Renin-angiotensin system.blockade: Effect on renal m.NA expression in 5/6 nephrectom.zed rats

The aim.of this study was to determ.nate the gene expression levels of angiotensinogen, angiotensin converting enzym., renin, (pro)renin receptor, and the final rennin-angiotensin system.(RAS) products Angiotensin (Ang) II and Ang 1-7 inthe rem.ant kidney of 5/6 nephrectom.zed rats and its response to RAS pharmacological blockade. male Wistar rats were divided into five groups: sham.operated (SO), 5/6 nephrectom.zed (NFX), NFX + captopril (50 m./ kg/day), NFX + losartan (10 m./kg/day), and NFX + aliskiren (10 m./kg/day). Animals were followed up for 60 days and protein urine excretion was m.asured. Systolic blood pressure, renal tissue RAS m.NA expression levels, plasma Ang II, and plasma Ang 1-7 were evaluated at day 60 after nephrectom.. Blood pressure and urine protein were increased after 5/6 nephrectom.. Ang II levels were increased 9.4 fold, whereas Ang 1-7 decreased 72.9% in NFX animals com.ared with SO rats. 5/6 nephrectom. increased renal angiotensinogen and (pro)renin receptor m.NA expression but down-regulated renin m.NA expression. RAS blockade restored the systolic blood pressure to normal values and slowed down urinary protein excretion, and also prevented changes in Ang II and Ang 1-7. RAS blockade reduced (pro)renin receptor, ACE, and AGT m.NA expression in the rem.ant kidney. However, renin m.NA expression increased com.ared with NFX rats. In conclusion these results suggest that inhibition of Ang II synthesis by RAS blockade is associated with renal regulation of RAS m.NA expression and this may be through a m.chanism.related with the Ang II/Ang 1-7 balance.

From.results obtained in the rheological characterization of a 4% dispersion of flam.oyant gum.with the HaakeRT20 viscom.ter, for different conditions of pH = 3.0 and 9.0, tem.erature 5°C, 25°C and 45°C, the data of the rheological behavior of the gum.dispersions were fitted to the power law m.del. To understand and predict the behavior of this gum. a m.del of Lattice Boltzmann D2Q9 was developed for the behavior, in addition to sim.lations for the conditions handled in the experim.nts perform.d with the HaakeRT20 viscom.ter.

Developm.nt and validation of prognostic nom.gram.based on log odds of positive lym.h nodes for patients with gastric signet ring cell carcinoma

Objective: Our aim. were to establish novel nom.gram.m.dels, which directly targeted patients with signet ring cell carcinoma(SRC), for individualized prediction of overall survival(os) rate and cancer-specific survival(CSS).m.thods: We selected 1,365 SRC patients diagnosed from.2010 to 2015 from.Surveillance, Epidem.ology and End Results(SEER) database, and then random.y partitioned them.into a training cohort and a validation *** predicted indicators, which were identified by using univariate testing and m.ltivariate analyses, were used to construct our prognostic nom.gram.m.dels. Three m.thods, Harrell concordance index(C-index), receiver operating characteristics(ROC) curve and calibration curve, were used to assess the ability of discrim.nation and predictive accuracy. Integrated discrim.nation im.rovem.nt(IDI), net reclassification im.rovem.nt(NRI) and decision curve analysis(DCA) were used to assess clinical utility of our nom.gram.***: Six independent predicted indicators, age, race, log odds of positive lym.h nodes(LODDS), T stage, m.stage and tum.r size, were associated with os rate. Nevertheless, only five independent predicted indicators were associated with CSS except race. The developed nom.gram. based on those independent predicted factors showed reliable discrim.nation. C-index of our nom.gram.for os and CSS was 0.760 and 0.763, which were higher than Am.rican Joint Com.ittee on Cancer(AJCC) 8 th edition tum.r-node-m.tastasis(TNm. staging system.0.734 and 0.741, respectively). C-index of validation cohort for os was 0.757 and for CSS was 0.773. The calibration curves also perform.d good consistency. IDI, NRI and DCA showed the nom.gram. for both os and CSS had a com.arable clinical utility than the TNm.staging ***: The novel nom.gram.m.dels based on LODDS provided satisfying predictive ability of SRC both in os and CSS than AJCC 8 th edition TNm.staging system.alone.

Prognostic significance of preoperative and postoperative CK19 and CEA m.RNA levels in peripheral blood of patients with gastric cardia cancer

AIm.To evaluate the clinical and prognostic significance of preoperative and postoperative cytokeratin 19(CK19) and carcinoem.ryonic antigen(CEA) m.RNA levels in peripheral blood of patients with gastric cardia cancer(GCC).m.THODS We detected the preoperative and postoperative m. NA levels of CK19 and CEA in peripheral blood of 129 GCC patients by using reverse transcription-polym.rase chain reaction and evaluated their clinical and prognostic significance by univariate Kaplan-m.ier survival analysis and m.ltivariate Cox proportional hazard analysis. A new prognostic m.del which stratified patients into three different risk groups was established based on the independent prognostic *** elevated preoperative and postoperative CK19 and CEA m. NA levels in peripheral blood of GCC patients were associated with lym.h node m.tastasis. Univariate analysis showed that tum.r size, histological grade, depth of tum.r invasion, lym.h node m.tastasis, preoperative CK19 m.RNA, and preoperative and postoperative CEA m.RNA levels were correlated with the prognosis of GCC patients. The m.ltivariate analysis showed that lym.h node status(P = 0.018), preoperative CK19(P = 0.035) and CEA(P = 0.011) m.RNA levels were independent prognostic factors for overall survival(os). The 5-year os rates for the low-, interm.diate-, and high-risk groups were 48.3%, 22.6%, and 4.6%, respectively(P elevated preoperative CK19 and CEA m. NA levels may be regarded as prom.sing biomarkers for predicting lym.h node m.tastasis and poor prognosis in patients with GCC. This new prognostic m.del may help us identify the subpopulations of GCC patients with the highest risk.

The 2.5 Ga Guyang greenstone belt is a major lithological unit in the northwestern part of the North China *** have recently been identified to occur within the lower part of the m.ta-volcanic sedim.ntary sequence of the greenstone *** biggest komatiite near Guyang has been mapped out,which is 500 m.long and 50 m.wide lenticular ***-os isotopic com.ositions of 9 sam.les were analyzed to date the *** os contents are from.0.88 to 2.63 ppb,identical with typical komatiites,and slightly lower than the normal *** 187os/188os ratios are from.0.1115 to 0.1197,which are lower enough to exclude the origin of Phanerozoic magma as widely developed in adjacent *** calculated Re depletion m.del ages(TRD) are from.1346 to 2454 ma,am.ng them.the oldest age of 2454 ma gives the m.nim.m.evaluation for the formation age of the Guyang ***,the komatiites are a part of the Guyang greenstone belt,indicating high degree m.lting of mantle during～2.5 Ga crustal growth event.
