Enantioselective organocatalytic synthesis of the chiral chromenes by domino oxa-michael-aldol reaction

The proline based chiral organocatalyst has been found to be an efficient catalyst for the facile synthesis of substituted 2-aryl-2 H-chromenes-3-carbaldehyde. We envisioned that the iminium interaction between chiral amino catalysts and a,b-unsaturated carbonyl group was beneficial along with thiourea group as hydrogen bond donor, heterocyclic amines as general base in the domino oxa-michael-aldol reaction. This catalytic system provided the products in good to high yields（73%–96%） with excellent enantioselectivity（up to 97%） and reasonable reaction time. The atom economy, high yield and mild reaction conditions are some of the important features of this protocol.

Results of oxidation 2-(N-acetylamine)-3-(3,5-di-tert-butyl-4-hydroxyphenyl)-propionic acid oxygen depend on temperature. At 55℃?- 60℃, 2,4-di-tert-butylbicyclo(4,3,1)deca-4,6-dien-8-(N-acetylamine)-3,9-dion-1-oxa is formed. The constitution is based on dates of spectrums 1Н and 13С NmR. At 95℃?- 97℃, mixtures of 2,4-di-tert-butylbicyclo(4,3,1)deca-4,6-dien-8-(N-acetylamine)-3,9-dion-1-oxa and of 6,8-di-tert-butyl-3-(N-acetylamine)spiro(4,5)deca-1-oxa-5,8-dien-2,7-dione are produced. Structures are calculated by the method of Hartrii-Foka. values of enthalpies and of entropies allow to assume dynamic isomerism.

Novel Hydralazine Schiff Base Derivatives and Their Antimicrobial, Antioxidant and Antipla.modial Properties

Two novel Schiff bases, 3-[1-(2-(phthalazin-1-yl)hydrazono)ethyl)-1,3-oxa- zinane (PHEO) and 2-[(2-(phthalazin-1-yl)hydrazono)methyl]phenol (PHmP), derived from hydralazine hydrochloride, an effective drug against hypertension, were synthesized and characterized by spectroscopic methods, Infrared (IR), Proton Nuclear magnetic Resonance (1H NmR) and Carbon-13 Nuclear magnetic Resonance (13C NmR). PHEO showed low antimicrobial activity against one bacterial strain with mIC value of 250 μg/ml while PHmP showed interesting activity against 4 bacterial strains with mIC of 31.25 - 250 μg/ml compared to the standard drug, amoxicillin. PHEO and PHmP showed higher reducing activity on ferric ions compared to vitamin C. On lipid peroxidation, PHEO showed higher inhibition while PHmP showed lower inhibition compared to vitamin C. Both compounds presented lower stimulating effect and lower catalase activity compared to the standard vitamin C. PHEO and PHmP showed less than 80% inhibition in the preliminary antipla.modial assay and so were not considered for the dose-response studies.

The pathophysiology of oxaliplatin-induced neurotoxicity remains unclear, a lthough in vitro studies suggest involvement of voltage- gated Na+ channels. In the present study, clinical assessment was combined with nerve conduction stu dies (NCS) and nerve excitability studies in 16 patients after completion of oxa liplatin therapy. Chronic neuropathic symptoms persisted in 50% of patients. N CS confirmed abnormalities in symptomatic patients: sensory potentials were sign ificantly low, whereas motor studies remained essentially normal. At 12- month follow- up of symptomatic patients, positive sensory symptoms improved but NCS abnormalities persisted. Cumulative oxaliplatin dose was a predictor of neuropat hy, and long- term effects appeared to be minimized by low single- infusion do sages. Nerve excitability measures in symptomatic patients established that axon s were of high threshold. Refractoriness was significantly greater in patients ( symptomatic group, 56.3± 24.9% ; entire patient group, 46.3± 12.5% ; control s, 27.1± 1.9% ; P ve sensory symptoms of oxalipl atin- induced neuropathy improved, negative sensory symptoms and abnormalities of sensory nerve conduction persisted. Differences in nerve excitability measure s, particularly refractoriness, support in vitro studies indicating involvement of voltage- gated transient Na+ - channel dysfunction in the development of o xaliplatin- induced neurotoxicity.
