Antidiarrheal activity of hexane extract of Citrus limon peel in an experimental animal model

OBJECTIVE： Acute diarrhea is one of the major illnesses that cause death in children, despite clinical interventions and the use of oral rehydration therapy. Thus, there is need to discover other effective, affordable and accessible treatments for this disease. Therefore, this study was carried out to investigate the effects of hexane extract of Citrus limon peel （HECLP） on castor oil-induced diarrhea in rats. METHODS： Diarrhea was induced in male albino Wistar rats weighing 100-150 g. The antidiarrheal activity of HECLP at different oral dosages （5, 10 and 20 mg/kg） was investigated by counting the number of wet fecal pellets. Animals were further treated with propranolol, prazosin, nifedipine and atropine to assess the effects of receptor blockers on the activities of the extract. The effects of HECLP on castor oil-induced enteropooling and the intestinal transit time of activated charcoal were also evaluated. RESULTS： Each of the 3 doses of C. limon significantly reduced （P 〈 0.05） the number of wet fecal pellets produced by animals, with 20 mg/kg HECLP producing the highest percentage inhibition （34.2%） Wet fecal pellet inhibition by the standard drug Ioperamide （3 mg/kg p.o.） was 68.4% relative to the negative control. Blockage of 13 adrenergic receptors by propanolol abolished the antidiarrheal effects of HECLP. Intestinal fluid accumulation was inhibited by 68.7% and 78.5% by 20 mg/kg HECLP and Ioperamide respectively. Furthermore, 20 mg/kg HECLP significantly （P 〈 0.05） reduced the percentage intestinal transit time （21.4% ± 1.42%）, relative to the control （34.2% ±4.29%）; atropine （5 mg/kg, intraperitoneal injection） significantly （P 〈 0.001） reduced the percentage intestinal transit time to 11.2% ±0.85%. CONCLUSION： These results suggest that C. limon peel possesses antidiarrheal effects through antisecretory and antimotility mechanisms that act through the β adrenergic system.

High Fat Diet Alteration of Gut Microbiota Impacts Learning, Memory and Anxiety Response in Mice: Cannabidiol and Omega 3 Possible Remedies

Chronic intake of High Fat Diet (HFD) has the potential of causing a number of metabolic disorders. It is also theorized to be involved in perturbation of gut microbiota. Cannabididol (CBD) and omega 3 are known to possess a number of medicinal usefulness. Their combined use in experimental interventions is quite limited. A total of 15 mice were used for this research divided into three groups of five animals each. Group 1 was administered water and normal chow ad libidum. Group 2 had HFD and water ad libidum. Group 3 had HFD plus CBD (10 mg/kg) and omega 3 (200 mg/kg) all for a total of 12 weeks. They were tested on the elevated plus maze (EPM) and average entry time into the closed arm was recorded. They were also tested on the Y-maze and spontaneous alternations were measured. Thereafter animals were sacrificed and faecal content in the caecum was collected in sterile bottles and cultured for E. coli count. It was found that HFD group at p value E. coli count (2.4 × 106 ± 4.5) compared to group 1 (1.4 × 106 ± 5.6) and group 3 (1.42 × 106 ± 6.3). The findings revealed that HFD enhanced gut E. coli overgrowth which was reduced by CBD and Omega 3. The memory impairment and anxiety induction by HFD was also significantly ameliorated by CBD and omega 3. E. coli known to be implicated in dementia induction was suppressed by the interventions. Possible mechanisms proposed are actions of CBD and omega 3 on CB1, TRVP and 5HT receptors in reducing anxiety and their antioxidant/anti-inflammatory actions in mitigating the neuro-inflammatory effect of HFD and immune hyperstimulation of E. coli via the gutbrain-axis.

Omega 3 Fatty Acid and Cannabidiol Prolong Lifespan and Ameliorates Brain Ischaemia in Mice Fed Chronic High Fat Diet

Diets these days contain more fats. High fat diet (HFD) is a model of unhealthy eating in experimental animals. It is known to induce inflammatory responses and oxidative stress. Cannabidiol (CBD), the non-psychoactive component of Cannbis sativa has been legalized for medicinal use in many countries of the world. Omega 3 fatty acid, commonly found in fish oil is medicinal and necessary for brain development. The antioxidant and neuroprotective functions of CBD and omega 3 have made them relevant in many researches. In this study, 45 mice were used and divided into three groups of 15 animals each: Group 1 (normal feed and water ad libidum);Group 2 (HFD ad libidum);Group 3 (HFD + CBD + Omega 3) for 16 weeks. Thereafter, five animals from each group were selected and their frontal lobes were harvested for histological analyses using H and E staining. The remaining mice were allowed to feed on normal diet and observed till death. At the end, HFD significantly reduced life span (57.4 ± 0.3) when compared to control (78.9 ± 1.6) and HFD + CBD + omega 3 group (74.5 ± 0.8) at p < 0.05. HFD also caused significant brain ischemic damage, neuronophagia and significant perivascular oedema. CBD + Omega 3 induced significant astrocytosis compared to control and HFD group. The immune stimulation by the CBD + Omega 3 could be responsible for tissue survival and longevity by protection from inflammatory and oxidative injuries. Ischaemic tissue death could have been prevented by amelioration of artheroma formation due to HFD. Further studies will be required to ascertain other possible mechanisms behind these findings.

A Study of EM Algorithm as an Imputation Method: A Model-Based Simulation Study with Application to a Synthetic Compositional Data

Compositional data, such as relative information, is a crucial aspect of machine learning and other related fields. It is typically recorded as closed data or sums to a constant, like 100%. The statistical linear model is the most used technique for identifying hidden relationships between underlying random variables of interest. However, data quality is a significant challenge in machine learning, especially when missing data is present. The linear regression model is a commonly used statistical modeling technique used in various applications to find relationships between variables of interest. When estimating linear regression parameters which are useful for things like future prediction and partial effects analysis of independent variables, maximum likelihood estimation (MLE) is the method of choice. However, many datasets contain missing observations, which can lead to costly and time-consuming data recovery. To address this issue, the expectation-maximization (EM) algorithm has been suggested as a solution for situations including missing data. The EM algorithm repeatedly finds the best estimates of parameters in statistical models that depend on variables or data that have not been observed. This is called maximum likelihood or maximum a posteriori (MAP). Using the present estimate as input, the expectation (E) step constructs a log-likelihood function. Finding the parameters that maximize the anticipated log-likelihood, as determined in the E step, is the job of the maximization (M) phase. This study looked at how well the EM algorithm worked on a made-up compositional dataset with missing observations. It used both the robust least square version and ordinary least square regression techniques. The efficacy of the EM algorithm was compared with two alternative imputation techniques, k-Nearest Neighbor (k-NN) and mean imputation (), in terms of Aitchison distances and covariance.

DEM investigation of strength and critical state behaviours of granular materials under true triaxial loadings

This paper investigates the strength and critical state(CS)behaviours of granular materials via DEM simulations of true triaxial drained tests under three different loading modes including constant b(in-termediate stress ratio)tests with constantσ'1(major principal stress),constant p(mean pressure)and constantσ'3(minor principal stress)*** this end,a series of samples are generated with the same particle size distribution,and with the confining stresses ranging from 100 kPa to 900 *** CS is achieved for all *** the macroscopic behaviours and the microscopic behaviours are examined and compared considering different loading modes,confining stresses and intermediate stress ratios(b).The critical state lines(CSLs)are found to be unique and independent of the loading modes,but dependent on the b *** CSLs with b=0 and b=1 form the two boundaries of CSLs respectively beyond which CSLs under all other b tests cannot go *** different strength criteria are examined and compared in terms of both peak and CS *** Mohr-Coulomb strength criterion is found to be only suitable for axisymmetric loading *** Lade-Duncan criterion is only suitable for describing peak strengths,which is dependent on the loading modes and confining *** Satake criterion and Matsuoka-Nakai criterion are the more appropriate strength criteria for describing CS failures,indicating that the CS values of both the Satake parameter and the Matsuoka-Nakai parameter describe an inherent property that characterizes the CS failure for a given type of *** CS mechanical coordination number is fitted by a curved line for a given b value,which is unique regardless of the loading *** peak and CS values of both major and minor principal fabric tensors decrease with increasing b values,while they increase with increasing b values for the intermediate principal fabric tensor.

The recent mpox outbreak(in 2022e2023)has different clinical and epidemiological features compared with previous outbreaks of the *** this outbreak,sexual contact was believed to be the primary transmission route of the *** addition,the community of men having sex with men(MSM)was disproportionately affected by the *** population is also disproportionately affected by HIV *** that both diseases can be transmitted sexually,the endemicity of HIV,and the high sexual behavior associated with the MSM community,it is essential to understand the effect of the two diseases spreading simultaneously in an MSM ***,we aim to understand the potential effects of HIV on an mpox outbreak in the MSM *** develop a mechanistic mathematical model of HIV and mpox *** model incorporates the dynamics of both diseases and considers HIV treatment with antiretroviral therapy(ART).In addition,we consider a potential scenario where HIV infection increases susceptibility to mpox,and investigate the potential impact of this mechanism on mpox *** analysis shows that HIV can facilitate the spread of mpox in an MSM population,and that HIV treatment with ART may not be sufficient to control the spread of mpox in the ***,we showed that a moderate use of condoms or reduction in sexual contact in the population combined with ART is beneficial in controlling mpox *** on our analysis,it is evident that effective control of HIV,specifically through substantial ART use,moderate condom compliance,and reduction in sexual contact,is imperative for curtailing the transmission of mpox in an MSM population and mitigating the compounding impact of these intertwined epidemics.

Hairy cell leukemia(HCL)is an uncommon mature B-cell malignancy characterized by a typical morphology,immunophenotype,and clinical *** vast majority of HCL patients harbor the canonical BRAF V600E mutation which has become a rationalized target of the subsequently deregulated RAS-RAF-MEK-MAPK signaling pathway in HCL patients who have relapsed or who are refractory to front-line ***,several HCL patients with a classical phenotype display non-canonical BRAF mutations or *** include sequence variants within alternative exons and an oncogenic fusion with the IGH *** must be taken in the molecular diagnostic work-up of patients with typical HCL but without the BRAF V600E to include investigation of these uncommon ***,functional characterization,and reporting of further such patients is likely to provide insights into the pathogenesis of HCL and enable rational selection of targeted inhibitors in such patients if required.

DCFNet:Discriminant Correlation Filters Network for Visual Tracking

CNN(convolutional neural network)based real time trackers usually do not carry out online network update in order to maintain rapid tracking *** inevitably influences the adaptability to changes in object *** filter based trackers can update the model parameters online in real *** this paper,we present an end-to-end lightweight network architecture,namely Discriminant Correlation Filter Network(DCFNet).A differentiable DCF(discriminant correlation filter)layer is incorporated into a Siamese network architecture in order to learn the convolutional features and the correlation filter *** correlation filter can be efficiently updated *** previous work,we introduced a joint scale-position space to the DCFNet,forming a scale DCFNet which carries out the predictions of object scale and position *** combine the scale DCFNet with the convolutional-deconvolutional network,learning both the high-level embedding space representations and the low-level fine-grained representations for *** adaptability of the fine-grained correlation analysis and the generalization capability of the semantic embedding are complementary for visual *** back-propagation is derived in the Fourier frequency domain throughout the entire work,preserving the efficiency of the *** evaluations on the OTB(Object Tracking Benchmark)and VOT(Visual Object Tracking Challenge)datasets demonstrate that the proposed trackers have fast speeds,while maintaining tracking accuracy.
