Automated evaluation of parapapillary choroidal microvasculature in crowded optic discs:a controlled,optical coherence tomography angiography study

AIM:To compare superficial and deep vascular properties of optic discs between crowded discs and controls using optical coherence tomography angiography(OCT-A).METHODS:Thirty patients with crowded discs,and 47 control subjects were enrolled in the study.One eye of each individual was included and OCT-A scans of optic discs were obtained in a 4.5×4.5 mm^(2) rectangular area.Radial peripapillary capillary(RPC)density,peripapillary retinal nerve fiber layer(pRNFL)thickness,cup volume,rim area,disc area,cup-to-disc(c/d)area ratio,and vertical c/d ratio were obtained automatically using device software.Automated parapapillary choroidal microvasculature(PPCMv)density was calculated using MATLAB software.When the vertical c/d ratio of the optic disc was absent or small cup,it was considered as a crowded disc.RESULTS:The mean signal strength index of OCT-A images was similar between the crowded discs and control eyes(P=0.740).There was no difference in pRNFL between the two groups(P=0.102).There were no differences in RPC density in whole image(P=0.826)and peripapillary region(P=0.923),but inside disc RPC density was higher in crowded optic discs(P=0.003).The PPCMv density in the inner-hemisuperior region was also lower in crowded discs(P=0.026).The pRNFL thickness was positively correlated with peripapillary RPC density(r=0.498,P<0.001).The inside disc RPC density was negatively correlated with c/d area ratio(r=-0.341,P=0.002).CONCLUSION:The higher inside disc RPC density and lower inner-hemisuperior PPCMv density are found in eyes with crowded optic discs.

Exosomes are important biomarkers for clinical diagnosis.It is critical to isolate secreted exosomes from bodily fluids such as blood,saliva,breast milk,and urine for liquid biopsy applications.The field of microfluidics provides numerous benefits for biosample processing,diagnostics,and prognostics.Several microfluidics-based methods have been employed for the isolation and purification of exosomes in the last ten years.These microfluidic methods can be grouped into two categories based on passive and active isolation mechanisms.In the first group,inertial and hydrodynamic forces are employed to separate exosomes based on their size differences.In the second group,external forcefields are integrated into microfluidic platforms to actively isolate exosomes from other bioparticles.In this paper,the application of microfluidic methods in exosome isolation is discussed,and future perspectives on this field are highlighted.

Background:Raloxifene,a selective estrogen receptor modulator,is also known to be a lysosomotropic agent.The bioavailability of raloxifene is around 2%due to extensive hepatic transport.Exosomes are nanosized vesicles that are naturally released from cells.Method:In this study,exosomes released from HeLa cervical cancer cells were loaded with raloxifene to increase its bioavailability,and an aptamer was attached to the exosome membrane for targeting only HeLa cells.Characterization of exosomes isolated from HeLa cells was performed by transmission electron microscopy,zeta sizer,and western blotting.In addition,the cytotoxic,apoptotic,autophagic,and lysosomotropic effects of the prepared Exo-Apt-Ral formulation on HeLa cervical cancer cells were investigated.Results:According to zeta analysis,the sizes of the empty exosome and Exo-Apt-Ral formulation were measured as 66±12 and 120±21 nm,respectively.There was a rise in the lysosomal permeability of HeLa cells after the Exo-Apt-Ral application.In addition,both apoptotic and autophagic death mechanisms were triggered in HeLa cells after the Exo-Apt-Ral application.Conclusion:This study showed that raloxifene functionalized by loading into aptamer-bound exosomes can be a new targeted drug carrier system for cervical cancer.

Nursing students’experiences of horizontal violence and occupational belonging during clinical placements

Background:Horizontal violence has negatively affected the students’active learning,interpersonal relationship,and occupational belonging.However,little attention has been paid to the relationship between horizontal violence and occupational belonging among nursing students.Objective:The objective of this study was to explore the frequency,types,perpetrators,effects,and responses of horizontal violence experienced by nursing students and the relationship between occupational belonging and horizontal violence.Materials and Methods:The present study used a descriptive,cross-sectional survey design.The data were collected from the 277 undergraduate nursing students selected by convenience sampling method using personal characteristics form,horizontal violence survey,and occupational belonging subscale at nursing faculty in Istanbul.Results:The first three horizontal violence behaviors experienced by nursing students were found,respectively:being forced to do trivial and unimportant work which are not the students’responsibility(84.1%),being exposed to sarcastic remarks by nurses(80.9%)and unwillingness of the nurse to help and lack of support(80.9%)and being ignored by nurses(77.3%).Nearly half of the students(48.4%)considered leaving nursing profession as a result of horizontal violence.There was a negative relation between horizontal violence and occupational belonging(P<0.05).As horizontal violence scores increased,occupational belonging scores decreased.Conclusions:Findings from this study shown that horizontal violence decreasing occupational belonging,and so nursing students consider leaving nursing profession or education.Further studies are needed to better demonstrate addressing the horizontal violence bilaterally and the effective preventive interventions.

This study develops a holistic approach that underlines the effect of spatial,behavioral,and perceptional dimensions on walkability at the neighborhood scale.The study recognizes quantitatively measured tangible factors and qualitatively assessed intangible values to evaluate the level of walkability in Besiktas,Istanbul(Turkey).The research explores the independent and combined effects of physical characteristics,individual perceptions,and walking behavior on walkability.Geographic Information Systems urban analyses,space syntax analysis,and observation to establish an analytical framework for spatial and behavioral dimensions of walkability in Besiktas central district are used as a methodology.Additionally,perceptional data is gathered through an experimental approach,which involves participants walking along pre-determined routes using cartography techniques and employing“shared walks”as a mobile method.The results of study confirmed previous findings from studies that the physical,social,and typological attributes of the built environment have a significant correlation between walking behavior and the physical and social aspects of the built environment.This study provides a comprehensive walkability assessment framework,encompassing quantitative and qualitative dimensions.By incorporating spatial,behavioral,and perceptual dimensions,this approach provides an efficient and integrated analytical methodology for evaluating walkability,distinguishing itself with its originality in the field of walkability research.

This research provides a mixed method of investigating and analyzing pedestrian behavior in pedestrianized areas,which is one of the human-based urban development practices.The relationship between pedestrian behavior and perception of the city is discussed through a combination of architect and urban planner Gordon Cullen’s theory and Space Syntax methodology.This study examines the development and implementation of the methodology to understand the relationship between pedestrian behavior and space in Besiktas Koyici settlement,Istanbul.The methodology,integrated with on-site observations(observing 10 min at five different observation points in the settlement at predetermined times on weekdays and weekends),surveys(determining pedestrian movement and perception in terms of Cullen’s theory)and spatial configuration(Space Syntax analysis(connectivity,integrity)done with DepthMapX software),aims to analyze pedestrian behavior patterns.The concepts of sense of place and content,as well as the effects of their sub-concepts on their behaviors,were revealed because of a survey conducted with students and architects of variety of educational levels who are daily users of the field.Pedestrian counting and observations were used to create density,activity,and mobility maps of the area.It has been found that pedestrianized areas have a significant impact on pedestrian behavior through influencing vitality and appeal and that Cullen’s theory may be used to analyze pedestrian behavior and perceptions of the city.The study showed that spatial configuration and Cullen’s theory effect pedestrian movement by complementing each other.

An alternative approach to predict human response to blast induced ground vibration

The aim of this study is to create alternative approaches to predict human response to ground vibration.Site measurements were conducted in an urban quarry.The multiple discriminant analysis technique was used to develop classification models.The measured ground vibration values were grouped based on the limit values appearing in the literature.Two classification models were created to predict human response to ground vibration.In addition,classical predictor equations were developed to compare to the classification models.The best equations were chosen considering several error measures.All the models were tested on the independent data.The prediction accuracy of the discriminant analysis models is quite promising.Nearly eighty-nine percent of the test data were correctly classified.The classification models have a combined structure.The results of the models can be directly presented to the neighbors of the studied quarry.The models are not complex.They do not require additional software or information to practice and can be easily used by site engineers.

The aim of this paper is to construct the integral representation of the solution of Sturm-Liouville equation with eigenparameter-dependent discontinuity conditions at an interior point of the finite interval. Moreover, we examine the properties of the kernel function of this integral representation and obtain the partial differential equation provided by this kernel function.
