The Pr.state Cancer.Pr.vention Tr.al （PCPT） is a seminal study in the field of ur.logy. Mor. than 10year. after.its initial publication, updated data fr.m this tr.al continue to shape our.under.tanding of pr.state cancer. Among the major.findings fr.m the PCPT has been the demonstr.tion that pr.state cancer.is common in men with pr.state-specific antigen （PSA） once thought to be in the nor.al r.nge,~ finaster.de pr.vents the development of benign pr.static hyper.r.phy,2 it incr.ases the sensitivity of PSA3 and digital r.ctal examination.4 Fur.her.or. the PCPT helped to establish the link between er.ctile dysfunction and car.iovascular.disease,5 and per.aps most impor.antly finaster.de demonstr.ted a 25% r.lative r.sk r.duction in the diagnosis of pr.state cancer.compar.d with placebo.

Applying the technical advantages of blockchain technology to this complex system to manage peer.to-peer.ener.y shar.ng, tr.nsmission, data stor.ge and build smar. contr.cts between networ. par.icipants can develop an optimal consensus mechanism within the new power.gr.d. This paper.pr.poses a new fr.mewor. for.the application of blockchain in a decentr.lised ener.y networ.. The micr.gr.d is assumed to be pr.vate and managed by local pr.sumer.. AIM： To evaluate whether.intr.tumor.l expr.ssion of measles vir.s fusogenic membr.ne glycopr.teins H and F （MV-FMG）, encoded by an adenovir.s vector.***-HI F, alone or.in combination with local coexpr.ssion of cytokines （IL-2, IL-12, IL-18, IL-21 or.GM-CSF）, can ser.e as a platfor. for.inducing tumor.specific immune r.sponses in colon cancer.METHODS： We used confocal laser.scanning micr.scopy and flow cytometr. to analyze cell-cell fusion after.expr.ssion of MV-FMG by dye colocalization. In a syngeneic bilater.l subcutaneous MC38 and Colon26 colon cancer.model in C57BL/6 and BALB/c mice, we assessed the effect on both the dir.ctly vector.tr.ated tumor.as well as the contr.later.l, not dir.ctly vector. tr.ated tumor. We assessed the induction of a tumor.pecific cytotoxic T lymphocyte （CTL） r.sponse with a lactate dehydr.genase （LDH） r.lease assay.r.SULTS： We demonstr.ted in vitr. that tr.nsduction of MC38 and Colon26 cells with ***-H/F r.sulted in dye colocalization, indicative of cell-cell fusion, in addition, in the syngeneic bilater.l tumor.model we demonstr.ted a significant r.gr.ssion of the dir.ctly vector.inoculated tumor.upon intr.tumor.l expr.ssion of MV-FMG alone or.in combination with the tested cytokines. We obser.ed the highest anti-neoplastic efficacy with MV-FMG and lL-21 coexpr.ssion. The degr.e of tumor.r.gr.ssion of the not dir.ctly vector.tr.ated tumor.cor.elated with the anti-neoplastic r.sponse of the dir.ctly vector.tr.ated tumor. This r.gr.ssion was mediated by a tumor.specific CTL r.sponse.CONCLUSION： Our.data indicate that intr.tumor.l expr.ssion of measles vir.s fusogenic membr.ne glycopr.teins is a pr.mising tool both for.dir.ct tumor.tr.atment as well as for.tumor.vaccination appr.aches that can be fur.her.enhanced by cytokine coexpr.ssion.

In this wor. we pr.sent the fir.t efficient algor.thm for.unsuper.ised tr.ining of multi-class r.gular.zed least- squar.s classifier.. The appr.ach is closely r.lated to the unsuper.ised extension of the suppor. vector.machine classifier.known as maximum mar.in cluster.ng, which r.cently has r.ceived consider.ble attention, though mostly consider.ng the binar. classification case. We pr.sent a combinator.al sear.h scheme that combines steepest descent str.tegies with power.ul meta-heur.stics for.avoiding bad local optima. The r.gular.zed least-squar.s based for.ulation of the pr.blem allows us to use matr.x algebr.ic optimization enabling constant time checks for.the inter.ediate candidate solutions dur.ng the sear.h. Our.exper.mental evaluation indicates the potential of the novel method and demonstr.tes its super.or.cluster.ng per.or.ance over.a var.ety of competing methods on r.al wor.d datasets. Both time complexity analysis and exper.mental compar.sons show that the method can scale well to pr.ctical sized pr.blems.
