决定母猪窝产仔数的其中一个主要因素是产前死亡率,最常发生于妊娠的头几周,这是由于胚胎发育过程(包括滋养层伸长和胚泡着床)出现异常。人们一直试图利用表型选择的方式提高产窝产仔数,然而,另一个有希望实现此目的的有效方法是利用基因型信息。一般而言,繁殖性状很适合使用标记辅助选择(MAS),在分子水平确立选择标准可以缩短世代间隔,因为能够在动物生命周期中较早地作出选择。此外,考虑到繁殖性状受性别限制的这一特性,基因型信息可以在不能直接观察到性状的性别上进行选择。据此,人们对参与繁殖性状调控的基因进行绘制和识别以及对基因表达图谱进行阐述产生了极大兴趣。本文就该领域目前所进行的大量研究与所采取的方法进行综述:一种方法是因其编码的蛋白质的生理重要性和其在其它哺乳动物繁殖中的作用而被选为候选基因。然后,通过各候选基因的基因多态和繁殖性状表型之间的相关研究鉴定候选基因的有效性。本文讨论的另一种方法是利用已存在或设想的家系作连锁分析,对感兴趣的繁殖性状绘制数量性状位点(quantitative trait lo.i,QTL)图谱。虽然各研究所报道的结果不一致,但检测到的QTL区域对将来利用分子遗传学研究识别定位候选基因可能会有帮助。然而,要更好地了解猪繁殖性能,这些功能基因方法须与蛋白组的详细分析相结合和整合,以便在遗传因素与生理学之间建立连锁关系。

The wall shear stress （WSS） that a mo.ing fluid exerts o. a surface affects many pro.esses including tho.e relating to.vascular functio.. WSS plays an impo.tant ro.e in no.mal physio.o.y （e.g. angio.enesis） and affects the micro.asculature＇s primary functio. o. mo.ecular transpo.t. Po.nts o. fluctuating WSS sho. abno.malities in a number o. diseases; ho.ever, there is no.established technique fo. measuring WSS directly in physio.o.ical systems. All current metho.s rely o. estimates o.tained fro. measured velo.ity gradients in bulk flo. data. In this wo.k, we repo.t a nano.enso. that can directly measure WSS in micro.luidic chambers with sub-micro. spatial reso.utio. by using a specific type o. virus, the bacterio.hage M13, which has been fluo.escently labeled and ancho.ed to.a surface. It is demo.strated that the nano.enso. can be calibrated and adapted fo. bio.o.ical tissue, revealing WSS in micro.do.ains o. cells that canno. be calculated accurately fro. bulk flo. measurements. 99mTc-Methylene dipho.pho.ate （99mTc-MDP） is widely used in clinical settings to.detect bo.e abno.malities. Ho.ever, the mechanism o. 99mTc-MDP uptake in bo.e is no. well elucidated. In this study, we utilized a mo.se tibia injury mo.el, single-pho.o. emissio. co.puted to.o.raphy （gamma scintigraphy o. SPECT）, ex vivo.micro.co.puted to.o.raphy, and histo.o.y to.mo.ito. 99mTc-MDP uptake in injury sites during skeletal healing. In an ex vivo.culture system, calvarial cells were differentiated into.o.teo.lasts with o.teo.enic medium, pulsed with 99mTc-MDP at different time po.nts, and quantitated fo. 99mTc-MDP uptake with a gamma co.nter. We demo.strated that 99mTc-MDP uptake in the injury sites co.respo.ded to.o.teo.last generatio. in tho.e sites thro.gho.t the healing pro.ess. The 99mTc-MDP uptake within the injury sites peaked o. day 7 po.t-injury, while the injury sites were o.cupied by mature o.teo.lasts also.starting fro. day 7. ~mTc-MDP uptake started to.decrease 14 days po.t-surgery, when we o.served the highest level o. bo.y tissue in the injury sites. We also.fo.nd that 99mTc-MDP uptake was asso.iated with o.teo.last maturatio. and mineralizatio. in vitro. This study pro.ides direct and bio.o.ical evidence fo. 99mTc-MDP uptake in o.teo.lasts during bo.e healing in vivo.and in vitro.
