Effects of glutamine-containing total parenteral nutrition on phagocytic activity and anabolic hormone response in rats undergoing gastrectomy

AIM: To investigate the effect of glutamine (Gln)-containing parenteral nutrition on phagocytic activity and to elucidate the possible roles of Gln in the secretion of anabolic hormones and nitrogen balance in rats undergoing a gastrectomy. METHODS: Rats with an internal jugular catheter were divided into 2 experimental groups and received total parenteral nutrition (TPN). The TPN solutions were isonitrogenous and identical in nutrient compositions except for differences in amino acid content. One group received conventional TPN (control), and in the other group, 25% of the total amino acid nitrogen was replaced with Gin. After receiving TPN for 3 d, one-third of the rats in each experimental group were sacrificed as the baseline group. The remaining rats underwent a partial gastrectomy and were killed 1 and 3 d, respectively, after surgery. Plasma, peritoneal lavage fluid (PLF), and urine samples were collected for further analysis. RESULTS: The Gln group had fewer nitrogen losses 1 and 2 d after surgery (d1, 16.6±242.5 vs-233.4±205.9 mg/d, d2, 31.8±238.8 vs-253.4±184.6 mg/d, P<0.05). There were no differences in plasma growth hormone (GH) and insulin-like growth factor-1 levels between the 2 groups before or after surgery. The phagocytic activity of peritoneal macrophages was higher in the Gln group than in the control group 1 d after surgery (A 1185±931 vs323±201, P<0.05). There were no differences in the phagocytic activities of blood polymorphonuclear neutrophils between the 2 groups at the baseline or on the postoperative days. No significant differences in interleukin-1β or interleukin-6 concentrations in PLF were observed between the 2 groups. However, tumor necrosis factor-α level in PLF was significantly lower in the Gln group than in the control group on postoperative d 3. CONCLUSION: TPN supplemented with Gln can improve the nitrogen balance, and enhance macrophage phagocytic activity at the site of injury. However, Gin supplementation has no effect on p

BACKGROUND Based on the breakthrough of genomics analysis, The Cancer Genome Atlas Research Group recently proposed an integrative genomic analysis, dividing gastric cancer(GC) into four subtypes, characterized by the chromosomal instability(CIN) status. However, the CIN status of GC is still vaguely characterized and lacking the valuable easy-to-use CIN markers to diagnosis in molecular and histological *** To explore the associations of CIN with downstream lipidomics *** We collected cancerous and noncancerous tissue samples from 18 patients with GC; the samples were divided into CIN and non-CIN types based on the system of The Cancer Genome Atlas Research Group and 409 sequenced oncogenes and tumor suppressor genes. We identified the lipidomics profiles of the GC samples and samples of their adjacent noncancerous tissues by using liquid chromatography–mass spectrometry. Furthermore, we selected leading metabolites based on variable importance in projection scores of > 1.0 and P <*** Twelve men and six women participated in this study; the participants had a median age of 67.5 years(range, 52–87 years) and were divided into CIN(n = 9)and non-CIN(n = 9) groups. The GC samples exhibited distinct profiles of lysophosphocholine, phosphocholine, phosphatidylethanolamine,phosphatidylinositol, phosphoserine, sphingomyelin, ceramide, and triglycerides compared with their adjacent noncancerous tissues. The glycerophospholipid levels(phosphocholine, phosphatidylethanolamine, and phosphatidylinositol)were 1.4-to 2.3-times higher in the CIN group compared with the non-CIN group(P < 0.05). AIM To explore the correlation of metabolomics profiles ofgastric cancer(GC) with its chromosomal instability(CIN) *** Nineteen GC patients were classified as CIN and nonCIN type by The Cancer Genome Atlas Research Group system, based on 409 oncogenes and tumor suppressor genes sequenced. The aqueous metabolites of the GC tumor and its surrounding adjacent healthy tissues were identified through liquid chromatographymass spectrometry. Groups were compared by defining variable importance in projection score of > 1.2, a fold change value or its reciprocal of > 1.2, and a P value of < 0.05 as a significant *** In total,twelve men and seven women were enrolled, with a median age of 66 years(range, 47-87 years). The numbers of gene alterations in the CIN GC group were significantly higher than those in the non-CIN GC(32-218 vs 2-17; P < 0.0005). Compared with the adjacent healthy tissues, GC tumors demonstrated significantly higher aspartic acid, citicoline, glutamic acid, oxidized glutathione, succinyladenosine, and uridine diphosphate-Nacetylglucosamine levels, but significantly lower butyrylcarnitine, glutathione hydroxyhexanoycarnitine, inosinic acid, isovalerylcarnitine, and threonine levels(all P < 0.05). CIN tumors contained significantly higher phosphocholine and uridine 5'-monophosphate levels but significantly lower beta-citryl-L-glutamic acid levels than did non-CIN tumors(all P < 0.05). CIN GC tumors demonstrated additional altered pathways involving alanine, aspartate, and glutamate metabolism, glyoxylate and dicarboxylate metabolism, histidine metabolism, and phenylalanine, tyrosine, and tryptophan *** Metabolomic profiles of GC tumors and the adjacent healthy tissue are distinct, and the CIN status is associated with downstream metabolic alterations in GC.

Single Channel Source Separation Using Filterbank and 2D Sparse Matrix Factorization

We present a novel approach to solve the problem of single channel source separation (SCSS) based on filterbank technique and sparse non-negative matrix two dimensional deconvolution (SNMF2D). The proposed approach does not require training information of the sources and therefore, it is highly suited for practicality of SCSS. The major problem of most existing SCSS algorithms lies in their inability to resolve the mixing ambiguity in the single channel observation. Our proposed approach tackles this difficult problem by using filterbank which decomposes the mixed signal into sub-band domain. This will result the mixture in sub-band domain to be more separable. By incorporating SNMF2D algorithm, the spectral-temporal structure of the sources can be obtained more accurately. Real time test has been conducted and it is shown that the proposed method gives high quality source separation performance.

We propose a modified residual income model that incorporates Tobin’s Q as a proxy for growth and find that this model outperforms many other alternative valuation models that have been extensively employed in *** assuming that stocks are mispriced if they deviate from the estimated intrinsic values,we can then develop an arbitrage strategy that sells the overvalued stocks and buys the undervalued *** arbitrage strategy based on the proposed model also outperforms the Ohlson’s residual income model(1995),the Campbell-Shiller’s valuation model(1995),and other contrarian strategies based on various accounting-fundamental-to-price *** further trace the source of mispricing and conclude that,by controlling the growth variables,it can be attributed to a simple truth that investors are unable to distinguish discretionary accruals from reported earnings.

Yulin section is a typical sedimentary record for reflecting the environmental evolution of Ordos Desert, china in the past 1.1Ma. By analyzing its sequence and grain-size composition some views have been put forward in this paper as follows. The layers of sand, loess and palaeosol in Yulin section were respectively formed by wind and the pedogenesis on parent material of the sand and loess. Since 1.1Ma B. P., Ordos Desert has alternately experienced 11 stages of shifting dunes under extreme cold-dry climatic environment, 7 stages of fixed and semi-fixed dunes and 8 stages of dust (loess) under cold-dry climatic condition; and the pedogenesis environment under 15 times of warm-humid climate and 3 times of temperate-humid climate (brownish-drab soils and black soils formed respectively). The aeolian sand had already existed in Ordos Desert at latest by 1.1Ma B. P., and from that time on it has undergone a series of alternative processes of shifting sands, fixed and semi-fixed dunes, loess and soils. Ordos Desert has been situated in the transitional belt of the Mongolian High Pressure and margin of the southeast summer monsoon since 1.1Ma B. P., and influenced repeatedly by migration of the lithofacies belts of shifting sands, fixed and semi-fixed dunes, loess and soils, which have been caused by the climatic fluctuations of glacial and interglacial periods.

Background： CD4 count is used to determine antiretroviral therapy （ART） eligibility. In china, flow cytometers are mostly located in urban areas with limited access by patients residing in remote areas. In an attempt to address this issue, we conducted a study to validate the performance ofAlere PIMA point-of-care CD4 analyzer. Methods： Venous and finger-prick blood specimens were collected from HIV-positive participants from two voluntary counseling and testing sites in Yunnan Province. Both venous and finger-prick blood specimens were tested with the PIMA analyzer. Venous blood specimens tested with the Becton Dickinson FACSCalibur were used as a reference. Results： Venous specimens from 396 and finger-prick specimens from 387 persons were available for analysis. CD4 counts by PIMA correlated well with those from FACSCalibur with an R2 of 0.91 for venous blood and 0.81 for finger-prick blood. Compared to FACSCalibur, the PIMA analyzer yielded lower counts with a mean bias of- 47.0 cells/μl （limit of agreement, [LOA]： -204-110 cells/μl） for venous blood and -71.0 cells/μl （LOA： -295-153 cells/μl） for finger-prick blood. For a CD4 threshold of 350 cells/μl, the positive predictive value （PPV） of PIMA was 84.2% and 75.7% and the negative predictive value （NPV） was 97.6% and 95.8% for venous and finger-prick blood, respectively. For an ART threshold of 500 cells/μl, the corresponding PPV was 90.3% and 84.0% and NPV was 94.3% and 93.4%, respectively. Conclusions： CD4 counting using venous blood with PIMA analyzers is a feasible alternative to a large flow cytometer to determine ART eligibility.
