Polycarcin V induces dNA-damage response and enables the profiling of dNA-binding proteins

To maintain genomic integrity and avoid diseases, the dNA-damage response(ddR) not only detects and repairs dNA lesions, but also contributes to the resistance to dNA-damaging *** the ddR plays a significant role in drug discovery using the principle of synthetic lethality. The incomplete current knowledge of the ddR encouraged us to develop new strategies to identify and study its components and pathways. Polycarcin V, belonging to the C-aryl glycoside natural products, is a light-activatable dNA-intercalating agent that causes dNA damage by forming a covalent [2+2]cycloadduct with thymine residue under 365–450 nm of light irradiation in a dNA-sequence-independent manner. Taking advantage of the light-activatable feature and temporal control of ddR, we designed and synthesized polycarcin V-based bifunctional chemical probes, including one that cross-links dNA to dNA-binding protein to explore the ddR induced by polycarcin V and uncover novel dNA–protein interactions. Utilizing this chemical probe and activity-based protein profiling-stable isotope labeling with amino acids in cell culture, we identified 311 dNA-binding protein candidates, including known ddR factors and additional proteins that may be of interest in discovering new biology. We validated our approach by showing that our probe could specifically cross-link proteins involved in nucleotide excision repair(NER) that repair bulky dNA adducts. Our studies showed that the [2+2] cycloadduct formed by polycarcin V could indeed be repaired by NER in vivo. As a dNA-damaging agent, polycarcin V or its drug-like derivative plus blue light showed promising properties for psoriasis treatment, suggesting that it may itself hold promise for clinic applications.

Bryophyte-like Fossil （Parafunaria sinensis） from Early-Middle Cambrian Kaili Formation in Guizhou Province, China

Abundant well-preserved animal and macroalgal fossils were discovered in the Early-MiddleCambrian Kaili Formation of Taijiang County, Guizhou Province, China, which constitute the Kaili Biota,named as one of the three important Burgess Shale-type faunas. The bryophyte-like fossil (Parafunariasinensis Yang gen. et sp. nov) was discovered in the horizon of occurrence of Macroalgal fossils. The fossilof P. sinensis possesses typical characters of bryophyte, such as whorled leaves, capsule and seta, andcomplex rhizoid or foot. We infer that the bryophyte-like fossil could be the ancestor of bryophyte. Thediscovery provides a new lead for further study on the origin and evolution of bryophyte or land plant.

Treatment of Aged Skin with a pH 4 Skin Care Product Normalizes Increased Skin Surface pH and Improves Barrier Function: Results of a Pilot Study

The physiological skin surface pH is just below 5. With age the skin surface pH increases up to 6. An increased pH correlates with reduced barrier integrity/cohesion. The present pilot study assesses possible normalization of an increased skin surface pH of the elderly and improvement of barrier function via application of ≈pH 4.0 skin care products. Baseline skin surface pH was determined in elderly (80+ years old;n = 15) compared to middle aged adults (31 - 50 years old;n = 15). The effect of o/w emulsions at pH-values of 3.5, 4.0, 4.5 and 5.5 on the skin surface pH was determined in both groups. Further, the effect of a 4-week treatment with a pH 4.0 skin care product on the skin surface pH, skin hydration and barrier integrity was assessed. Thirteen elderly females were involved in this home-in-use test. Increased baseline skin surface pH of the elderly normalizes to the physiological pH of 4.5 - 5.0 over 7 hours after single application of o/w-emulsions with a given pH of 3.5 or 4.0. A 4 week treatment employing the pH 4.0 skin care product improves the epidermal barrier integrity of the elderly significantly (p = 0.005). Reduction of the increased baseline skin surface pH of the elderly is accompanied by improved epidermal barrier integrity. Skin care products for the elderly have to be adjusted in the pH range of 3.5 to 4.0.

Investigation of an eclogite xenolith,discovered in a Cretaceous granite from the Central domain of the dabieshan massif in eastern China, yields new petrological insights into the high to ultrahigh -pressure metamorphism,experienced by the Qinling-dabie *** to inclusion as a xenolith in the granite during the Early Cretaceous,this eclogite xenolith had recorded a complex metamorphic evolution that complies with subduction and exhumation processes experienced by the continental crust of the South China ***-preserved mineral parageneses substantiate the prograde and retrograde stages revealed by inclusions in porphyroblastic garnet and zoned minerals such as garnet,omphacite and amphibole in the *** relatively low P/T re-equilibration during a late metamorphic stage was textually inferred by the presence of aluminous and calcic-subcalcic amphiboles such as katophorite,edenite,taramite and pargasite as main matrix *** to our U/Pb,Rb/Sr and new Ar/Ar geochronological results,namely109±1 and 112±2 Ma plateau ages for muscovite and amphiboles,respectively,two successive but distinct cooling stages account for the thermal history of the granite-migmatite gneiss dome that forms the Central dabieshan *** argue that prior to the Cretaceous doming,the Central dabieshan domain experienced a tectono-metamorphic evolution similar to that observed in the high-pressure to ultra high-pressure units developed in the Southern dabieshan domain and Hong’an massif.

The south-Liaodong peninsula massif is the easternmost Mesozoic metamorphic core complex,recognized in Eastern *** provides a good example of the combination of ductile shearing,syn-kinematic plutonism and polyphase *** Jurassic granodioritic plutons,located at the footwall of the detachment normal fault,and dated here at ca 160 Ma,recorded two different phases of cooling.A slow cooling regime of about 3-10℃/my prevailing before 122 Ma,was followed by a significant increase in cooling rate of about 40-55℃/my after that *** contrast,a single fast cooling path was recorded by the Cretaceous monzogranite situated in the footwall of the detachment normal *** result indicates that the Jurassic and Cretaceous plutons recorded different exhumation processes:a Jurassic slow or negligible exhumation and a Cretaceous fast one assisted by normal *** two cooling stages correspond to distinct geodynamic processes during the Jurassic and *** tectonics seems not significant before Early *** second stage,dominated by an extensional regime which develops after ca 120 Ma,is tentatively correlated to the lithosphere removal of the North China Craton.

A Commercially Available Skin Care Lotion with a pH of 4.5 and 10% Urea Improves Skin Surface pH, Stratum Corneum Hydration and Epidermal Barrier Function in Subjects with dry Skin and Atopic diathesis

OBJECTIVE: The physiological skin surface pH is crucial for several epidermal barrier functions, like stratum corneum integrity, cohesion and restoration. Alterations of the “normal” acidic nature of the skin surface have been shown to correlate with specific skin conditions like aged or inflamed skin and are leading to impaired skin barrier function and formation. It is previously demonstrated that topical acidification in atopic dermatitis improves stratum corneum function, skin barrier structure and clinical signs in dermatitis. Against this background, we examined the impact of a slightly acidic skin care product containing urea on stratum corneum hydration, skin surface pH and epidermal barrier function in subjects with dry skin and atopic diathesis. METHODS: Stratum corneum hydration, skin surface pH and transepidermal water loss were biophysically measured before and after a 4-week treatment period with the test product (pH 4.5, 10% urea) compared to the reference product in 25 volunteers. In addition, dynamic epidermal barrier parameters like stratum corneum integrity, cohesion and recovery were investigated by using a previously described tape stripping approach. RESULTS: It was shown that the test product (pH 4.5, 10% urea) significantly elevated stratum corneum hydration and improved the acidic nature of the skin surface by lowering the skin surface pH to a greater extent compared to the reference product. After the 4-week treatment period a significant faster barrier restoration was detected on the test site treated with the test product compared to the reference product.
