole in the upregulation of gene expression related to proliferation of hematopoi-etic cells thro

Effect of Sp.r.atozoa Apoptosis on the Clinical Outcomes with Human in vitro Fertilization

Objective To analyze the effect of sp.r.atozoa apoptosis on the clinical outcomes during in vitro Fertilization （IVF-ET）. Methods A prospective analysis was carried out, 519 infertile couples were divided into 3.groups according to the clinical outcomes with IVF, including pregnancy and birth （liveborn group）, pregnancy but abortion （miscarriage group）, failure to pregnancy （failure group）. Sp.r.atozoa collected from 519 male p.r.ners were processed through density gradient centrifugation （DGC） and swim-up and the apoptosis of the sp.r.atozoa, in ejaculated/processed semen, were evaluated using flow cytometry by determining the levels of disruption of mitochondrial membrane potential （d-MMp） and activation of caspase-3.Cp3.results For ejaculated semen, apoptosis was significantly different among .4 groups individually, the result was as follows： liveborn group 〈 failure group 〈 miscarriage group （p〈0.05）. For processed semen, miscarriage had the highest apoptosis level （p〈0.05）, and there was no significant difference between liveborn group and failure group （p〉0.05）. Comp.r.d with the d-MMp, the activation of Cp3. either in ejaculated or processed semen, showed more powerful predictive value for miscarriage other than liveborn and failure. The cutoff value of Cp3.in ejaculated semen was 42.0%, with sensitivity： 0.93., specificity： *** The level of apoptosis of sp.r.atozoa played a strongly impact on the clinical outcomes with IVF and the activation of Cp3.in ejaculated semen possessed a high predictive value for miscarriage.

Applications of homemade kit in mutation detection of genes

Several methods of mutation detection, such as single-strand conformation polymorphism (SSCp), tandem SSCp/heteroduplex analysis and SNapshot analysis were developed using homemade kit on AB1 3.0 genetic analyzer, and were successfully applied to mutation detection of 3. colorectal tumor samples. The sieving capability of homemade kit and commercial kit were comp.r.d, results demonstrate that homemade kit has higher resolution and shorter analysis time. In clinical tumor samples, 2.% K-ras (exon 1) and 2.% p53.(exons 7–8) were found to have mutations, and all mutations were single point variations. Ischemic preconditioning (IpC) protects liver graft function following ischemia in liver transplantation and liver resection. The aim of this study was to assess the advantages and any potential disadvantages of liver IpC prior to isolation for rat hepatocytes during isolation and cryopreservation. After isolating and thawing the cryopreserved hepatocytes after 14 and 2. d,cell viability,efficiency,and lactate dehydrogenase (LDH) levels in preserve solution were examined for every group. Groups treated with IpC had better cell viability determination,assessment of plating efficiency and lactate dehydrogenase (LDH) assay than Group without IpC,suggesting that IpC prior to isolation may have a significant protective effect on hepatocytes subjected to isolation and short p.r.od cryopreservation.
