cytoplasmic domain of tissue factor promotes liver fibrosis in mice

AIM To evaluate the role of tissue factor(TF) and protease activated receptor(PAR)-2 in liver *** Using CCl4 administration for eight weeks, we induced hepatic fibrosis in wild-type C57BL/6 mice and in mice with deletion of the cytoplasmic signalling domain of TF(TF§CT/§CT), deletion of PAR-2(PAR-2-/-) and combined deletion of TF signalling domain and PAR-2(TF§CT/§CT/PAR-2-/-). Hepatic fibrosis area was assessed by quantitative imaging of picrosirius red staining. Hepatic collagen content was assessed by hydroxyproline levels. Hepatic stellate cells(αSMA positive) and hepatic macrophages(CD68 positive) were identified by immunohistochemistry. Hepatic gene expression was determined by PCR and liver TGFβ1 content by *** CCl4 treated mice with deletion of the PAR-2 gene(PAR-2-/-) and the cytoplasmic domain of TF(TF§CT/§CT) developed significantly less hepatic fibrosis, characterised by reduced liver fibrosis area and hydroxyproline content, compared to control wildtype mice treated with CCl4. The observed reduction in histological fibrosis was accompanied by a significant decrease in the hepatic content of TGFβ, the prototypic fibrogenic cytokine, as well as fewer activated hepatic stellate cells and hepatic macrophages. Deletion of the TF cytoplasmic signalling domain reduced hepatic fibrosis to levels similar to thoseobserved in mice lacking PAR-2 signalling but combined deletion provided no added protection against fibrosis indicating a lack of mutual modulating effects that have been observed in other contexts such as angiogenic *** Tissue factor cytoplasmic domain is involved in TF-PAR-2 signalling initiating hepatic fibrosis and is a potential therapeutic target, as its deletion would not impact coagulation.

Intra-urban variation in tuberculosis and community socioeconomic deprivation in Lisbon metropolitan area:a Bayesian approach

Self-assembled PEG-b-PDPA-b-PGEM copolymer nanoparticles as protein antigen delivery vehicles to dendritic cells:preparation,characterization and cellular uptake

Antigen uptake by dendritic cells(DCs)is a key step for initiating antigen-specific T cell *** the present study,novel synthetic polymeric nanoparticles were prepared as antigen delivery vehicles to improve the antigen uptake by ***-defined cationic and acid-responsive copolymers,monomethoxy poly(ethylene glycol)-block-poly(2-(diisopropyl amino)ethyl methacrylate)-block-poly(2-(guanidyl)ethyl methacrylate)(mPEG-b-PDPA-b-PGEM,PEDG)were synthesized by reversible addition-fragmentation chain transfer polymerization of 2-(diisopropylamino)ethyl methacrylate and N-(tert-butoxycarbonyl)amino ethyl methacrylate monomers,followed by deprotection of tert-butyl protective groups and guanidinylation of obtained primary amines.1H NMR,13C NMR and GPC results indicated the successful synthesis of well-defined PEDG *** copolymers could self-assemble into nanoparticles in aqueous solution,which were of cationic surface charges and showed acid-triggered disassembly contributed by PGEM and PDPA moieties,***,PEDG nanoparticles could effectively condense with negatively charged model antigen ovalbumin(OVA)to form OVA/PEDG nanoparticle formulations with no influence on its secondary and tertiary structures demonstrating by far-UV circular dichroism and UV-vis *** vitro antigen cellular uptake by bone marrow DCs(BMDCs)indicated using PEDG nanoparticles as antigen delivery vehicles could significantly improve the antigen uptake efficiency of OVA compared with free OVA or the commercialized Alum ***,as the surface cationic charges of OVA/PEDG nanoparticle formulations reduced,the uptake efficiency decreased ***,our work suggests that guanidinylated,cationic and acid-responsive PEDG nanoparticles represent a new kind of promising antigen delivery vehicle to DCs and hold great potential to serve as immunoadjuvants in the development of vaccines.

How reliable is the ReaxFF potential for describing the structure of alkanethiols on gold?A molecular dynamics study

The structures of self-assembled monolayers(SAMs)of short(methyl)and long(hexyl)chain alkyl thiols on the clean gold(111)surface were modeled using for the Au–S interactions either the reactive ReaxFF potential or the well-known nonreactive Morse potential,while for the Au–Au interactions either the ReaxFF potential or an embeddedatom method(EAM).Analysis of the molecular dynamics(MD)trajectories of possible SAM structures suggests that disordering of interfacial Au atoms is definitely driven by the gold–sulphur *** MD results reveal a novel structure where two methanethiol molecules are bound to a gold adatom that has been lifted from the surface at 300 K,and the same kind of RS–Au–SR motif was also observed for hexanethiol at 600 K but not at 300 *** is more,the above motif is only observed for the reactive ReaxFF ***,these results are in clear agreement with recent experiments and more costly first principles-based MD *** findings strongly support the use of reactive potentials such as ReaxFF for gathering an accurate description of Au–S interactions in inexpensive classical MD simulations.

Role of dynamic computed tomography scans in patients with congenital craniovertebral junction malformations

AIM To evaluate the role of dynamic computed tomography(CT)scan imaging in diagnosing craniovertebral junction(CVJ)instability in patients with congenital CVJ *** Patients with symptomatic congenital CVJ malformations who underwent posterior fossa decompression and had a preoperative dynamic CT scan in flexion and extended position were included in this *** of the following craniometrical parameters were taken in flexed and extended neck position:Atlanto-dental interval(ADI),distance of the odontoid tip to the Chamberlain’s line,and the clivus-canal angle(CCA).Assessment of the facet joints congruence was also performed in both *** of the values obtained in flexion and extension were compared using a paired Student’s *** A total of ten patients with a mean age of 37.9 years were *** flexion imaging,the mean ADI was1.76 mm,the mean CCA was 125.4°and the mean distance of the odontoid tip to the Chamberlain’s line was+9.62 *** extension,the mean ADI was1.46 mm(P=0.29),the mean CCA was 142.2°(Poth underwent CVJ fixation).The other two patients with a fixed subluxation were not initially *** patient with atlantoaxial assimilation and C23 fusion without initial facet subluxation developed a latter CVJ instability diagnosed with a dynamic CT *** with basilar invagination had a lower CCA variation compared to the whole *** Craniometrical parameters,as well as the visualization of the facets location,may change significantly according to the neck *** imaging can provide additional useful information to the diagnosis of CVJ *** studies addressing the relationship between craniometrical changes and neck position are necessary.

A survey of executive compensation contracts in China's listed companies

We analyze 228 executive compensation contracts voluntarily disclosed by Chinese listed firms and find that central-government-controlled companies disclose more information in executive compensation contracts than localgovernment-controlled and non-government-controlled companies. Cashbased payments are the main form of executive compensation, whereas equity-based payments are seldom used by Chinese listed companies. On average, there are no significant differences in the value of basic salaries and performance-based compensation in executive compensation ***, compared with their counterparts in non-government-controlled companies, executives in government-controlled companies are given more incentive compensation. Accounting earnings are typically used in executive compensation contracts, with few firms using stock returns to evaluate their executives. However, the use of non-financial measures has increased significantly since 2007.

Hydrocarbons in particulate samples from wildfire events in central Portugal in summer 2010

In summer 2010, twenty eight （14 PM=2.5 samples plus 14 samples PM2.5-10） smoke samples were collected during wildfires that occurred in central Portugal. A portable high-volume sampler was used to perform the sampling, on quartz fibre filters of coarse （PM=2.5-10） and fine （PM2.5） smoke samples. The carbonaceous content （elemental and organic carbon） of particulate matter was analysed by a thermal-optical technique. Subsequently, the particulate samples were solvent extracted and fractionated by vacuum flash chromatog- raphy into three different classes of organic compounds （aliphatics, polycyclic aromatic hydrocarbons （PAHs） and carbonyl compounds）. The organic speciation was performed by gas chromatography-mass spectrometry （GC-MS）. Emissions were dominated by the fine particles, which represented around 92% of the PM10. A clear predominance of carbonaceous constituents was observed, with organic to elemental carbon （OC/EC） ratios ranging between 1.69 and 245 in both size fractions. The isoprenoid ketone 6,10,14-trimethyl-2-pentadecanone, a tracer for secondary organic aerosol formation, was one of the dominant constituents in both fine and coarse particles. Retene was the most abundant compound in all samples. Good correlations were obtained between OC and both aliphatic and PAH compounds. Pyrogenic processes, thermal release of biogenic com- pounds and secondary processing accounted for 97% of the apportioned PM2.5 levels.
